The sex-biased dispersal and kinship dynamics are important factors shaping the spatial distribution of individuals and are key parameters affecting a variety of ecological and evolutionary processes.Here,we studied the spatial distribution of related individuals within a population of corn mice Calomys musculinus in a seasonal cycle to infer dispersal patterns.The sampling was carried out from spring 2005 to winter 2006 in field borders of intensively managed agroecosystems.Genotyping data from 346 individuals with 9 Microsatellites showed spatial genetic structure was weak for males,but not for females.The results indicate a complex spatial kinship dynamic of related females across all seasons.Which,contrary to our expectations,dispersal distances decrease with the increase of the population abundance.Meanwhile,male dispersal distances were greater when population abundance increased and thus the availability of active females.Males disperse greater distances to mate and sire offspring with distant females as a possible inbreeding avoidance mechanism.This study shows that C.musculinus is capable of much greater scattering distances than previously reported and that dispersal occurs fluidly and without barriers across the agroecosystem.The indirect benefit of dispersal on individual fitness could be related to relaxing the competition in the natal area and increasing the mating rate.Our study highlights the value of combining genetic relatedness,fieldwork observations,and behavioral data to estimate dispersal at a fine geographical scale.

Across two phylogeographic breaks: Quaternary evolutionary history of a mountain aspen (Populus rotundifolia) in the Hengduan Mountains

Biogeographical barriers to gene flow are central to plant phylogeography.In East Asia,plant distribution is greatly influenced by two phylogeographic breaks,the Mekong-Salween Divide and Tanaka-Kaiyong Line,however,few studies have investigated how these barriers affect the genetic diversity of species that are distributed across both.Here we used 14 Microsatellite loci and four chloroplast DNA fragments to examine genetic diversity and distribution patterns of 49 populations of Populus rotundifolia,a species that spans both the Mekong-Salween Divide and the Tanaka-Kaiyong Line in southwestern China.Demographic and migration hypotheses were tested using coalescent-based approaches.Limited historical gene flow was observed between the western and eastern groups of P.rotundifolia,but substantial flow occurred across both the Mekong-Salween Divide and Tanaka-Kaiyong Line,manifesting in clear admixture and high genetic diversity in the central group.Wind-borne pollen and seeds may have facilitated the dispersal of P.rotundifolia following prevalent northwest winds in the spring.We also found that the Hengduan Mountains,where multiple genetic barriers were detected,acted on the whole as a barrier between the western and eastern groups of P.rotundifolia.Ecological niche modeling suggested that P.rotundifolia has undergone range expansion since the last glacial maximum,and demographic reconstruction indicated an earlier population expansion around 600 Ka.The phylogeographic pattern of P.rotundifolia reflects the interplay of biological traits,wind patterns,barriers,niche differentiation,and Quaternary climate history.This study emphasizes the need for multiple lines of evidence in understanding the Quaternary evolution of plants in topographically complex areas.

A spaceborne advanced storage system for remote sensing Microsatellites

With the development of satellite miniaturization and remote sensing,the establishment of Microsatellite constellations is an inevitable trend.Due to their limited size,weight,and power,spaceborne storage systems with excellent scalability,performance,and reliability are still one of the technical bottlenecks of remote sensing Microsatellites.Based on the commercial off-the-shelf field-programmable gate array and memory devices,a spaceborne advanced storage system(SASS)is proposed in this paper.This work provides a dynamic programming,queue scheduling multiple-input multiple-output cache technique and a high-speed,high-reliability NAND flash controller for multiple Microsatellite payload data.Experimental results show that SASS has outstanding scalability with a maximum write rate of 2429 Mb/s and preserves at least 78.53%of the performance when a single NAND flash fails.The scheduling technique effectively shortens the data scheduling time,and the data remapping method of the NAND flash controller can reduce the retention error by at least 50.73%and the program disturbance error by at least 37.80%.

