BACKGROUND Vestigial like family member 3(VGLL3)is associated with the prognosis of epithelial ovarian cancer and soft tissue sarcoma,but its role in gastric cancer(GC)is unclear.AIM To explore the expression pattern and clinical significance of VGLL3 in GC.METHODS Integrative analysis was performed on the GC transcriptome profiles and survival information deposited in the ONCOMINE,GEPIA,and ONCOLNC databases.The expression levels of VGLL3 mRNA and protein were analyzed in the freshly resected tumor and normal gastric tissues from GC patients by quantitative RT-PCR and Western blot,respectively.In addition,the in situ expression of VGLL3 in the GC tissues was determined by immunohistochemistry(IHC),and the patients were accordingly classified into the high and low expression groups.The correlation of VGLL3 expression status with patient prognosis was then determined by univariate and multivariate Cox regression analyses.RESULTS Analysis of the ONCOMINE and GEPIA databases showed that VGLL3 was significantly up-regulated in GC tissues(P=0.003),and associated with the tumor TNM stage(P=0.0163).The high VGLL3 expression group had a significantly worse prognosis compared to the low expression group,as per both GEPIA(P=0.0057)and ONCOLNC(P=0.01).The bioinformatics results were validated by the significantly higher VGLL3 mRNA and protein levels in the GC tissues compared to the adjacent normal tissues(P<0.001)in a cohort of 30 GC patients.Furthermore,high in situ expression of VGLL3 protein was associated with more advanced N and TNM stages and HER2 mutation(P<0.05)in a cohort of 172 patients.Kaplan-Meier analysis showed that the high VGLL3 expression group had a worse prognosis compared to the low expression group(P=0.019).Multivariate analysis showed that VGLL3 expression status was an independent risk factor for prognosis.In addition,the prognostic risk model nomogram showed that VGLL3 was the most important indicator,with an area under the receiver operating characteristic(ROC)curve(AUC)of 0.613 for 3-year survival

Pore Size Distribution of a Tight Sandstone Reservoir and its Effect on Micro Pore-throat Structure: A Case Study of the Chang 7 member of the Xin’anbian Block, Ordos Basin, China

Pore distribution and micro pore-throat structure characteristics are significant for tight oil reservoir evaluation, but their relationship remains unclear. This paper selects the tight sandstone reservoir of the Chang 7 member of the Xin’anbian Block in the Ordos Basin as the research object and analyzes the pore size distribution and micro pore-throat structure using field emission scanning electron microscopy(FE-SEM), high-pressure mercury injection(HPMI), highpressure mercury injection, and nuclear magnetic resonance(NMR) analyses. The study finds that:(1) Based on the pore size distribution, the tight sandstone reservoir is characterized by three main patterns with different peak amplitudes. The former peak corresponds to the nanopore scale, and the latter peak corresponds to the micropore scale. Then, the tight sandstone reservoir is categorized into three types: type 1 reservoir contains more nanopores with a nanopore-to-micropore volume ratio of 82:18;type 2 reservoir has a nanopore-to-micropore volume ratio of 47:53;and type 3 reservoir contains more micropores with a nanopore-to-micropore volume ratio of 35:65.(2) Affected by the pore size distribution, the throat radius distributions of different reservoir types are notably offset. The type 1 reservoir throat radius distribution curve is weakly unimodal, with a relatively dispersed distribution and peak ranging from 0.01 μm to 0.025 μm. The type 2 reservoir’s throat radius distribution curve is single-peaked with a wide distribution range and peak from 0.1 μm to 0.25 μm. The type 3 reservoir’s throat radius distribution curve is single-peaked with a relatively narrow distribution and peak from 0.1 μm to 0.25 μm. With increasing micropore volume, pore-throat structure characteristics gradually improve.(3) The correlation between micropore permeability and porosity exceeds that of nanopores, indicating that the development of micropores notably influences the seepage capacity. Tight Carbonate Microstructure and its Controls: A Case Study of Lower Jurassic Da'anzhai member, Central Sichuan Basin

