A new phenolic compound, 6-(2-acetyl-3,5-dihydroxybenzyl)-4-hydroxy-3-methyl-2H-pyran-2-one(1), along with other six known phenolic derivatives(2-7), were isolated from the mangrove rhizosphere fungus Penicillium janthinellum HK1-6 cultured in potato dextrose broth medium containing 30 g L^(-1) of natural sea salt. The structure of the new compound(1) was elucidated by comprehensive analysis of spectroscopic data including 1D and 2D NMR spectra. The proposed biosynthetic pathway of compound 1 was also studied in this research. Interestingly, a brominated phenolic derivative, aryl bromide(compound 8), was obtained from this fungal strain cultured in medium containing 30 g L^-1 of NaBr instead of natural sea salt. Compound 8 is proposed as a new natural product and formed through bromination of compound 7 when the fungus was cultured with NaBr. The neuroprotective effect of compound 1 on oxygen-glucose deprivation(OGD)-induced injury was investigated in rat spinal cord astrocytes. MTT assay demonstrated that compound 1 can attenuate OGD-induced cell viability loss in rat spinal cord astrocytes.

Penicildiones A-D(1-4), four new steroids derivatives together with three known compounds including 16α-methylpregna-17α,19-dihydroxy-(9,11)-epoxy-4-ene-3,18-dione-20-acetoxy(5), stachybotrylactone B(6) and stachybotrin(7) were isolated from the soft coral-derived fungus Penicillium sp. SCSIO41201, cultured in the 1% NaCl PDB substrate. Their structures were determined through spectroscopic methods and X-ray crystallography. Biological evaluation results revealed that 6 exhibited significant cytotoxic activity against HL-60, K562, MOLT-4, ACHN, 786-O, and OS-RC-2 cell lines with IC50 values of 5.23, 4.12, 4.31, 23.55, 7.65 and 10.81 μmol·L-1, respectively, while other compounds showed weak or no cytotoxicity at 50 μmol·L-1.

Mechanism of anti-breast cancer compound produced by marine fungus

Terpenoids are the most diverse family of natural products. The artemisinin and paclitaxel are well known antimalarial and anticancer terpenoids drugs,respectively. A marine fungus strain Aspergillus ustus 094102 produced sesterterpeneophiobolins, among which MHO7 is the major product and showed cytotoxicity on different breast cancer cel lines, including the estrogen positive breast cancer cell lines, the triple negative breast cancer cell lines, and the drug resistant breast cancer cell lines.We found that MHO7 can down regulate estrogen receptor α(ERα) both at protein level and transcription level of the estrogen positive breast cancer cell lines,which is different from the known ERα degrader or down regulator. It is evident by detecting binding ability of MHO7 to ERα using fluorescence polarization method,and a RNA-seq analysis which showed that the most affected was estrogen signaling pathway. While for the drug resistant breast cancer cells and the triple negative breast cancer cells which were estrogen receptor negative, we found that MHO7 can inhibit pathways related to apoptosis and cell cycle. We propose that 6 G is a novel multi targets compound, and ERα is one of the target for those cell lines that contain estrogen receptor. More evidence should be obtained on the non ERα containing cells.

The present study was aimed at determining the activity of the metabolite MFS from Fusarium solani FG319 on HMG-CoA reductase in vitro,in order to establish the HMG-CoA reductase inhibiting activity.In vitro evaluation system,marine fungus FG319 metabolite(MFS)was identified as an inhibitor of 3-hydroxy-3-methylglutaryl coenzyme A(HMG-CoA)reductase,the rate-limiting enzyme of the cholesterol biosynthetic pathway.An HMG-CoA reductase inhibiting reaction protocol was constructed in vitro using lovastatin as the positive control.HMG-CoA(2.5 mmol/L),HMG-CoA reductase(2-8 U/mg,656μg/mL),NADPH(10 mmol/L),PBS buffer(50 mmol/L),MFS and lovastatin were added into the wells of a 96-well plate at the final reaction volume of 50μL.The reaction was started by adding HMG-CoA and its initial velocity was measured as the decreasing rate of absorbance at 337 nm(ΔOD337/min).Molecular level investigations were performed for acquiring information on how MFS affects the enzymatic reaction.To investigate an inhibitory effect,test compound MFS dissolved in PBS buffer at various concentrations was added to the reaction mixture.For the HMG-CoA inhibition study,the NADPH concentration was fixed at 1000μmol/L,and HMG-COA was used in the range of125-1000μmol/L.With respect to NADPH,HMG-CoA was fixed at 1000μmol/L,and NADPH was varied from 125-1000μmol/L.The type of inhibition and Ki(the inhibition constant value)of the inhibitor was identified using the Lineweaver-Burk plots.It was concluded that the in vitro inhibiting effect of MFS on HMG-CoA reductase was higher than lovastatin,at concentrations ranging from 10 to 150μg/mL.By using the catalytic domain of human HMG-CoA reductase,MFS was found to inhibit the enzyme activity by competing with HMG-CoA whereas it was uncompetitive towards NADPH.The inhibition constants(K,)with respect to HMG-CoA and NADPH were 0.158±0.019μg/mL and(1.685±0.088)×μg/mL,respectively.Due to these findings,we suggest that MFS would occupy the active site of the enzyme,thereby blocking access of its substrate.The p

Streptococcus agalactiae is a hazardous pathogen that can cause great harm to human and fish.In the present study,the known fungal metabolite helvolic acid(10), seven new helvolic acid derivatives named 6-O-acetyl-aspergitone A(2),6-O-propionyl-aspergitone A(3),1,2-dihydroaspergitone A(4),6-O-acetyl-1,2-dihydroaspergitone A(5),aspergitones B-D(6,7,and9),and two known ones(1 and 8) were isolated from the marine-derived fungus Aspergillus fumigatus HNMF0047 isolated from an unidentified sponge from the beach of Wenchang,Hainan province of China.Compounds 6,7,and 10 showed stronger antibacterial activity than the positive drug tobramycin against Streptococcus agalactiae with MIC values of 16,2,and 8 μg/mL,respectively,which proves that they are potential anti-S.agalactiae drug lead compounds.

To increase the efficiency of dye removal from wastewater using mycelial pellets, a bubble column reactor with a simple structure was designed and efficiently used to remove dyes from solution containing dyes. The mycelial pellets were prepared by marine fungus Aspergillus niger ZJUBE-1. Eight dyes were tested as dye targets for the adsorption capacity of mycelial pellets and good removal results were obtained. Eriochrome black T was selected as a model dye for characterizing the adsorption processes in detail. The measurement results of Zeta potential and FT- IR analysis indicate that the electrostatic attraction may play a key role in the biosorption process. The bubble column reactor was utilized to study the batch dye-removal efficiency of mycelial pellets. A re-culture process between every two batches, which was under non-sterile condition, successfully enhanced the utilization of mycelium bio- mass. The dye removal rate is 96.4% after 12 h in the first batch and then decreases slowly in the following batches. This semi-continuous mode, which consists of commutative processes of dye-removal and re-culture, has some outstanding advantages, such as low power consumption, easy operation, high dye removal rate, and efficient biomass utilization.
