背景与目的:小细胞肺癌(smallcelllungcancer,SCLC)是一种侵袭性极强的恶性肿瘤,具有增长迅速、早期转移等特点。目前公开的数据库中尚未见到小细胞肺癌的双向电泳参考图谱及其蛋白表达谱。本研究目的是建立高分辨率的小细胞肺癌细胞系nci-H446细胞双向凝胶电泳图谱,并初步分析其蛋白质表达情况。方法:用固相pH梯度双向凝胶电泳技术(IPG-DALT)分离nci-H446细胞总蛋白,凝胶银染显色,ImageMaster2D图像分析系统分析,从凝胶中选取分离较好的蛋白质点,应用基质辅助激光解吸电离飞行时间质谱(MALDI-TOF-MS)技术和数据库搜索鉴定蛋白质。结果:获得了背景清晰、分辨率和重复性较好的双向凝胶电泳图谱,三块胶平均蛋白质点数为1506±74,匹配点数为1412±56,匹配率达93.4%,三块胶蛋白质点在位置上有较好的重复性,不同胶间蛋白质点在IEF方向偏差是(0.96±0.27)mm,在SDS-PAGE方向偏差是(1.24±0.41)mm。胶内酶解-肽质指纹图分析鉴定了58个蛋白质,其中有原癌蛋白、细胞周期调控和信号转导相关蛋白质。结论:建立了小细胞肺癌细胞系nci-H446双向电泳参考图谱,并应用质谱技术鉴定了部分蛋白质点,为进一步构建其蛋白质表达数据库提供了基础。

The Role of Chemokine Receptor CXCR7 in Lung Cancer

To investigate whether CXCL12 and its receptor, CXCR7, play a role in lung cancer invasion and metastasis. Western blot and immunocytochemistry were used to detect CXCR7 protein expression in 8 lung cancer cell lines, EKVX, HOP62, HOP92, nci-H23, nci-H226, nci-H322M, nci-H446, and A549, and cell migration experiment was conducted to observe mobility of the lung cancer cells. The concentration of intracellular calcium in cytoplasm was measured under the fluorescence microscopy. CXCR7 protein was positively expressed in the 8 lung cancer cell lines EKVX, HOP62, HOP92, nci-H23, nci-H226, nci-H322M, nci-H446, and A549. Following CXCL12 stimulation, obvious pseudopodia of lung cancer cells was observed under the microscope. The cell migration experiment showed that after incubation with CXCL12, the number of EKVX cells, which passed through the polycarbonate microporous filter membranes increased to a great extent. This phenomenon can be reversed by CXCR7-siRNA. After CXCL12 incubation, the intracellular Ca2+ level in the EKVX cells was increased to a great extent. Chemokine CXCL12 facilitates the migration of lung cancer cells by changing the concentration of intracellular Ca2+. The CXCL12-CXCR7 axis may play an important role in lung cancer invasion and metastasis. It could be a potential target for lung cancer therapy and an effective way to prevent recurrence and metastasis of lung cancer.

The National Institutes of Health （NIH）, a part of the U.S. Department of Health and Human Services, is the nation＇s medical research agency-making important discoveries that improve health and save lives.Thanks in large part to NIH-funded medical research, Americans today are living longer and healthier. Life expectancy in the United States has jumped from 47 years in 1900 to 78 years as reported in 2009, and disability in people over age 65 has dropped dramatically in the past 3 decades. In recent years, nationwide rates of new diagnoses and deaths from all cancers combined have fallen significantly.

In this paper the changes of NCl(a1△) and NCl(b1∑) emission intensity is presented when I2/He is introduced into the stream of Cl/Cl2/He/HN3/NCl(a1△)/NCl(b1∑). The dependences of atomic iodine I(2P1/2) on flow rates of gases were also investigated. The optimum parameters for I(2P1/2) production are given.