Cast thin section observation, scanning electron microscopy(SEM), high-pressure mercury injection(HPMI), and nuclear magnetic resonance(NMR) were used to examine the microstructure of tight carbonate reservoirs in the Lower Jurassic Da’anzhai member, the central Sichuan Basin. The pore space in the Da’anzhai member is classified into 2 types and 17 subtypes, with nano-scale pore throats of ‘O’, ‘S’, ‘Z’, and ‘I’ shapes. Poorly sorted pore throats vary greatly in diameter;thus, it is difficult for fluid flow to pass through these pore throats. There are three classes of pore throats in carbonate reservoirs, i.e. isolated pores, pores coexisting with fractures, and large pores and fractures. Isolated pores may provide some pore space, but the permeability is low. Pores and fractures coexisting in the reservoir may have a great impact on porosity and permeability;they are the major pore space in the reservoir. Large pores and fractures have a great impact on reservoir properties, but they only account for a limited proportion of total pore space. The microstructure of Da’anzhai reservoirs, which dominates fluid mobility, is dependent on sedimentary environment, diagenesis, and tectonic process. Pore structure is related to sedimentary environment. The occurrence of microfractures, which may improve reservoir properties, is dependent on tectonic process. Diageneses are of utmost importance to pore evolution, cementation and growth of minerals have played an important role in destroying reservoir microstructure.

Effect of Shale Reservoir Characteristics on Shale Oil Movability in the Lower Third member of the Shahejie Formation, Zhanhua Sag

To reveal the effect of shale reservoir characteristics on the movability of shale oil and its action mechanism in the lower third member of the Shahejie Formation(Es3l), samples with different features were selected and analyzed using N2 adsorption, high-pressure mercury injection capillary pressure(MICP), nuclear magnetic resonance(NMR), high-speed centrifugation, and displacement image techniques. The results show that shale pore structure characteristics control shale oil movability directly. Movable oil saturation has a positive relationship with pore volume for radius > 2 μm, as larger pores often have higher movable oil saturation, indicating that movable oil is present in relatively larger pores. The main reasons for this are as follows. The relatively smaller pores often have oil-wetting properties because of organic matter, which has an unfavorable effect on the flow of oil, while the relatively larger pores are often wetted by water, which is helpful to shale oil movability. The rich surface provided by the relatively smaller pores is beneficial to the adsorption of immovable oil. Meanwhile, the relatively larger pores create significant pore volume for movable oil. Moreover, the larger pores often have good pore connectivity. Pores and fractures are interconnected to form a complex fracture network, which provides a good permeability channel for shale oil flow. The smaller pores are mostly distributed separately;thus, they are not conducive to the flow of shale oil. The mineral composition and fabric macroscopically affect the movability of shale oil. Calcite plays an active role in shale oil movability by increasing the brittleness of shale and is more likely to form micro-cracks under the same stress background. Clay does not utilize shale oil flow because of its large specific surface area and its block effect. The bedding structure increases the large-scale storage space and improves the connectivity of pores at different scales, which is conducive to the movability of shale oil.

Sedimentary and geochemical characteristics of the Triassic Chang 7 member shale in the Southeastern Ordos Basin,Central China

The Ordos Basin is the largest petroliferous basin in China, where the Chang 7 member shale serves as the major source rock in the basin, with an area of more than 100,000 km^2 So far, sedimentary and geochemical characterizations have rarely been conducted on the shale in shallow(member in the Tongchuan area were observed and sampled, and sedimentary and geochemical characterizations were conducted for the well-outcropped YSC section. The study results show that the Chang7 member shale is widely distributed laterally with variable thickness. The organic-rich shale is 7-25 m thick in total and exhibits obvious horizontal variation in mineral composition. In the eastern sections, the shale contains organic matter of TypeⅡ_2-Ⅲ and is low in thermal maturity, with high clay mineral content, low K-feldspar content, and no pyrite. In the western sections, the shale contains Type Ⅱ_1 organic matter and is low in thermal maturity, with high clay mineral, K-feldspar, and pyrite contents. The YSC section reveals three obvious intervals in vertical mineral composition and organic abundance.The Chang 7 member organic-rich shale(TOC > 10%) contains mainly sapropelite and liptinite, with Type Ⅱ kerogen. It is generally characterized by a hydrocarbon potential of more than 70 mg/g, low maturity, and shallow-semideep lacustrine facies. In the western sections, the shale, still in a low maturity stage, has a higher hydrocarbon potential and is optional for shale oil recovery. However, the Chang 7 member shale in the study area is highly heterogeneous and its shale oil recovery is practical only in the organic-rich intervals.

