breast cancer and metastasis remain great challenges in clinical *** with monotherapy,combination therapy,especially mediated by nanomedicine delivery strategy,significantly improves the therapeutic efficacy and reduces undesired ***-2(COX-2)inhibitors are widely used for adjuvant chemotherapy because COX-2 is overexpressed in virtually all cancer cell lines to regulate tumor progression by catalyzing prostaglandin E2(PGE2)*** drug combination strategy is still required to be improved due to some unsatisfactory clinical trial *** processes of tumor growth and metastasis are orchestrated by multiple proteins in addition to COX-2,which are modified and transported by Golgi ***,disrupting the structure and function of Golgi apparatus can inhibit the secretion of tumor-related proteins and further suppress carcinoma progression and *** COX-2 is also enriched within Golgi apparatus in tumor cells,COX-2 inhibitors and Golgi disrupting agents can be co-delivered to Golgi apparatus to maximize the *** this work,we developed a human serum albumin(HSA)nanoparticle encapsulating pirarubicin(THP),retinoic acid(RA),and indomethacin(IMC),called TIR-HSA,which was observed to be localized in Golgi complex of 4T1 *** to the synergistic effect of these three drugs,TIR-HSA inhibited the proliferation,migration,and invasion of tumor cells,enhanced the apoptotic rate,and improved the immunosuppressive tumor microenvironment,which remarkably regressed the tumor growth and metastasis and prolonged the survival period of 4T1-bearing mice.

Association of SIPA1545 C>T polymorphism with survival in Chinese women with metastatic breast cancer

It has been demonstrated that single nucleotide polymorphisms(SNPs)of SIPA1(signal-induced proliferation associated gene 1)are associated with metastatic efficiency in both human and *** purpose of this study was to determine whether SIPA1545 C>T polymorphism was associated with overall survival in patients with metastatic breast *** this study,SIPA1545 C>T polymorphism was detected in 185 metastatic breast cancer patients using polymerase chain reaction-restriction fragment length polymorphism assay(PCR-RFLP).Survival curves for patients with SIPA1545 C>T polymorphism was compared using the Kaplan-Meier method with log-rank *** found that SIPA1545 C>T polymorphism was significantly associated with survival in 185 patients with metastatic breast *** with SIPA1545 T/T genotype had a significantly worse overall survival(OS)than did patients with C/T or C/C genotype(50.0%vs.62.9%,P=0.042).Moreover,in multivariate analysis,as compared with the C/C or C/T genotype,the T/T genotype remained an independent unfavorable prognostic marker of OS in this cohort(hazard ratio[HR]=2.16;95%CI=1.12–4.15;P=0.022).Our findings indicate that metastatic breast cancer patients with SIPA1545 T/T genotype have a poorer survival compared to patients with C/C or C/T genotype.

Endocrine therapy(ET)remains the mainstay of treatment for steroid hormone receptor-positive,human epidermal growth factor 2(HER2)-negative metastatic breast cancer(MBC).Tumor resistance to hormone therapy has led to the development of novel endocrine drug combinations,transforming the landscape of MBC *** options for ET are expanding,with promising agents in the *** MBC remains incurable,many patients can enjoy years of survival with good quality of life by cycling through the many available *** the plethora of available agents and rapid approvals,clinicians look to evidencebased guidelines to assist in treatment selection to maximize patient *** this review,we provide a contemporary review of the advances in ET and a suggested algorithm to guide clinicians in daily management of patients with hormone receptor-positive,HER2-negative *** will discuss landmark trials and highlight their impact in reshaping treatment ***,we will provide a glimpse into advances on the horizon and the promise they bring to improve outcomes in patients with advanced breast cancer.

Effectiveness of second-line anti-HER2 treatment in HER2-positive metastatic breast cancer patients previously treated with trastuzumab:A real-world study

Background: A phase II clinical trial of the comparison between eribulin plus gemcitabine (EG) and paclitaxel plus gemcitabine (PG) as first-line chemotherapy for patients with metastatic breast cancer (MBC) found that the EG regimen was less neurotoxic, but was similar in efficacy to the PG regimen. In the present study, we analyzed functional assessment of cancer therapy-taxane (FACT-Taxane) questionnaires from patients in this clinical trial to determine their quality of life (QoL). Methods: QoL was assessed using the Korean version of the FACT-Taxane questionnaires. After baseline assessment, QoL was assessed every 2 cycles for 12 cycles and every 3 cycles thereafter. The linear mixed model was used to evaluate the difference in QoL between the EG and PG arms. Results: Of the 118 enrolled patients, 117 responded to the FACT-Taxane questionnaires at baseline, 1 in the PG arm did not. Baseline QoL scores were not different between the EG and PG arms. During treatment, taxane subscale scores were significantly higher in the PG arm than in the EG arm after 2-13 cycles of chemotherapy (all P < 0.05), except for the 11th cycle. Neuropathy-specific analysis showed that patients in the PG arm had earlier and more severe neuropathic symptoms than those in the EG arm (P < 0.001). Conclusions: In our QoL analysis, the EG regimen delayed and decreased neuropathy as compared with the PG regimen. Therefore, eribulin would be a reasonable substitute for paclitaxel as first-line chemotherapy for MBC.