Preliminary study of the electrospray DPE peculiarities from the liquid surface in the presence of the CSWs

The Ultrasonic Electric Propulsion(UEP) system is a cutting-edge propulsion technology that is mostly used on platforms for small satellites(less than 10 kg). The characteristics of droplet partial emissions(DPEs) in the UEP system are investigated using a high-speed imaging technique(an ultra-high speed camera(NAC HX-6) and a long-distance microscope) in this work.The experiments demonstrate that there are a few partial emission modes, including left-side emission, double-side emission, and right-side emission, that are present in the droplet emission process of the UEP system. These modes are primarily caused by the partial formation of capillary standing waves(CSWs) on the emission surface of the ultrasonic nozzle. The emission rate for single-and double-sided emissions varies at different times, indicating that there are different CSWs engaged in droplet emission due to variations in the liquid film thickness and charge state of the liquid cones. Additionally, as the droplets emit continuously, a raised area on the emission surface appears, with several droplets emitting there as a result of charge accumulation. Additionally, photos of the CSWs with emitting droplets are obtained, which highlights the CSWs’ distinctive wave morphology.

Simulation of a soft-gamma-ray polarimeter on board a Microsatellite

Gamma-ray polarimetry is a new and prospective tool for studying extremely high-energy celestial objects and is of great significance for the field of astrophysics.With the rapid development of Microsatellite technology,the advantages of space exploration have become increasingly apparent.Therefore,we simulated a soft-gamma-ray polarimeter for a Microsatellite based on the Compton scattering principle.We performed detailed Monte Carlo simulations using monoenergetic gamma-ray linear-polarization sources and Crab-like sources in the energy range of 0.1-10 MeV considering the orbital background.The polarimeter exhibited excellent polarization detection performance.The modulation factor was 0.80±0.01,and the polarization angles were accurate within an error of 0.2°at 200 keV for on-axis incidence.For the Crab-like sources for on-axis incidence,the polarization degrees were consistent with the set values within the error tolerance,the modulation factor was 0.76±0.01,and the minimum detectable polarization reached 2.4%at 3σfor an observation time of10^(6) s.Additionally,the polarimeter exhibited recoil electron tracking,imaging,and powerful background suppression in a large field of view(FoV;∼2πsr).The proposed polarimeter meets the requirements of a space soft-gamma-ray polarization detector and has promising research prospects.

Genome-wide development of interspecific Microsatellite markers for Saccharum officinarum and Saccharum spontaneum

Sugarcane has a large,complex,polyploid genome that has hindered the progress of genomic research and molecular marker-assisted selection.The user-friendly SSR markers have attracted considerable attention owing to their ideal genetic attributes.However,these markers were not characterized and developed at the genome-wide scale due to the previously lacking high-quality chromosome-level assembled sugarcane genomes.In this present study,744305and 361638 candidate SSRs were identified from the genomes of S.officinarum and S.spontaneum,respectively.We verified the reliability of the predicted SSRs by using 1200 interspecific SSR primer pairs to detect polymorphisms among 11 representative accessions of Saccharum,including S.spontaneum,S.officinarum,S.robustum,and modern sugarcane hybrid.The results showed that 660 SSR markers displayed interspecific polymorphisms among these accessions.Furthermore,100 SSRs were randomly selected to detect the genetic diversity for 39 representative Saccharum accessions.A total of 320 alleles were generated using 100 polymorphic primers,with each marker ranging from two to seven alleles.The genetic diversity analysis revealed that these accessions were distributed in four main groups,including group I(14 S.spontaneum accessions),group II(two S.officinarum accessions),group III(18 modern sugarcane hybrid accessions),and group IV(five S.robustum accessions).Experimental verification supported the reliability of the SSR markers based on genome-wide predictions.The development of a large number of SSR markers based on wet experiments is valuable for genetic studies,including genetic linkage maps,comparative genome analysis,genome-wide association studies,and marker-assisted selection in Saccharum.

Analysis of genetic diversity and population structure of Chinese yak breeds (Bos grunniens) using Microsatellite markers

