Objective Abelson helper integration site 1（AHI1） gene is associated with several neuropsychiatric and brain developmental disorders such as schizophrenia, depression, autism and Joubert syndrome. AHI1 deficiency in mice leads to typical depressive behaviors. However, the mechanisms by which AHI1 regulates mental and behavioral processes are largely unknown. Methods Western blot and quantitative RT–PCR（q RT-PCR） were performed to examine the protein and m RNA levels. Immunohistochemistry and immunofluorescence was applied to examine the TH neuron activity and content in mice midbrains. Dual luciferease assay was used to examine the transcriptional activity. The interaction between AHI1 and RORα was identified by co-immunoprecipition assay. Depression-like behaviors were measured by tail suspension test（TST） and forced swim test（FST）. Results We found that down-regulation of tyrosine hydroxylase（TH） expression in Ahi1 knockout（KO） mouse midbrains is responsible for AHI1-deficiency-mediated mood and behavior disturbance. REV-ERBα, a TH transcriptional repressor, is up-regulated in Ahi1 KO mouse midbrains. Knockdown of Ahi1 decreases TH but increases REV-ERBα expressions, respectively. AHI1 interacts with RORα and represses RORα transcriptional activity to decrease BMAL1 expression, leading to a decrease of REV-ERBα, a downstream target of BMAL1. In BMAL1 deficient or REV-ERBα inhibitor SR8278-treated cells, the repression of TH expression by AHI1-deficiency is completely blocked. In addition, microinfusion of SR8278 into ventral midbrain（VMB） of Ahi1 KO mice dramatically improves their performance in depressive behaviors as well as increases TH expression in ventral tegmental area（VTA） and substantia nigra（SN）. Conclusion These findings provide a mechanistic explanation between AHI1-related behaviors and the circadian clock pathway, suggesting that the circadian clock pathway could be targeted as one of therapies in mood-related disorders.

BACKGROUND People with bipolar disorder(BD)frequently struggle with the recurrence of affective ***,the interplay between coping mechanism and positive mood state remains *** To explore the associations among behavioral approach system(BAS)sensitivity level,coping,and positive mood states among people with *** Using a cross-sectional study design,90 participants with BD were presented with four BAS-activating life event scenarios and assessed with regard to their BAS trait sensitivity,coping flexibility,and mood states.A hierarchical clustering method was used to identify different groups with different styles of *** hierarchical regression analyses were conducted to examine the mediating and moderating roles of different components of coping on mood *** A three-cluster solution was found to best fit the present data *** findings showed that a low mass of coping combined with low BAS sensitivity level protects people with BD from detrimentally accentuating mood states when they encounter BAS-activating life ***,coping flexibility is demonstrated to mediate and moderate the relationships between BAS sensitivity level and mood ***,subduing the perceived controllability and reducing the use of behavioral-activation/emotion-amplifying coping strategies could help buffer the effect of positive *** The judicious use of coping in emotion regulation for people with BD when encountering BAS-activating life events was *** applications and theoretical implications are highlighted.

Baicalin influences the dendritic morphology of newborn neurons in the hippocampus of chronically stressed rats

Chronic stress models, established in adult Sprague-Dawley rats through a 14-day subcutaneous injection of 40 mg/kg corticosterone, once per day, were given a daily oral feeding of 50 mg/kg baicalin. The study was an attempt to observe the effect of baicalin on neurogenesis in chronically stressed rats. Results showed that subcutaneous injection of corticosterone significantly decreased the total number of doublecortin-positive neurons in the hippocampus. The reduced cell number caused by corticosterone was mainly due to the decrease of class II doublecortin-positive neurons, but the class I doublecortin-positive neurons were unaffected. Baicalin treatment increased the number of both class I and class II doublecortin-positive neurons. In addition, doublecortin-positive neurons showed less complexity in dendritic morphology after corticosterone injection, and this change was totally reversed by baicalin treatment. These findings suggest that baicalin exhibits a beneficial effect on adult neurogenesis.
