Biphasic taxonomic approaches for generic relatedness and phylogenetic relationships of Teichosporaceae

The placement of the dothideomycetous family Teichosporaceae has been controversial.Recent phylogenetic investigations have used a taxonomic lumping approach with the Floricolaceae and its genera have been synonymized under the earlier fam-ily name,Teichosporaceae.Intergeneric relationships were therefore obscure and proper generic delimitation was needed in upcoming studies.We here taxonomically revised the family Teichosporaceae based on both morphological and phylogenetic evidence.Teichosporaceae species have immersed or semi-immersed,erumpent to superficial,ostiolate ascomata,cellular or trabeculate pseudoparaphyses,cylindrical to oblong or sub-clavate asci and ellipsoid to oblong or fusiform,cylindric-fusiform or oblong to elliptical,ovoid to clavate,symmetric or asymmetric,initially hyaline or pale brown to dark brown or yellowish brown,1-3-septate or muriform ascospores.Asexual morphs are coelomycetous.Type or representative speci-mens of Teichosporaceae were loaned and fresh specimens were collected from China and Thailand.Maximum likelihood and Bayesian analyses of a combined ITS,LSU,SSU,tef1-αand rpb2 dataset were performed to clarify the phylogenetic affinities of taxa and examine monophyly of newly proposed genera.One new species(Floricola festucae),one new host record(Ramusculicola thailandica)and four new combinations(Aurantiascoma nephelii,A.quercus,Magnibotryascoma acaciae,M.melanommoides)are introduced.The broad genus concept of Teichospora is dismissed based on morphological dissimilarities and the monophyletic status of the proposed genera.We accept Asymmetrispora,Aurantiascoma,Floricola,Magnibotryascoma,Misturatosphaeria,Pseudoaurantiascoma,Pseudomisturatosphaeria,Ramusculicola and Teichospora as distinct genera in the Teichosporaceae.All recognized genera are phenotypically characterized and phylogenetically well-supported.The phylogenetic placements of three genera(Chaetomastia,Loculohypoxylon and Sinodidymella),which do not have molecular data cannot be conclusively clarified at present,but

The genus Lactarius Pers.(Russulales)is a cosmopolitan group of Basidiomycota that forms ectomycorrhizal relationships primarily with both deciduous and coniferous trees.Although the genus has been well-studied in Europe and North America,only fragmentary researches have been carried out on Asian species.In particular,the distribution of Lactarius species in South Korea is poorly understood due to insufficient morphological descriptions and a lack of DNA sequence data.In addition,the misuse of European and North American names has added to confusion regarding the taxonomy of Asian Lactarius species.In this study,the diversity of Lactarius in South Korea was evaluated by employing both morphological and phylogenetic approaches.A multi-locus phylogenetic analysis of 729 Lactarius specimens collected between 1960 and 2017 was performed using the internal transcribed spacer(ITS)region,partial nuclear ribosomal large subunit(nrLSU),partial second largest subunit of RNA polymerase II(rpb2),and minichromosome maintenance complex component 7(mcm7).49 Lactarius species were identified in three Lactarius subgenera:L.subg.Russularia(17 spp.),L.subg.Lactarius(22 spp.),and L.subg.Plinthogalus(10 spp.).Among them,28 Lactarius species were identified as new to science,while just 17 were previously described Lactarius species.Four of the taxa remain un-named due to paucity of materials.A key to Korean Lactarius species,molecular phylogenies,a summary of diversity,and detailed description are provided.

Culturable plant pathogenic fungi associated with sugarcane in southern China

Sugarcane(Saccharum officinarum)is one of the oldest crops cultivated by mankind.Numerous fungal taxa have been reported from this host,although most were not identified beyond genus level.In this study,we explored 31 sampling sites in Guangxi and Guangdong Provinces(China),and collected 370 diseased samples from leaves and roots of sugarcane,from which 762 strains were isolated.Our preliminary analysis based on internal transcribed spacer(ITS)sequences suggested that these isolates belonged to 143 species in 51 genera,but we could not assign 129 strains.Bipolaris,Chaetomium,Curvularia,Phoma and Nigrospora represented the top five most common genera identified,while 27 rare genera comprised only one identified species.In this study,we chose above mentioned common genera for in-depth morphological observations and multi-locus analyses,in order to identify these strains to species level.In this paper,we described one new genus,32 new species,and reported 19 new records for China and the asexual morph of Chaetomium olivaceum.Hitherto,this is the most comprehensive study with molecular identification and illustration of fungi associated with sugarcane,which greatly improves our understanding of culturable mycota associated with this host.

