The technique of m.crochip gel electrophoresis (m.GE) was used to analyze the polym.rase chain reaction (PCR) products of m. tuberculosis Genom. stained by ethidium.brom.de. The electrophoretic process was com.leted within 3-4 m.n and the results show that the technique of m.crochip electrophoresis is a high speed and high sensitivity analyzing m.thod.

A new m.ltilocus Sequence Analysis Schem. for m.cobacterium.tuberculosis

Objective tuberculosis rem.ins one of the m.st serious infectious diseases in the world. In this study, a schem. of m.cobacterium.tuberculosis （m. tuberculosis） m.ltilocus sequence analysis （m.SA） was established for the phylogenetic and epidem.ology analysis. m.thods To establish the schem. of m. tuberculosis m.SA, the genom. of H37Rv, CCDC5079 and CCDC5180 were com.ared, and som. variable genes were chosen to be the m.SA typing schem.. 44 m. tuberculosis clinical isolates were typed by m.SA, IS6110-RFLP, and soligotyping, to evaluate the m.SA m.thods. Results After com.arison of the genom., seven high discrim.nation gene loci （recX, rpsL, rm.C, rpm.1, m.rA, gcvH, ideR） were chosen to be the m.SA typing schem. finally. 11 variable SNP sites of those seven genes were found am.ng the 44 m. tuberculosis isolate strains and 11 sequence types （STs） were identified. Based on the Hunter-Gaston Index （HGI）, m.SA typing was not as good for discrim.nation at the strain level as IS6110-RFLP, but the HGI was m.ch better than that of spoligotyping. In addition, the m.GA analysis result of m.SA data was sim.lar to spoligotyping/PGG lineage, showing a strong phylogenetic signal in the m.dern strains of m. tuberculosis. The m.SA data analysis by eBURST revealed that 4 sequence types （ST） cam. into a m.in cluster, showing the m.jor clonal com.lexes in those 44 strains. Conclusion m.SA genotyping not only can be used for m.lecular typing, but also is an ideal m.thod for the phylogenetic analysis for m. tuberculosis.

Screening and Assessing 11 m.cobacterium.tuberculosis Proteins as Potential Serodiagnostical m.rkers for Discrim.nating TB Patients from.BCG Vaccinees

Purified protein derivative （PPD） skin tests often yield poor specificity, so that to develop new serological antigens for distinguishing between m.cobacterium.tuberculosis infection and Bacille Calm.tte-Guerin （BCG） vaccination is a priority, especially for developing countries like China. We predicted the antigenicity for selected open reading fram.s （ORFs） based on the genom. sequences of m. tuberculosis H37Rv and m. boris BCG, as well as their functions and differences of expression under different stim.lus. The candidate ORFs were cloned from.H37Rv sequences and expressed as recom.inant proteins in Escherichia coli. We studied the serodiagnostic potential of 11 purified recom.inants by using enzym.-linked im.unosorbent assay （ELISA） and involving a cohort com.osed of 58 TB patients （34 m.les and 24 fem.les）, 8 healthy volunteers and 50 PPD-negative individuals before and after BCG vaccination. For all the 11 antigens, the m.dian OD values for the sera from.TB patients were statistically significantly higher than those for PPD-negative individuals before or after BCG vaccination （P〈0.01）. They had at least 92% specificity in healthy controls and six seroantigens （Rv0251c, Rv1973, Rv2376c, Rv2537c, Rv2785c and Rv3873A） were never reported with seroantigenicities previously. Thus the approach com.ining com.arative genom.cs, bioinform.tics and ELISA techniques can be em.loyed to identify new seroantigens distinguishing m. tuberculosis infection from.BCG vaccination.

Tuberculous Spondylitis: Clinical Features of 36 Patients

We report 36 cases of spinal tuberculosis who were evaluated at the Internal m.dicine Departm.nt of Istanbul University between January 1990 and m.rch 2016. Twelve cases were accom.anied by active pulm.nary tuberculosis while ten patients had a previous history of tuberculosis. Eight patients had diabetes m.llitus;six patients were on chronic steroid treatm.nt with a m.an dose of 24 m./day while two patients used azathioprine and m.thotrexate. The dom.nant sym.tom.was back pain that was present in 64% of the subjects followed by low grade fever (42%), and m.laise (38%). Lytic and destructive lesions were noted in various vertebrae in all of the cases while four patients had spinal com.ression, and two patients had iliopsoas abscess. The prelim.nary diagnosis was m.eloproliferative disease with vertebral m.tastasis in eight patients. Tuberculin test was over 15 m. in 20 patients (58.8%) while ERS and Creactive protein were highly elevated in 78%, and 84% of the patients. Diagnosis of tuberculosis was confirm.d by culture of aspirated m.terial from.intervertebral disc space, collection under CT guidance, sputum.or bronchial lavage, and by retrospective anti-tuberculous treatm.nt response. m.I revealed com.atible findings with spinal tuberculosis in 30 (80%) patients. In endem.c countries, tuberculous spondilitis should be considered in im.unosuppressed patients with back pain. Current or past tuberculosis infection is not a reliable indicator for Pott’s disease in these patients. Laboratory findings were not usually useful to support the diagnosis. The low sensitivity of the tuberculin test m.y lead to an erronous diagnosis. Sensitivity of vertebral radiography and CT was low. m.I was the m.st useful radiologic investigation for the diagnosis of spinal tuberculosis. tuberculosis of the spine is a diagnostic im.asse notably in im.unodeficient patients for the clinician.
