In recent decades,the global prevalence of metabolic syndrome has surged,posing a significant public health *** disorders,encompassing diabetes,obesity,nonalcoholic fatty liver disease,and polycystic ovarian syndrome,have been linked to alterations in the gut ***,the connection between gut microbiota and host metabolic diseases warrants further *** this review,we delve into the associations between various metabolic disorders and the gut microbiota,focusing on immune responses and bile acid(BA)***,T helper cells,innate lymphoid cells,macrophages,and dendritic cells have been shown to modulate host metabolism through interactions with intestinal microorganisms and the release of ***,secondary BA metabolites,derived from the microbiota,are involved in the pathogenesis of metabolic diseases via the farnesoid X receptor and Takeda G protein-coupled receptor *** covering both aspects of this immune system-microorganism axis,we present a comprehensive overview of the roles played by the gut microbiota,microbiota-derived BA metabolites,and immune responses in metabolic diseases,as well as the interplay between these systems.

Circadian rhythms play crucial roles in orchestrating diverse physiological processes that are critical for health and *** circadian rhythms are closely associated with various human metabolic diseases,including type 2 diabetes,cardiovascular disease,and non-alcoholic fatty liver *** lifestyles are frequently associated with an irregular circadian rhythm,which poses a significant risk to public *** the central clock has a set periodicity,circadian oscillators in peripheral organs,particularly in the liver,can be entrained by metabolic alterations or stress *** the molecular level,the signal transduction pathways that mediate stress responses interact with the key determinants of circadian oscillation to maintain metabolic homeostasis under physiological or pathological *** the liver,a number of nuclear receptors or transcriptional regulators,which are regulated by metabolites,hormones,the circadian clock,or environmental stressors,serve as direct links between stress responses and circadian *** this review,we summarize recent advances in the understanding of the interactions between stress responses(endoplasmic reticulum stress response,oxidative stress response,and inflammatory responses)and circadian metabolism,and the role of these interactions in the development of metabolic diseases.

Obesity has a multifactorial etiology and is known to be a state of chronic low-grade inflammation,known as *** state is associated with the development of metabolic disorders such as glucose intolerance and nonalcoholic fatty liver *** is a glycolytic metabolite and a crucial node in various metabolic ***,its role and molecular mechanism in obesity and associated complications are *** this study,we reported that pyruvate substantially inhibited adipogenic differentiation in vitro and its administration significantly prevented HFD-induced weight gain,white adipose tissue inflammation,and metabolic *** identify the target proteins of pyruvate,drug affinity responsive target stability was employed with proteomics,cellular thermal shift assay,and isothermal drug response to detect the interactions between pyruvate and its molecular ***,we identified cytosolic phospholipase A2(cPLA2)as a novel molecular target of pyruvate and demonstrated that pyruvate restrained diet-induced obesity,white adipose tissue inflammation,and hepatic steatosis in a cPLA2-dependent *** with global ablation of cPLA2 in mice showed that the protective effects of pyruvate were largely abrogated,confirming the importance of pyruvate/cPLA2 interaction in pyruvate attenuation of inflammation and ***,our study not only establishes pyruvate as an antagonist of cPLA2 signaling and a potential therapeutic option for obesity but it also sheds light on the mechanism of its ***’s prior clinical use indicates that it can be considered a safe and viable alternative for obesity,whether consumed as a dietary supplement or as part of a regular diet.

It has been well established that most of the age-related diseases such as insulin resistance,type 2 diabetes,hypertension,cardiovascular disease,osteoporosis,and atherosclerosis are all closely related to metabolic *** the other hand,interventions on metabolism such as calorie restriction or genetic manipulations of key metabolic signaling pathways such as the insulin and mTOR signaling pathways slow down the aging process and improve healthy *** findings raise an important question as to whether improving energy homeostasis by targeting certain metabolic signaling pathways in specific tissues could be an effective antiaging *** a more comprehensive understanding of the tissue-specific roles of distinct metabolic signaling pathways controlling energy homeostasis and the crosstalks between these pathways during aging may lead to the development of more effective therapeutic interventions not only for metabolic dysfunction but also for aging.
