Hybrid Dextran-gadolinium Nano-suitcases as High-relaxivity mri contrast agents

Dextran-poly（glycidyl methacrylate）（Dex-PGMA） nano-suitcases were synthesized efficiently via a graft copolymerization induced self-assembly （GISA） approach. On this basis, the Dex-PGMA nano-suitcases were modified with hydrazide, and the attachment of multiple chelated Gd（III） ions to the interior of the nano-suitcases affords nanoscale mri contrast agents with high relaxivity values. The highly fenestrated dextran shell of the nano-suitcases assures water exchange which readily occurs between the surrounding environment and the Gd（III） ions encapsulated within the hybrid nano-suitcases. The complexation between the hydrophilic hydrazide interior of the nano-suitcases and Gd（III） ions results in an impressive Gd payload at 22.6 wt% in the hybrid nano-suitcases. The longitudinal relaxivity （rl） of the hybrid nano-suitcases is reported as 44.4 L/（mmol-s）, which is 9-14 folds of that of commercial Gd-DTPA agents. In vivo mri studies demonstrate that the hybrid nano-suitcases accumulated in the lymph node of the rat due to their nanoscale dimensions and displayed strong signals in vivo. The results indicated that the hybrid nano-suitcases provide a promising platform for the diagnosis of lymph node related diseases.

Stem cells are capable of replication and differentiation so that they are widely studied for its potential application for tissue engineering and *** stem cells are delivered into human body,it is hard to detect its existence and trace its migration route between different *** and superparamagnetic substances are used for labeling stem cells that could be visualized under magnetic resonance imaging(mri).However,the efficacy,sensitivity and feasibility between paramagnetic and superparamagnetic substances labeling are not fully investigated. We investigated paramagnetic and superparamagnetic substance labeling by choosing clinically widely used gadolinium chelates as choice for paramagnetic contrast agent and iron oxide nanoparticles as candidate for superparamagnetic substance because the toxicity and adverse effects to the cells are previously widely *** gadolinium chelates and iron oxide nanoparticles are labeled into human mesenchymal stem cell line at different concentration and the optimized concentration was determined by maintaining cell viability the same as cells without *** optimized concentration,labeled cells at numbers from 1000 to 2×10 are placed into test tubes for mri *** minimal cell numbers that could be detected by clinical mri was *** cells are further injected into bilateral flank of living *** clinical 1.5 Tesla mri scanning,the images are evaluated for stem cell detecting ability. We found there is 8 fold sensitivity increases in cells labeled with iron oxide nanoparticles,which has detecting limit of 12500 cells whereas only 50000 or more cells could be detected in gadolinium chelates labeled *** living animal study,although iron oxide nanoparticle labeled stem cells could be detected 8 times more sensitive,the magnetic susceptibility artifact interrupted the visualization of adjacent organ *** conclude that labeling stem cells with super

The uniform and extremely small-sized FeO nanoparticles decorated self-made graphene composites(G-FeO) have been successfully prepared via a facile coprecipitation method[1]. The amount of loading of FeO on graphene was estimated as 38.69 wt% with high stability, excellent supraparamagnetism, as well as good biocompatibility for G-FeO. The hydrophobic nanocomposites can be easily transformed to water-dispersible and stable G-FeO-NH nanocomposites through one-step solution-processing which can achieve to capping with the formational polyaniline(PAN) and bind or immobilize metal nanoparticles on the surface of graphene sheets[2]. The result indicated that the hydrophilic group-containing composite was loaded with FeO on monolayer graphene and the loading capacity was as high as 47.95 wt% and having the integrated properties of superior electrical conductivity and superparamagnetism,oxidation resistance and thermal stability, water solubility,magnetic sensitivity and cell compatibility, which was demonstrated that G-FeO-NH is a promising candidate for magnetic resonance imaging contrast agents[3, 4].
