Interaction of cationic cyanine dye with algal alginates: evidence for a polymer bound dye dimer

Under certain conditions algal alginates bind pinacyanol chloride in the form of dimers – this is the conclusion drawn from a uv/vis and circular dichroism (CD) spectroscopic study of aqueous solutions of the dye in a 10-fold molar excess of the polysaccharide from different sources. With its easily detected features the dimer holds promise as a diagnostic tool for alginate conformational analysis. Binding of the strongly blue-shifted (maximum wavelength = 485 nm) dimer is probably mediated by the charged groups and involves guluronate units only. By using the peakFit program, the two overlapping excitonic absorption bands together with the optically inactive band resulted from the interaction of pinacyanol with a specific alginate concentration were separated. The standard Gibbs energy of the interaction was calculated as –27.02 ***–1. The dimers are sensitive against acid and divalent cations.

＜正＞Alginate, polymannuronate, and polyguluronate were partially depolymerized by alginate lyase from Vibrio sp. 510 and Vibro sp. YWA and mild hydrochloric acid hydrolysis. Sixteen oligosaccharide fragments were obtained after fractionation by Gel-filtration and strong-anion exchange high performance liquid chromatography. The sequences of these oligos, which have been confirmed by the NMR in our published papers （Zhang ZQ, et al, 2004;Wang Y., et al, 2005）, were further assigned by electrospray collision-induced dissociation tandem mass spectrometry （ES/CID-MS/MS） in this paper. Besides the B, C, X, Y-type and minor 0.2 A-type glycosidic ions at each glycosidic bond, the difference of the mannuronic and guluronic acid residues, which existed in the middle of sugar chains, were clearly showed in the CID-MS/MS spectra. The unique type ions were easily observed with the appearance of mannuronate residues but not in homoguluronate of saturated oligosaccharide, and with the residues of M and its neighbors, M or G, in the unsaturated hetero-oligosaccharide except the non-reducing end and reducing end. The masses of unique type ions were the same both in saturated and unsaturated oligosaccharide, though the non-reducing end of them had difference of 18μ, and the unique type ions were disappeared after transformation of reducing end from aldehyde to alditol. So it is a kind of X-type ions. The presences of them were related to Mr 44 （CO2） losses from Y-type ions, which could distinguish β-D-mannuronic acid residues from α-L-guIuronic acid residues. ESI/CID-MS/MS is very sensitive and useful for the sequencing analysis of uronic oligosaccharides, and these structural determined oligos would be fixed to the Glycochip for the carbohydrate-protein interaction study.