The gut hormone apelin is a major therapeutic focus for several diseases involving inflammation and aberrant cell growth. We investigated whether apelin-36 contained alternative bioactive peptides associated with normal physiology or disease. Amino acid sequence analysis of apelin-36 identified an amidation motif consistent with the formation of a secondary bioactive peptide (SCNH2). SCNH2 is proven to be mitogenic and chemotactic in normal/malignant cells and augments angiogenesis via a PTX-resistant/CT-X-sensitive G protein-coupled receptor (GPCR). Notably, SCNH2 is substantially more potent and sensitive than apelin-13 and vascular endothelial growth factor-A. Endogenous SCNH2 is highly expressed in human tumors and placenta and in mouse embryonic tissues. Our findings demonstrate that SCNH2 is a new apelinergic member with critical pluripotent roles in angiogenesis related diseases and embryogenesis via a non-APJ GPCR.

Rolling Fatigue Test of Large-Sized UHPC member for Cable Stayed Bridge

Recently, research strives to apply Ultra High Performance Concrete (UHPC) to large-sized structures owing to its remarkable mechanical performance and durability compared to normal concrete. The Korea Institute of Construction Technology proposed SuperBridge800, an edge girder type UHPC cable stayed bridge with central span of 800 m, through its detailed design. The bridge is designed to be erected through the connection of precast UHPC segments. The precast UHPC segment is monolithically composed of one ribbed deck slab and edge girders at each side. The connection between the precast segments is achieved by steel bars at the edge girders and by UHPC cast-in-place wet joint at the slab. Despite of the outstanding mechanical performance of UHPC, the fabrication of large-sized members is a difficult task since UHPC hardens faster than normal concrete and requires a special curing process. Therefore, the constructability of large-sized UHPC segment should be secured to achieve SuperBridge800. Besides, the performance of the connection between segments should also be guaranteed, especially in terms of the fatigue performance of the UHPC cast-in-place joint, which constitutes a weak point. To that goal, two half-scaled UHPC segments are manufactured and the constructability is examined by fabricating a large-sized UHPC member connected with respect to the design conditions. This study conducts rolling fatigue test on the so-fabricated large-sized UHPC member. Rolling fatigue test is carried out up to 2 million cycles considering actual vehicle load at each center and quarter points of the member. The test results confirm that the service limit state is satisfied.

Kinetic simulation of hydrocarbon generation and its application to in-situ conversion of shale oil

The kinetic parameters of hydrocarbon generation are determined through experimental simulation and mathematical calculation using four typical samples selected from the Cretaceous Nenjiang Formation in the northwest of Songliao Basin,Chang 7 member of Triassic Yanchang Formation in the southwest of Ordos Basin,Paleogene in the southwest of Qaidam Basin,and Lucaogou Formation of Jimusar Sag in the east of Junggar Basin.The results show that activation energy of hydrocarbon generation of organic matter is closely related to maturity and mainly ranges between 197 kJ/mol and 227 kJ/mol.On this basis,the temperature required for organic matter in shale to convert into oil was calculated.The ideal heating temperature is between 270℃and 300℃,and the conversation rate can reach 90%after 50-300 days of heating at constant temperature.When the temperature rises at a constant rate,the temperature corresponding to the major hydrocarbon generation period ranges from 225 to 350℃at the temperature rise rate of 1-150℃/month.In order to obtain higher economic benefits,it is suggested to adopt higher temperature rise rate(60-150℃/month).The more reliable kinetic parameters obtained can provide a basis for designing more reasonable scheme of in-situ heating conversion.
