magic技术在原发性干燥综合征中的应用

目的探讨一站式弛豫定量（MAGi C）在原发性干燥综合征（pSS）中应用价值。方法选择33例pSS女性患者（pSS组），年龄29～81岁，平均年龄53.85岁；病程1～14个月，平均病程6.5个月。同期选择25例女性健康志愿者（健康组），年龄26～84岁，平均年龄50.19岁，无pSS相关病程。对所有受试者双侧腮腺进行常规序列和MAGi C序列扫描，通过后处理获得腮腺组织的MAGi C定量图谱并测量所有受试者腮腺的T1均值、T2均值、质子密度（PD）均值。利用组内相关系数（ICC）评估腮腺组织T1均值、T2均值、PD均值组内和组间测量的一致性。比较pSS组与健康组之间各均值的差异，并绘制受试者工作特性（ROC）曲线评估各均值对pSS的诊断效能，应用DeLong检验比较各均值诊断效能的差异。结果观察者组内和组间测量均值的一致性均较好（r> 0.9）。pSS组T1均值和T2均值均高于健康组[（711.17±90.30） ms vs （615.58±60.66） ms、（89.92±10.69） ms vs （77.88±6.69） ms。P 0.05），且高于PD均值（P <0.05）。T1均值联合T2均值诊断pSS的AUC值、灵敏度、特异度、95%CI、约登指数分别为0.935、100.0%、78.1%、0.837～0.983、0.781，均明显高于T1均值、T2均值、PD均值（P均<0.05）。结论 MAGi C技术可应用于鉴别pSS，且腮腺组织的T1均值、T2均值皆有一定的诊断效能，二者联合可以为pSS的MRI影像诊断提供更高的诊断价值

Lignin metabolism plays a pivotal role in plant defense against pathogens and is always positively correlated as a response to pathogen infection. Thus, understanding resistance genes against plant pathogens depends on a genetic analysis of the lignin response. This study used eight Upland cotton lines to construct a multi-parent advanced generation intercross(magic) population(n=280), which exhibited peculiar characteristics from the convergence of various alleles coding for advantageous traits. In order to measure the lignin response to Verticillium wilt(LRVW), the artificial disease nursery(ADN) and rotation nursery(RN) were prepared for magic population planting in four environments. The stem lignin contents were collected, and the LRVW was measured with the lignin value of ADN/RN in each environment, which showed significant variations. We employed 9 323 high-quality single-nucleotide polymorphism(SNP) markers obtained from the Cotton-SNP63K array for genotyping the magic population. The SNPs were distributed through the whole genome with 4.78 SNP/Mb density, ranging from 1.14(ChrA06) to 10.08(ChrD08). In addition, a genome-wide association study was performed using a Mixed Linear Model(MLM) for LRVW. Three stable quantitative trait loci(QTLs), qLRVW-A04, qLRVW-A10, and qLRVW-D05, were identified in more than two environments. Two key candidate genes, Ghi_D05G01046 and Ghi_D05G01221, were selected within the QTLs through the combination of variations in the coding sequence, induced expression patterns, and function annotations. Both genes presented nonsynonymous mutations in coding regions and were strongly induced by Verticillium dahliae. Ghi_D05G01046 encodes a leucine-rich extensin(LRx) protein involved in Arabidopsis cell wall biosynthesis and organization. Ghi_D05G01221 encodes a transcriptional co-repressor novel interactor of novel interactor of jasmonic acid ZIM-domain(JAZ–NINJA), which functions in the jasmonic acid(JA) signaling pathway. After being pumped from the 5s_(1/2) ground state to the 6p_(1/2) state,the population inversion between 6s_(1/2) and 5p_(1/2,3/2) can be established for a rubidium four-level active optical clock.We calculate the ac Stark shift due to lattice trapping laser which dominates the frequency shift of clock transition in a lattice trapped rubidium four-level active optical clock.Several magic wavelengths are found,which can form desired optical lattice trapping potential.By choosing a proper intensity and linewidth of the trapping laser,the fractional frequency uncertainty of clock transition due to the ac Stark shift of the trapping laser,is estimated to be below 10^(−18).

Similar to most of the other alkaline earth elements,barium atoms can be candidates for optical clocks,thus the magic wavelength for an optical lattice is important for the clock transition.We calculate the magic wavelength of a possible clock transition between 6s21S0 and 6s5d3D2 states of barium atoms.Our theoretical result shows that there are three magic wavelengths 615.9 nm,641.2 nm and 678.8 nm for a linearly polarized optical lattice laser for barium.

The wild Solanum pimpinellifolium(SP)and the weedy S.lycopersicum var.cerasiforme(SLC)are largely unexploited genetic reservoirs easily accessible to breeders,as they are fully cross-compatible with cultivated tomato(S.lycopersicum var.lycopersicum).We performed a comprehensive morphological and genomic characterization of four wild SP and four weedy SLC accessions,selected to maximize the range of variation of both taxa.These eight accessions are the founders of the first tomato interspecific multi-parent advanced generation inter-cross(magic)population.The morphoagronomic characterization was carried out with 39 descriptors to assess plant,inflorescence,fruit and agronomic traits,revealing the broad range of diversity captured.Part of the morphological variation observed in SP was likely associated to the adaptation of the accessions to different environments,while in the case of SLC to both human activity and adaptation to the environment.Whole-genome resequencing of the eight accessions revealed over 12 million variants,ranging from 1.2 to 1.9 million variants in SLC and from 3.1 to 4.8 million in SP,being 46.3%of them(4,897,803)private variants.The genetic principal component analysis also confirmed the high diversity of SP and the complex evolutionary history of SLC.This was also reflected in the analysis of the potential footprint of common ancestors or old introgressions identified within and between the two taxa.The functional characterization of the variants revealed a significative enrichment of GO terms related to changes in cell walls that would have been negatively selected during domestication and breeding.The comprehensive morphoagronomic and genetic characterization of these accessions will be of great relevance for the genetic analysis of the first interspecific magic population of tomato and provides valuable knowledge and tools to the tomato community for genetic and genomic studies and for breeding purposes.
