Resonance Scattering Detection of Trace Hg2＋ Using Aptamer-modified AuPd Nanoalloy Probe as Catalyst

The 5 nm AuPd nanoalloy in mole ratio of Au：Pd=32 ： 1 was prepared, using sodium citrate as the stabilizing agent and NaBH4 as the reductant. The AuPd nanoalloy was modified by the aptamer to prepare an aptamer- AuPd （AptAuPd） probe for resonance scattering （RS） detection of 5.（Y-1250 nmol/L Hg2＋. The AptAuPd-Hg2＋ aptamer reaction solution was filtrated by membrane, and the AptAuPd in the filtrate exhibited strong catalytic ef- fect on the slow NiP particle reaction between NiCI2 and NaHzPO2, and the NiP particles showed a RS peak at 508 nm. The RS intensity decreased when Hg2＋ concentration increased. The decreased RS intensity was linear to Hg2＋ concentration in the range of 0.5--1250 nmol/L. The RS assays were used to determine Hg2＋ in real samples, with good results.

Ill tliis work, two kinds of PEGylated salicylaldehyde azine(SA) polymers were prepared and investigated for cellular imaging applications. First, a diazido derivative of SA was synthesized and subsequently PEGylated with polyethylene glycol monomethyl ether(mPEG) by metal-free azide-alkyne 1,3-dipolar cycloaddition reaction. The formed triazole group in niPEG-SA was then converted into cationic triazolium group by N-alkylation reaction. Botli the synthesized polymers, niPEG-SA and N-alkylated niPEG-SA, showed good dispersibility in water, but differences in self-assembly of nanostructures. The niPEG-SA with triazole groups self-assembled into micelles, while the N-alkylated mPEG-SA with triazolium groups sell-assembled into vesicles. Furthemiore, mPEG-SA and N-alkylated niPEG?SA naiioparticles showed bright fluorescence due to the aggregation of AIE-active SA molecules in the nanoparticles and could be successfully used as fluorescent naiioprobes for bioimaging applications in HeLa cancer cells. Finally, both the synthesized polymers showed minimal cytotoxicity and low hemolytic activity. Therefore, these PEGylated SA polymers proved to be promising bioimaging nanoprobes or traceable dnig delivery vehicles.

In this study, we report the fabrication of engineered iron oxide magnetic nanoparticles (MNPs)functionalized with anti-human epidermal growth factor receptor type 2 (HER2) antibody totarget the tumor antigen HER2. The Fc-directed conjugation of antibodies to the MNPs aidstheir efficient immunospecific targeting through free Fab portions. The directional specificity ofconjugation was verified on a macrophage cell line. Immunofluorescence studies on macrophagestreated with functionalized MNPs and free anti-HER2 antibody revealed that the antibodymolecules bind to the MNPs predominantly through their Fc portion. Different cell lines with different HER2 expression levels were used to test the specificity of our functionalized nanoprobe formolecular targeting applications. The results of cell line targeting demonstrate that these engineered MNPs are able to differentiate between cell lines with different levels of HER2 expression.

Multimodality optical imaging probes have emerged as powerful tools that improve detection sensitivity and accuracy, important in disease diagnosis and treatment. In this review, we focus on recent developments of optical fluorescence imaging(OFI) probe integration with other imaging modalities such as X-ray computed tomography(CT), magnetic resonance imaging(MRI), positron emission tomography(PET), single-photon emission computed tomography(SPECT), and photoacoustic imaging(PAI). The imaging technologies are briefly described in order to introduce the strengths and limitations of each techniques and the need for further multimodality optical imaging probe development. The emphasis of this account is placed on how design strategies are currently implemented to afford physicochemically and biologically compatible multimodality optical fluorescence imaging probes. We also present studies that overcame intrinsic disadvantages of each imaging technique by multimodality approach with improved detection sensitivity and accuracy.

Chemodynamic therapy kills cancer cells with reactive oxygen species generated by endogenous triggers in the tumor *** chemodynamic therapy is blossoming in recent years,their therapy process still faces a series of *** unknown catalytic activity of chemodynamic therapy reagents may lead to unpredictable therapy effects,so it is necessary to reveal the therapeutic mechanism of chemodynamic therapy and develop self-monitoring *** this mini-review,we summarize and illustrate the most recent progress of chemodynamic therapy,focusing on the applications of magnetic imaging and optical imaging probe for monitoring cancer chemodynamic ***,we also discuss the potential challenges and the further directions of this field.

As an emerging hybrid imaging modality,microwave-induced thermoacoustic imaging(MTAI),using microwaves as the excitation source and ultrasonic signals as the information carrier for combining the characteristics of high contrast of electromagnetic imaging and high resolution of ultrasound imaging,has shown broad prospects in biomedical and clinical *** imaging contrast depends on the microwave-absorption coe±cient of the endogenous imaged tissue and the injected MTAI contrast *** systemically introduced functional nanoparticles,MTAI contrast and sensitivity can be further improved,and enables visualization of biological processes in *** recent years,functional nanoparticles for MTAI have been developed to improve the performance and application range of MTAI in biomedical *** paper reviews the recent progress of functional nanoparticles for MTAI and their biomedical *** challenges and future directions of microwave thermoacoustic imaging with functional nanoparticles in theeld of translational medicine are discussed.

Rational design of a multifunctional molecular dye for dual-modal NIR-Ⅱ/photoacoustic imaging and photothermal therapy

The continuous or real-time tracking of biological processes using biocompatible contrast agents over a certain period of time is vital for precise diagnosis and treatment,such as monitoring tissue regeneration after stem cell transplantation,understanding the genesis,development,invasion and metastasis of cancer and so on. As compared to the traditional fluorescent materials,aggregationinduced emission fluorogens(AIEgens) and conjugated polymers(CPs) enjoy the advantages of simultaneously possessing high brightness,excellent photostability,and good biocompatibility. As such,they have attracted great attention in modern biomedical research for challenging *** the last decade,we have been focusing on the development of new cell trackers for regenerative medicine and cancer *** CPs or AIEgens as the fluorescent domain,the obtained organic trackers have shown higher brightness,better stability in biological medium and comparable size/photostability as compared to their counterparts of inorganic quantum dots(QDs).In summary,the exploration and application of these advanced organic trackers in stem cell tracking,cancer theranostics,vascular imaging,and tissue regeneration will be introduced.
