China is a center of diversity for Malus Mill,with 27 native species including 21 wild species and six domesticated *** applied a set of 19 simple sequence repeat markers to genotype 798 accessions of 17 species(12 wild species and five cultivated species)of Malus originating from 14 provinces in China.A total of 500 alleles were *** statistics indicated a high level of genetic variation as quantified by the average values of the effective allele number(N_(e)).expected heterozygosity(H_(e)),and Shannon's Information Index(I)(10.309,0.886,and 2.545,respectively).Malus sieversii(MSR;H_(e)=0.814,I=2.041,N_(e)=6.054),***(MBB;H_(e)=0.848,/=2.350,N_(e)=8.652),***(MTH;He=0.663,I=1.355,N_(e)=3.332),and ***(MHR;H_(e)=0.539,I=0.912,N_(e)=0.579)showed a higher level of genetic diversity in this study than the previous *** and MBB contributed to the origin and evolution of some accessions of *** ***(MDC).However,other accessions of MDC showed a closer genetic distance with MBB and cultivated species,especially ***(MRB),***(MAN),and ***(MPB).Not all accessions of MDC were descended from MSR in Xinjiang Uygur Autonomous Region of *** research provides novel insights into the genetic relationships of Malus native to China,which will be useful for genetic association studies,germplasm conservation,and breeding programs.

The blue sheep is an endemic species to the Qinghai-Tibet Plateau and surrounding *** has been regarded as having 2 subspecies:Pseudois nayaur nayaur and ***,such a classification remains ***,we analyze 10 microsatellite loci and part of the mitochondrial control region for clarification in such taxonomic *** use samples from 168 individuals from 6 geographic populations covering almost all the distribution areas of the species in China to carry out *** trees derived from both the microsatellite and mitochondrial markers combined with the discriminant analysis of principal components(DAPC)and the STRUCTURE analysis reveal that the individuals in the Helan Mountains are well grouped with a distinct evolutionary lineage and are significantly different from the other populations of *** according to Fst values,implying that this isolated population should be categorized as a valid subspecies;namely,Pseudois nayaur *** isolation-by-distance(IBD)analysis shows a significant positive relationship between genetic and geographical distances among the populations.

Five Larix species(***,***,***,***,and *** ***),have survived on the Qinghai-Tibet Plateau(QTP)under specific climate conditions for *** lack of genomic information seriously hinders research on the evolution,conservation and ecology of these Larix *** this study,complete chloroplast(cp)genomes of the 5 species were assembled and compared based on next generation sequencing technology combined with polymerase chain reaction *** results show that the 5 cp genomes are relatively conservative in size,gene content and arrangement,and border *** analysis showed that the species are closely related as well as to seven other species of the same *** addition,the 5 cp genomes contained few simple sequence repeats and relatively low nucleotide variability;thus,12 candidate polymorphic cp DNA markers will be helpful for further research on relevant population *** results will provide valuable genetic information for the conservation,evolution and ecology of these species and their relatives.

Detailed information on the size and genetic structure of wildlife populations is critical for developing effective conservation strategies, especially for those species that have suffered population decline and fragmentation due to anthropogenic activities. In the present study, we used a non-invasive approach combining fecal pellet sampling with mitochondrial DNA and nuclear DNA microsatellite marker analysis to monitor and compare the population structure of the Chinese goral (Naemorhedus griseus) in Beijing and northeast Inner Mongolia in China. Of the 307 fecal samples confirmed to be from N. griseus, 15 individuals (nine females and six males) were found in the Beijing population and 61 individuals (37 females and 24 males) were found in the Inner Mongolian population. Among these 76 individuals, we identified eight haplotypes and 13 nucleotide polymorphic sites from mtDNA and 45 alleles from 10 microsatellite loci. Spatially structured genetic variation and a significant level of genetic differentiation were observed between the two populations. In both populations, the sex ratios were skewed toward females, indicating high reproductive potential, which is crucial for population recovery and conservation of this patchily distributed vulnerable species. We suggest that managing the two populations as evolutionarily significant units with diverse genetic backgrounds could be an effective solution for present population recovery, with the possible relocation of individuals among different groups to help ensure future goral species prosperity.

Migration and population genetics of the grain aphid Macrosiphum miscanti (Takahashi) in relation to the geographic distance and gene flow

microsatellites are found in taxonomically different organisms, and such repeats are related with genomic structure, function and certain diseases. To characterize microsatellites for macaques, we searched and compared SSRs with 1–6 bp nucleotide motifs in rhesus, cynomolgus and pigtailed macaque. A total of 1395671, 1284929 and 1266348 perfect SSRs were mined, respectively. The most frequent perfect SSRs were mononucleotide SSRs. The most GC-content was in dinucleotide SSRs and the least was in the mononucleotide SSRs. Chromosome size was positively correlated with SSR number and negatively correlated with the relative frequency and density of SSRs. The GC content of chromosome SSRs were negatively correlated with relative frequency of SSRs and GC content of chromosome sequences. The features of microsatellite distribution in assembled genomes of the three species were greatly similar, which revealed that the distributional pattern of microsatellites is probably conservative in genus Macaca. The degenerated number of repeat motifs was found to be different in pentanucleotide and hexanucleotide repeats. Species-specific motifs for each macaque were significantly underrepresented. Overall, SSR frequencies of each chromosome in rhesus macaque were higher than in cynomolgus macaque. The maximum repeat times of mono-to pentanucleotide repeats in cynomolgus macaque was more than other two macaques. These results emphasize the genetic diversity and phylogenetic relationship of genus Macaca species. Our data will be beneficial for comparative genome mapping, understanding the distribution of SSRs and genome structure between these animal models, and provide a foundation for further development and identification of more macaque-specific SSRs.

The population genetics of the grain aphid Macrosiphum miscanti (Takahashi) is analyzed by microsatellite markers. Samples collected from 15 locations in China have been examined at 5 polymorphic microsatellite loci. Overall, genetic diversity displays a relation between the migration and gene flow in the grain aphid; a free and frequent gene flow is found in the eastern populations, and gene isolation occurs in the two western populations, especially Datong population and Guiyang population. The natural barriers may present an insurmountable obstacle preventing gene flow and aphid migration. However, a spatial genetic differentiation between populations is correlated with their geographical separation, indicating the geographic differentiation may play an important role in shaping the genetic structure of M. miscanti populations. In addition, most populations of grain aphids are out of Hardy-Weinberg equilibrium and there is het-erozygote deficit. Based on F statistics, the average genetic differentiation among different geographical populations is relatively low. It seems that the long distance migration of the grain aphid may enhance gene flow and decrease genetic differentiation among different populations .