Nine Chinese yak breeds （Maiwa, Tianzhu White, Qinghai Plateau, Sibu, Zhongdian, Pali, Tibetan High Mountain, Jiulong, and Xinjiang） and Gayal were analyzed by means of 16 Microsatellite markers to determine the level of genetic variation within populations, genetic relationship between populations, and population structure for each breed. A total of 206 Microsatellite alleles were observed. Mean F-statistics （0.056） for 9 yak breeds indicated that 94.4% of the genetic variation was observed within yak breeds and 5.6% of the genetic variation existed amongst breeds. The Neighbor-Joining phylogenetic tree was constructed based on Nei＇s standard genetic distances and two clusters were obtained. The Gayal separated from the yaks far away and formed one cluster and 9 yak breeds were grouped together. The analysis of population structure for 9 yak breeds and the Gayal showed that they resulted in four clusters; one cluster includes yaks from Tibet Autonomous Region and Qinghai Province, one cluster combines Zhongdian, Maiwa, and Tianzhu White, and Jiulong and Xinjiang come into the third cluster. Pali was mainly in the first cluster （90%）, Jiulong was mainly in the second cluster （87.1%）, Zhongdian was primarily in the third cluster （83%）, and the other yak breeds were distributed in two to three clusters. The Gayal was positively left in the fourth cluster （99.3%）.

The analysis of genetic diversity and differentiation of six Chinese cattle populations using Microsatellite markers

A total of 321 individuals from six cattle populations of four species in a bovine subfamily in China were studied using 12 pairs of Microsatellite markers. The genetic diversities within and between populations were calculated. The phylogenetic trees were constructed by （δμ）^2 and DA distances, and the divergence times between populations were estimated by （δμ）^2. Altogether, 144 Microsatellite alleles were detected including 24 private alleles and nine shared alleles. Chinese Holstein had the largest number of private alleles （10）, whereas Bohai black and Buffalo had the smallest number of private alleles （2）. Chinese Holstein showed the highest genetic variability. Its observed number of alleles （Na）, mean effective number of alleles （MNA）, and mean heterozygosity （He） were 7.7500, 4.9722, and 0.7719, respectively, whereas, the Buffalo and Yak showed low genetic variability. In the phylogenetic trees, Luxi and Holstein grouped first, followed by Bohai and Minnan. Yak branched next and buffalo emerged as the most divergent population from other cattle populations. Luxi and Bohai were estimated to have diverged 0.039-0.105 million years ago （MYA）, however, buffalo and Holstein diverged 0.501-1.337 MYA. The divergence time of Yak versus Minnan, Holstein and buffalo was 0.136-0.363, 0.273-0.729, and 0.326-0.600 MYA, respectively.

Using MiddRAD-seq data to develop polymorphic Microsatellite markers for an endangered yew species

Microsatellites are highly polymorphic markers which have been used in a wide range of genetic studies.In recent years, various sources of next-generation sequencing data have been used to develop new Microsatellite loci, but compared with the more common shotgun genomic sequencing or transcriptome data, the potential utility of RAD-seq data for Microsatellite ascertainment is comparatively under-used.In this study, we employed MiddRAD-seq data to develop polymorphic Microsatellite loci for the endangered yew species Taxus florinii. Of 8,823,053 clean reads generated for ten individuals of a population, 94,851(~1%) contained Microsatellite motifs. These corresponded to 2993 unique loci, of which 526(~18%) exhibited polymorphism. Of which, 237 were suitable for designing Microsatellite primer pairs, and 128 loci were randomly selected for PCR validation and Microsatellite screening. Out of the 128 primer pairs, 16 loci gave clear, reproducible patterns, and were then screened and characterized in 24 individuals from two populations. The total number of alleles per locus ranged from two to ten(mean=4.875), and within-population expected heterozygosity from zero to 0.789(mean = 0.530),indicating that these Microsatellite loci will be useful for population genetics and speciation studies of T. florinii. This study represents one of few examples to mine polymorphic Microsatellite loci from ddRAD data.

Microsatellite instability(MSI)is a key biomarker for cancer therapy and prognosis.Traditional experimental assays are laborious and time-consuming,and next-generation sequencingbased computational methods do not work on leukemia samples,paraffin-embedded samples,or patient-derived xenografts/organoids,due to the requirement of matched normal samples.Herein,we developed MSIsensor-pro,an open-source single sample MSI scoring method for research and clinical applications.MSIsensor-pro introduces a multinomial distribution model to quantify polymerase slippages for each tumor sample and a discriminative site selection method to enable MSI detection without matched normal samples.We demonstrate that MSIsensor-pro is an ultrafast,accurate,and robust MSI calling method.Using samples with various sequencing depths and tumor purities,MSIsensor-pro significantly outperformed the current leading methods in both accuracy and computational cost.MSIsensor-pro is available at https://***/xjtu-omics/msisensor-pro and free for non-commercial use,while a commercial license is provided upon request.
