Synthesis and Structure-Activity Relationships of Novel neocryptolepine Derivatives

Herein we reported the synthesis of a novel series of neocryptolepine（5-methyl-5H-indolo[2,3-b]quinoline） derivatives containing different substituents at Cll using methyl 1H-indole-3-carboxylate and N-methylaniline as starting material. The target 21 compounds were evaluated for their axltibacterial activity in vitro against gram-positive bacteria（B, subtilis and S. aureus） and gram-negative bacteria（E, coli and ***）. Almost all the tested compounds showed moderate to high activities against the four bacterial strains at the minimum inhibitory concentrations（MICs） of 1-10μg/mL. The obtained results suggest that part of the novel synthetic neocryptolepine derivatives exhibit significant antibacterial effect against all the tested organisms.

It is well known that an essential component of the search for new leads in the designing program is the synthesis of molecules,which are novel yet resemblance to natural ***[2,3-b]quinoline ring system of neocryptolepine alkaloid have been deeply studied as an effective DNA intercalating agent and exhibited exceptionally pharmacological properties,such as strong antiplasmodial,antimicrobial and cytotoxic activities in vitro and significant antitumor properties in ***,in recent years enormous synthetic efforts have been devoted surrounding the unique structural array of indolo[2,3-b]quinoline skeleton of neocryptolepine alkaloid to design and synthesize new families of such poly-fused heterocyclic systems for current medicinal chemistry *** these syntheses are well established,alternatives are still desirable in order to avoid expensive transition metal catalyst,high reaction temperature or toxic *** the present investigation,we established a novel and efficient route to the direct construction indolo[2,3-b]quinoline ring system through the one-pot reaction between3-acetyl-2-ethoxyindole and *** comparison with some syntheses previously realized,our synthesis using 10%aqueous ethanol as solvent and KOH as base under mild conditions is very simple and constitutes a practical route to this class of alkaloid analogous in a single step with less of an impact on the *** believe this reaction was enough scope for further investigation,and could be useful tool for the short synthesis of indolo[2,3-b]quinoIine alkaloid derivatives.
