Cloning and Sequencing of a Putative Mitochondrial Intermediate Peptidase gene Fragment from the Edible Fungus Lentinula edodes

A 531 bp fragment of a putative mitochondrial intermediate peptidase (mip) gene from Lentinula edodes was amplified by PCR using a pair of degenerate primers designed according to conserved sequences of mip genes reported in other Basidiomycetes. Alignment analysis revealed that this putative partial mip gene from L. edodes exhibited high homology with corresponding segments of mip genes from the other Basidiomycetes. Since the gene encoding MIP appears to be very closely linked with the A type mating gene of Basidiomycetes, identification of mip may prove a useful tool for locating and cloning the A type mating gene in L. edodes and other edible fungi.

The Winter Mushroom or Enokitake is economically important and widely cultivated in commercial large-scale in East Asia. However, the phylogeny and species delimitation of the winter mushroom genus Flammulina have not been fully elucidated previously. In this study, 81 collections of Flammulina from East Asia, Europe and North America were studied, and their phylogeny and species delimitation were inferred from partial sequences of the internal transcribed spacer regions（ITS）,translation elongation factor 1-α（tef1-α）, the second largest subunit of RNA polymerase II（rpb2） and partial sequence（labled as HD1-A） of homeodomain1（HD1） of mating gene in sublocus ***, genetic structure analyses based on genomic SSR markers and haplotype network analyses based on HD1-A were also employed in delimitation of the closely related taxa. 12 phylogenetic species were recognized, which was largely in consistence with previous ***, our integrated studies indicated that Enokitake is not identical with the true European Winter Mushroom F. velutipes, and, thus, should be treated as a separated species, namely F. filiformis. All cultivated strains of "F. velutipes" in East Asia, including the strains labeled as F. velutipes from Korea and Japan with genome sequence, are in fact F. filiformis. In addition, a new species, F. finlandica, was unanticipatively discovered in Northern Europe. Eventually, morphological descriptions, color photos of fresh basidiomata, line-drawings of microscopic features of the two species were presented.

Wheat stripe rust,caused by Puccinia striiformis ***（Pst）,is a major disease threatening safety of wheat *** selection and deployment of disease-resistant varieties can effectively,economically and environmentally manage wheat stripe ***,the emergence and accumulation of new virulent races of Pst often leads to ineffectiveness of resistance in varieties,resulting in a large scale of epidemics of wheat stripe *** recent years,studies showed that sexual reproduction is a main way of virulence variation of *** sexual reproduction in Basidiomycetes includes two types:homothallism and *** the heterothallism includes the bipolar mating system and the tetrapolar mating *** mating type（MAT） genes of Basidiomycetes are limited to two types,one is the A mating type locus encoding the homeodomain transcription factor（HD gene）,and the other is the B mating type locus encoding the pheromone precursor and pheromone receptor（P/R gene）.At present,the MAT genes and the mating system of Pst has been not well understood,and the role of sexual reproduction on the genetic diversity of natural Pst population is still needed to be further *** this study,a total of 11 HD1 gene sequences were cloned from the natural population of Puccinia striiformis（Ps）,10 of which were from Pst and one from Puccinia striiformis ***（Psh）.The mating system of Pst is confirmed to be *** hybridization of Pst was the main reason for the diversity of HD1 gene combination types in Pst population,which can cause the genetic diversity of Pst natural *** of the epidemic of Pst in 11 provinces of China based on HD1 gene showed that the Pst populations in Gansu and Shaanxi provinces were abundant in HD1 gene combination types and had high genetic diversity,and the Pst populations in Tibet and Xinjiang were two relatively independent populations compared to other regions in *** results provide an insight
