Ulcerative colitis （UC） is a chronic inflammatory disease and its involvement area in colon is influenced by a complex network of gene interactions. We analyzed the weighted gene co-expression networks in mieroarray dataset from colonic mucosa of patients with UC and identified one gene co-expression module that was highly associated with the progression of involved area in UC colon （Pearson coefficient=0.81, P〈0.0001）. In total, 523 hub genes in this module were found to be involved in immune system process after enrichment analysis in Gene Ontology. By the STRING and Cytoscape analysis, we observed that interleukin-8 （IL- 8） and matrix metalloproteinase-9 （MMP-9） were centered in the network of hub genes. We then detected the expression of IL-8 and MMP-9 in mucosa from left-sided colon of patients using quantitative PCR and immunofluorescence assay respectively. Both quantitative PCR and immunofluorescence assay revealed the expression levels of IL-8 and MMP-9 were significantly different among the healthy controls, left-sided colitis group and pancolitis group （P〈0.05）. IL-8 and MMP-9 were detected with an enhanced expression in pancolitis as compared with leftsided colitis and healthy controls, respectively （P〈0.05）. This study demonstrates that immune system process is indispensable in the progression of disease in colon, and identifies that IL-8 and MMP-9 play potential critical roles for the progression.

AIM: To investigate the expression of matrix metalloproteinase-2 (MMP-2) and matrix metalloproteinase-9 (MMP-9) in retinoblastoma (Rb), and their relationships with tumor development stage. METHODS: Immunohistochemical technique was used to detect the expression of MMP-2 and MMP-9 in 41 cases of paraffin embedded Rb samples. Quantitative analysis of the expression of MMP-2 and MMP-9 was assessed by HMIAS-2000 Color Pathologic Analysis System. The differences of the expression of MMP-2 and MMP-9 in each clinical and pathological stage were analyzed statistically. RESULTS: In all the 41 Rb specimens, MMP-2 and MMP-9 expression was found in tumor cells. The expression of MMP-2 and MMP-9 was significantly higher in tumors with optic nerve invasion than in tumors without optic nerve invasion (P9 was significantly higher in tumors of extra-ocular stage than in tumors of glaucomatous stage or intra-ocular stage( P9 exist in retinoblastoma cells. The level of MMP-2 and MMP-9 is related to optic nerve invasion and clinical stage of Rb, which suggests the expression of MMP-2 and MMP-9 could be connected to the invasion and development of tumor cells. Further research is needed for deeper understanding of the biological behavior and better evaluation of the prognosis of Rb.

OBJECTIVE To investigate the significance and relationship between matrix metalloproteinase 9 （MMP-9） and infiltration of macrophages in the process of invasion and metastasis in oral squamous cell carcinoma （OSCC）. METHODS The immunohistochemical SABC method was used to detect the expression of MMP-9 and CD68 （for labeling macrophages） in 42 cases of OSCC and in 10 normal tissues. RESULTS The expression of MMP-9 and macrophage counts in the OSCC cases were significantly higher compared to normal tissues （P〈0.05）. The expression of MMP-9 and macrophage counts were related to lymphnode metastasis and the TNM stage （P〈0.05）, showing that there was a positive correlation among these parameters （y = 0.443, ,0〈0.01）. CONCLUSION Both MMP-9 and macrophages may play an important role in the process of invasion and metastasis in OSCC, and this cellular activity may relate to the macrophages which affect the tumor cells and upregulate the expression of MMP-9.