Myrica rubra is an important fruit tree with high nutritional and economic value and is widely cultivated in multiple regions of China.In January 2021,an unknown disease causing leaf spot was discovered on the leaves of M.rubra in Jiujiang City of Jiangxi Province(29.71°N,115.970 E).The initial symptoms were small pale brown spots on the leaves,which gradually expanded into round or irregular dark brown spots with the occurrence of the disease,the lesion developed necrotic tissues in the center at later stages,eventually led to the leaves turned chlorotic and wilted.Infected leaves with typical symptoms were collected and the leaf tissue(5 mm×5 mm) at junction of diseased and healthy portion were cut and disinfected with 75% ethanol for 45 s,followed by 1 % sodium hypochlorite for 1 min,rinsed in sterile water for 3 times and transferred to potato dextrose agar(PDA) at 28±1℃ for 3 days.Five fungal single isolates with similar morphology were purified from single spores.On PDA medium,the colonies initially appeared white with numerous aerial hyphae,and the center of the colony turned gray at later stages,less sporulation.On czapek-dox medium(CA),the mycelia of the colony was sparse and produced a large number of small bright orange particles(conidial masses).Conidia were single-celled,transparent,smooth-walled,1-2 oil globule,cylindrical with slightly blunt rounded ends,(14.45-18.44) μm×(5.54-6.98) μm(av=16.27 μm×.19 μm,n=50) in size.These morphological characteristics of the pathogen were similar to the descriptions of Colletotrichum fructicola(Ruan et al,2017;Yang et al,2021).To further confirm the identity of the pathogen,genomic DNA from isolates was extracted with DNA Extraction Kit(Biocolor,Shanghai,China),and the internal transcribed spacer(ITS),glyceraldehyde-3-phosphatedehydrogenase(GAPDH),calmodulin gene(CAL),actin(ACT) and chitin synthase 1(CHS 1) were amplified using the primers ITS1/ITS4(Gardes et al,1993),GDF/GDR(Templeton et al,1992),CL1 C/CL2 C(Weir et al,2012),ACT-512 F/ACT-783 R and CHS-79 F/CHS-

Didymellaceae（Pleosporales, Dothideomycetes） is one of the most species-rich families in the fungal kingdom. Species of Didymellaceae are geographically widespread and distribute in numerous ecological niches.Although most species have been shown to be important plant pathogens, with six species of quarantine significance, many species also infect human, insects, fungi or inhabit a wide range of other substrates as saprophytes. The taxonomy of Didymellaceae has undergone significant changes in recent years via morphological and phylogenetic analyses based on DNA sequence data of ex-type strains. Several poly-and paraphyletic genera, including Phoma, Didymella, and Ascochyta were redefined, along with the introductions of new genera. However, due to the tradition of host associated nomenclature, the wide host range and the widespread presence of Didymellaceae, taxonomy of the majority of species in this family remain unsolved and the real species diversity is largely unknown. To further resolve the phylogenetic relationships of Didymellaceae,in the present study, a systematic analysis of this family was carried out based on the hitherto largest number of cultures with a global distribution and diverse substrates. Total 1130 Didymellaceae strains from 76 countries, 98 plant families and 55 other substrates, including animals, fungi, soil, air, water, paint, glacier, were examined. The resulting phylogenetic tree of four loci（ITS, LSU, rpb2, tub2） resolved 38 well supported genera and recognized287 species. Among them, 7 new genera, 30 new species from plants, soil, coral, house dust were introduced.Besides, 20 new combinations were proposed, and five epitypes and four neotypes were designated to stabilize the taxonomy and use of elder names. On this basis, an extended phylogenetic backbone for Didymellaceae based on a more complete sequences dataset was generated, aiming to help the delimitation of novelties at the species,genus and family levels. A new kind of stem spot disease was detected on the stems of Aconitum carmichaeli（Chinese Wutou） in Gansu Province,China,in 2010.A Chaetosphaeronema-like fungus was isolated and completion of Koch’s postulates confirmed that the fungus was the causal agent of the stem spot disease.The morphology and molecular methods were combined to identify the pathogen.The conidiomata were thickwalled,250-600 μm wide and 150-500 μm high.The conidia were 0-1 septate,7.7（-11.4）-19.2μm×2.0（-2.8）×5.1μm.A multi-locus phylogenetic analysis of five loci,LSU,ITS,tefl-a,RPB2 and BTUB demonstrated that this Chaetosphaeronema-like fungus is closely related to C.hispidulum and C.achilleae.However,on the basis of larger pycnidia with no setae this pathogen is viewed as a new taxon of Chaetosphaeronema.
