Changes in myofibrillar and Mitochondrial Compartments during Increased Activity: Dependance from Oxidative Capacity of Muscle

Striated muscle tissue contains fibers with high oxidative capacity (heart muscle), higher oxidative capacity (type I and IIA fibers of skeletal muscle) and low oxidative capacity (type IIB/X fibers of skeletal muscle). Muscle fibers with higher oxidative capacity contain large mitochondria tightly packed with cristae as well as small forms of mitochondria containing relatively few cristae. The intensive development of the mitochondrial apparatus in the post-activity period reflects the adaptive processes, which is intended to supply the increased energy requirements of muscle fibers with higher oxidative capacity. Muscle fibers with low oxidative capacity contain significantly less mitochondria than fibers with higher capacity. It is typical to type IIB fibers that after intensive muscle activity there are damaged myofibrils in a relatively small area, some myofibrils are twisted and lose the connection with the neighboring structures. It is still not fully known how skeletal muscles with different oxidative capacity respond to an increased functional activity and what differences exist in these fibers between oxidative capacity and function of myofibrils. The aim of the present short review was to compare structural-functional changes in mitochondrial and myofibrillar compartments of heart and skeletal muscle fibers with different oxidative capacity and the effect of increased functional activity on the interaction of these compartments.

Influence of Bleaching Powder on the Quality of Giant Freshwater Prawn (<i>Macrobrachium rosenbergii</i>)

Calcium hypochlorite commercially known as bleaching powder is used as a bleaching agent in shrimp processing industries in many countries and known to effect biochemical alteration in shrimp muscle. Studies were, therefore, undertaken to determine their effect in different concentrations viz., 10, 20, 30, 40 and 50 ppm with different time intervals on the quality of head-on, headless shell-on and peeled giant freshwater prawn (Macrobrachium rosenbergii) by determining biochemical and organoleptic aspects. myofibrillar protein solubility of fresh head-on, headless shell-on and peeled samples were 90.5%, 90% and 88%, respectively indicating a gradual decrease in protein solubility with increasing concentration of bleaching powder. Decrease in protein solubility was also higher in samples kept at longer duration in different concentration of bleaching powder. At a given concentration of 50 ppm for 30 min treatment, the loss of myofibrillar protein was higher (26.14%) in peeled samples than those of head-on and headless shell-on samples (20.44% and 21.11%). Shelf life of bleaching powder treated prawn samples was found to be reduced to 4 - 5 days in iced condition compared to 6 - 7 days for control samples. Peeled samples were also found to be more susceptible to bleaching powder than that of head-on and headless shell-on samples.

Myosin Heavy Chain Expression and Oxidative Modifications in Diabetic Rat Hearts

In this study, we tested the hypotheses that 1) diabetes-induced disturbances in cardiac my-ATPase activity would be attributed to not only myosin heavy chain (MHC) isoform transitions, but also reduced amounts in MHC protein;and 2) if diabetes results in declines in the MHC protein content, this change would relate to oxidative damage to MHC. Diabetes was induced by a single intraperitoneal injection of streptozotocin. After 6 weeks of injection, the left ventricles were excised for mechanical and biochemical analyses. Peak twitch tension and the rate of force development in papillary muscles were decreased by 23.4% and 34.1%, respectively. A 33.5% reduction in myofibrillar ATPase activity occurred in conjunction with a 9.5% decrease in MHC protein as well as MHC isoform transitions towards a slower phenotype. The decreased MHC content was not accompanied by elevations in carbonyl groups present in MHC. Whole muscle analyses indicated that the contents of malondialdehyde and reduced glutathione were elevated. These results suggest that decreases in the MHC content may be associated, at least in part, with a diabetes-related inactivation of cardiac my-ATPase and may not be due to accumulation of oxidative damage to protein.