建立了一种可用于水产品及食用油中氟乐灵残留量分析的分散型固相萃取-气相色谱-负化学离子源质谱方法。水产品及食用油经乙腈提取，4℃冷藏后，采用分散型固相萃取法净化，由气相色谱-负化学离子源质谱选择离子监测技术进行测定与确证，同位素内标法定量。在1～40μg / L 范围内氟乐灵农药的线性关系良好；方法定量限（LOQ）为0.02μg / kg；对鳗鱼、烤鳗、梭子蟹、小龙虾、猪油和橄榄油等6种复杂基质进行1.0、2.0和3.0μg / kg 等3个水平的添加回收试验，平均回收率均处于80％～100％之间，RSD≤10.3％；无干扰现象出现。该方法可作为水产品及食用油中氟乐灵残留检测的确证方法。

国际癌症研究署(IARC)和美国环境保护局(EPA)都将苯列为确证的人类致癌物.尹松年、李桂兰等[1,2]与美国国家癌症研究所(nci)合作对中国74 828名接苯工人和35 805名对照工人进行的队列研究证明,接苯工人发生血液淋巴系统恶性肿瘤的相对危险性明显增加,但是,由于苯在工业上的重要作用,至今仍是应用最为广泛的化工原料和工业溶剂之一.据报道,2002年亚洲地区(包括澳大利亚)苯的年产量约为1 170万吨,并预计将以每年206万吨的速度增长,中国每年将增加88万吨.我国职业苯暴露工人达50万以上,占各种化学物职业接触人数之首,同时,苯又是香烟烟雾和汽车尾气的组分,广泛存在于生活环境中.由此看来,目前苯暴露是无法避免的,因此寻找苯致癌危险性的生物标志物,早期预防苯白血病的发生,对于保护工人健康具有重要意义。

Outcome of Management of Pseudomyxoma Peritonei: National Cancer Institute Experience

Background: The current treatment for pseudomyxoma peritonei (PMP) consists of radical cytoreductive surgery (CRS) followed by hyperthermic intra-peritoneal chemotherapy (HIPEC). Aim: To assess PMP patients regarding the clinical and pathological characteristics, the treatment including surgery (CRS) and chemotherapy either HIPEC type or post-operative systemic chemotherapy aiming to evaluate end results regarding recurrence and survival. Patients and Methods: This retrospective study included 39 patients with PMP who were diagnosed, treated and followed-up from 2009-2014 at National Cancer Institute, Cairo, Egypt. Results: High grade mucinous adenocarcinoma was found in 23.1% of patients. Patients with low grade tumor showed higher survival rate compared with patients with high grade disease. The mean operative PCI score (peritoneal cancer index) that was done to all patients who were explored was 15.81. Our study reported success to achieve complete cytoreduction that was combined with HIPEC in 44% of patients who were planned for this modality. Treatment related postoperative grade (3 - 5) complications mainly surgery related developed in 17.3 of patients. Operative mortality was 22.2%. The follow up period in our study was quite short (mean 22.9). However the overall survival at the end of the follow up in our study was 48.7%, 1 year survival was 82%, and 2 year survival was 41%. The overall survival in patients treated with CRS and HIPEC was 66.6%, with 1 year and 2 year survival of 91% and 66.6% respectively. Only 2 patients developed recurrent disease during the follow up period. Conclusions: The outcome of PMP treatment process is extremely variable. Combined CRS and HIPEC is considered the best therapeutic approach for patients with PMP. Surgical experience combined with proper patient selection have to be built up together to improve the outcome. That could only be achieved through more centralization of patients’ treatment in specialized units or center.

本文建立了气相色谱质谱法负化学离子化方式检测运动场地面层合成材料中短链氯化石蜡(SCCPs)(C_(10-13)C_(l5-10))的定量方法.样品经正己烷超声萃取60 min,提取液使用浓硫酸净化后,使用GCMS对SCCPs的24组同类物进行分离和检测.在5.0 mg·L^(-1)浓度下,连续6次进样,SCCPs的24组同系物峰面积RSD%值均小于10.0%.以3倍信噪比计算检测限,24组同类物检测限分布在0.049—2.53 mg·L^(-1)之间.加标回收实验中,加标浓度为25 mg·kg^(-1)和100 mg·kg^(-1),SCCPs平均加标回收率为122.2%和107.5%.实验结果证明,此方法能够较为准确测定运动场地面层合成材料中短链氯化石蜡的含量.
