Paternal inheritance of mitochondrial dna in the sheep (Ovine aries)

Paternal inheritance of mitochondria dna in sheep was discovered by examination of 152 sheep from 38 hybrid families for mtdna D-loop polymorphisms using PCR-RFLP, amplification of repeated sequence somain, and PCR-SSCP of the D-loop 5′ end region of a 253 bp fragment. Our findings have provided the first evidence of paternal inheritance of mtdna in sheep and possible mechanisms of paternal inheritance were discussed.

Pancreatic cancer is a more aggressive and refractory *** and toxicity limit drug ***,we report a lower toxic and higher effective miriplatin(MPt)-loaded liposome,LMPt,exhibiting totally different anti-cancer mechanism from previously reported platinum *** in gemcitabine(GEM)-resistant/sensitive(GEM-R/S)pancreatic cancer cells,LMPt exhibits prominent anti-cancer activity,led by faster cellular entry-induced larger accumulation of *** level of caveolin-1(Cav-1)determines entry rate and switch of entry pathways of LMPt,indicating a novel role of Cav-1 in nanoparticle *** endosome-lysosome processing,in unchanged metabolite,MPt is released and targets mitochondria to enhance binding of mitochondria protease LONP1 with POLG and TFAM,to degrade POLG and ***,via PINK1-Parkin axis,mitophagy is induced by POLG and TFAM degradation-initiated mitochondrial dna(mtdna)replication ***,POLG and TFAM are identified as novel prognostic markers of pancreatic cancer,and mtdna replication-induced mitophagy blocking mediates their pro-cancer *** findings reveal that the target of this liposomal platinum agent is mitochondria but not dna(target of most platinum agents),and totally distinct mechanism of MPt and other formulations of ***-assembly offers LMPt special efficacy and *** action and characteristic mechanism make LMPt a promising cancer candidate.

An improved method of mitochondrial dna isolation for XL-PCR

Objective： To obtain high quality of mitochondrial dna （mtdna） and carry out extra-long PCR （XL-PCR）. Methods： mitochondria were isolated by differential centrifugation, and membranes were disrupted using 10%SDS （pH 7.0）. mtdna was then extracted using phenol and chloroform. Resuits： The mtdna obtained by using our improved method can be used as effective template for XL-PCR, and total mtdna （16 kb） can be amplified easily. Conclusion： Our improved method is effective in preparing high quality of mtdna, which can be used as template for XL-PCR.

The phenotypic manifestations of mitochondrial dna(mtdna) mutations were modulated by mitochondrial haplotypes,nuclear modifier genes and environmental *** yeast mitochondrial 15S rRNA C1477Gmutation(PR) corresponds to human 12S rRNA C1494T and A1555G mutations,which are well known as a primary factor for aminoglycoside-induced and nonsyndromic *** we report that the nuclear modifier gene MT02 deletion suppressed the aminoglycoside-sensitivity of mitochondrial 15S rRNA C1477G mutation in Saccharomyces ***,the strain with a single mtdna C1477G mutation exhibited hypersensitivity to *** assays indicated that the steady-state level of mitochondrial dna,the mitochondrial respiratory rate,and the membrane potential decreased significantly after neomycin *** impaired mitochondria could not produce sufficient energy to maintain ***, when the mto2 null and the mitochondrial C1477G mutations co-existed(mto2(PR)),oxygen consumption rate in the double mutant decreased markedly compared to that of control strains(MT02(PS),mto2(PS) and MT02(PR)).The expression levels of key glycolytic genes HXK2,PFK1 and PYK1 in mto2(PR) strain were stimulated by neomycin and up-regulated by 89%,112%and 55%,*** enhanced glycolysis might compensate for the respiratory energy *** findings in yeast will provide a new insight into the pathogenesis of human deafness.