AIM:To evaluate the protective role of AE-941,a matrix metalloproteinase(MMP) inhibitor,on ulcerative colitis(UC) in ***:Sprague Dawley(SD) rats were randomly divided into three groups:a control group,an AE-941 treatment group,and an UC model *** were sacrificed on days 7,21,or 56 following administration of treatment by enema and the disease activity index(DAI),colonic mucosa damage index(CMDI) and colonic expression of MMP-2 and MMP-9 were ***:DAI and CDMI scores in the UC model group increased significantly compared to the control group at all timepoints(P 941-treated group(DAI:21-and 56-d = 2.09 ± 0.25,1.52 ± 0.30 vs 1.55 ± 0.28,0.59 ± 0.19,respectively,P = 0.040 and 0.007,CMDI:21-and 56-d = 3.03 ± 0.42,1.60 ± 0.35 vs 2.08 ± 0.46,0.86 ± 0.37,respectively,P = 0.040 and 0.005).Furthermore,the colonic expression of MMP-2 and MMP-9 in the UC model group increased significantly compared to the control group(P 941-treated group on the 21-and 56-d timepoints(MMP-2:21-and 56-d = 0.6048 ± 0.0522,0.4163 ± 0.0330vs 0.3983 ± 0.0218,0.1093 ± 0.0072,respectively,P = 0.010;MMP-9:21-and 56-d = 0.6873 ± 0.0472,0.4328 ± 0.0257vs 0.5179 ± 0.0305,0.2673 ± 0.0210,respectively,P = 0.010 and 0.040).CONCLUSION:Expression of MMP-2 and MMP-9 increased significantly in rats with ***-941 can reduce colonic mucosal damage by downregulating the expression of MMP-2 and MMP-9.

AIM： To determine the proteolytic activity and expression of gelatinase B in serum of gastric cancer patients and their correlation with the stage of the tumor. METHODS： Sera from 23 patients who underwent surgery for primary gastric cancer as the experimental group and from 11 as the control group were used to determine the proteolytic activity and its inhibition by EDTA and 1,10-phenanthroline. Gelatinase B activity was detected by SDS polyacrylamide gel electrophoresis （SDS-PAGE） and SDS-PAGE zymography. RESULTS： Proteolytic enzyme activity was increased in gastric cancer patients when compared to the control group （,0〈0.05）. The proteinases were determined to be metalloproteinases upon inhibition test with specific metalloproteinase inhibitors 1,10-phenanthroline （P〈0.05） and EDTA （P〈0.01）. SDS-PAGE and SDS-PAGE zymography revealed gelatinase B （proMMP-9） activity and its molecular mass of 92 ku. CONCLUSION： Proteinase activity is overexpressed in serum of gastric cancer patients. Background The changes in matrix metalloproteinase-2 (MMP-2) and matrix metalloproteinase-9 (MMP-9) expressions were examined in the kidneys of diabetic rats to investigate the degradative pathway of collagen type Ⅳ (C-Ⅳ) and the protective effects of pioglitazone on an experimental model of diabetic *** In 54 SD rats used in our study, 18 served as normal controls. Diabetes mellitus was induced in 36 age- and weight-matched rats by intraperitoneal injection of streptozotocin (70 mg/kg); 18 of the diabetic rats were allocated at random to receive pioglitazone (20 mg·kg -1·d -1) in their drinking water and 18 served as diabetic controls. Rats were killed after 2, 4, or 8 weeks of treatment. Kidneys were examined pathomorphologically and the expressions of MMP-2, MMP-9, and C-Ⅳ were analyzed by immunohistochemistry, and the results were quantified by image analysis *** Diabetes mellitus was associated with a decrease in the expression of MMP-2 in the glomeruli (P0.05, vs control). The expression of MMP-9 did not show any change when comparing the three groups (P>0.05, vs control). STZ-diabetic rats were also associated with an increase in the expression of C-Ⅳ in the glomeruli and the interstitium (Pmatrix accumulation in diabetic nephropathy. Pioglitazone treatment, which can attenuate the decrease of glomerular MMP-2 and the increase of C-Ⅳ degradation, has curative effects on diabetic nephropathy.

Triptolide protects astrocytes from hypoxia/reoxygenation injury

Astrocytes in an in vitro murine astrocyte model of oxygen and glucose deprivation/hypoxia and reoxygenation were treated with different concentrations of triptolide （250, 500, 1 000 ng/mL） in a broader attempt to elucidate the protection and mechanism underlying triptolide treatment on astrocytes exposed to hypoxia/reoxygenation injury. The results showed that the matrix metalloproteinase-9, interleukin-1β, tumor necrosis factor α and interleukin-6 expressions were significantly decreased after triptolide treatment in the astrocytes exposed to hypoxia/ reoxygenation injury, while interleukin-10 expression was upregulated. In addition, the vitality of the injured astrocytes was enhanced, the triptolide＇s effect was apparent at 500 ng/mL. These experimental findings indicate that triptolide treatment could protect astrocytes against hypoxia/ reoxygenation injury through the inhibition of inflammatory response and the reduction of matrix metalloproteinase-9 expression.
