Our previous study verified the protective effects of lycium barbarum polysaccharides(LBP)on retinal neurons and blood vessels in acute ocular hypertension(AOH)*** investigate the effect of LBP on the reactivity of retinal glial cells,an AOH mouse model was established in one eye by maintaining ocular hypertension of 90 mm Hg for 60 *** LBP solution(1 mg/kg)or phosphate-buffered saline was administrated to the mice by gavage daily,starting 7 days before the AOH insult and continuing until the mice were sacrificed for specimen collection on day 4 *** AOH insult,increased numbers of astrocytes and microglia were observed,together with decreased expression of the following glial cell biomarkers in the retinal ganglion cells of AOH mice:glial fibrillary acidic protein,glutamine synthetase,aquaporin-4,S-100 proteins,ionized calcium-binding adaptor molecule 1,amyloid precursor protein and receptor of advanced glycosylation *** intervention with LBP,the above changes were significantly ***,morphological remodeling of blood vessel-associated retinal astrocytes,marked by glial fibrillary acidic protein,was also *** results,taken together,suggest that LBP regulated the production of amyloid-βand expression of receptor of advanced glycosylation end-products,as well as mediating the activity of retinal glial cells,which may lead to the promotion of better maintenance of the blood-retinal barrier and improved neuronal survival in AOH *** study was approved by the Committee for the Use of Live Animals in Teaching and Research(approval ***-#1664-08).

Exploring the effect of in vitro digestion on the phenolics and antioxidant activity of lycium barbarum fruit extract

To assess the effect of the digestive system on the phenolics and antioxidant capacity of lycium barbarum fruit,its extract was digested and analyzed in *** results revealed that the phenolics/flavonoids contents and antioxidant capacity in the pH-treated group and digested group generally decreased by 4.39%-21.22%in the oral and intestinal digestions but increased by 1.26%-16.14%in the gastric *** phenolic content and antioxidant capacity were highly correlated in digested groups(r>0.899,p<0.01).Five main phenolics including rutin,p-coumaric acid and its hexoside derivatives were identified and quantified,and these phenolics displayed higher stability during the digestion,implying that they may be the potential functional phenolics of *** fruit.

THE EFFECT OF ACTIVE COMPONENTS OF lycium barbarum AND GARLIC (LB-GO) ON THE SYNTHESIS OF DNA AND ULTRASTRUCTURE OF U_(14) CERVIX CANCER CELLS IN MICE

Mice, inoculated with U_(14) ascitic type cervix cancer cells, were administered with effective components of lycium barbarum (LB) and garlic (GO).On the 4th day after ***, general condition of the animals were found *** of ascitic fluid revealed damage of the cancer cells, blanching of fluorescence staining of DNA and RNA, and the cancer cells besieged by large numbers of macrophages and leucocytes. Flow cell metric (FCM) analysis found: accumulation of cells of G_1 stage. Ultrastructure study disclosed: swelling of mitochondria in cytoplasm and damage of mitochondrial crests even with cavity formation, enlargement and degranulation of rough ER. It seemed the effect of LBGO was affirmable. It was postulated that macrophages,being activated by LB, came in close contact with the cancer cells giving rise to carcinolysis. In addition to the direct but transient killing effect of GO, the anti-cancer results could be greatly enhanced.

Storage Stability of Imidacloprid Residues in Different Matrixes of lycium barbarum

The storage stability and degradation trends of imidacloprid residues in different matrix of lycium barbarum were investigated by directly adding 0.1 mg/kg imidacloprid, frozen storage and phase detection. The results showed that under frozen condition, the storage stability of imidacloprid residues remained good for half a year in dry fruit, and a year in fresh fruit of L. barbarum . There was no significant change of imidacloprid residues between different processing of dry fruit and fresh fruit, and sample processing methods （crushing or homogenizing） had no significant effect on the degradation trends of imidacloprid. Therefore, the samples could be processed into crushed samples or homogenized samples for storage as soon as possible according to the needs, and the dry fruit should be detected within half a year, while the fresh fruit should be detected within a year.

Three new dimers of phenolic amides,named lyciumamides A（3）,B（4）and C（5）,along with two known monomers,N-E-coumaroyl tyramine（1）and N-E-feruloyl tyramine（2）,were isolated from the fruits of lycium barbarum for the first time,with help of the activity-guided *** amides were also firstly found in the genus *** structures were determined using spectral analysis and comparing with literature *** 1–5 were evaluated for their antioxidant activities in scavenging DPPH free radical and inhibiting lipid peroxidation in rat liver microsomes induced by ascorbate/Fe2+,cumine hydroperoxide or CCl4/reduced form of nicotinamide-adenine dinucleotide phosphate,and the results showed that all of them exhibited strong activities while compounds 1 and 2 were more potent than the reference *** data supported that these phenolic amides played an important role on the natural antioxidant properties of the fruits of ***,similar to other phenolic constituents such as flavonoids and phenolic acids of the *** conclusion,the results complemented the current knowledge of the antioxidative composition of the fruits of *** and confirmed that this agricultural product contained phenolic amide compounds,which might offer interesting potential applications in the food,cosmetic,and pharmaceutical industries.

Investigation on Prescription Screening of a Polyherbal Preparation Based on Splenocyte Proliferation Activity and its Preparation Method

Objective: The present study aims to optimize the extraction conditions of polysaccharides and flavonoids from a polyherbal preparation consisting of three kinds of Chinese medicinal herbs, Codonopsis pilosula, Crataegus pinnatifida, and lycium barbarum, and evaluation of its immunomodulatory activity in immunosuppressed mice. Materials and Methods: An orthogonal design(L_9 [3]~4) was constructed to achieve the optimal extraction conditions. The immunomodulatory action of the polyherbal preparation was studied at three doses of 10, 20, and 40 mL/kg/day orally by measuring splenocyte proliferation in mice model of cyclophosphamide-induced immunosuppression. Results: The chosen parameters, including the ratio of solvent-to-raw material, duration of extraction, and extraction times, were the fundamental variables that influenced the extract yields. The highest yield of total polysaccharides content was 54.3 mg/mL when the ratio of solvent-to-raw material,duration of extraction, and number of extractions were 12:1, 1.5 h, and 3, respectively. The maximum extraction yield of the flavonoids was 3.5 mg/mL when the ratio of solvent-to-raw material was 12:1, the extraction time was 2 h, and the number of extractions was 3. The prescription screening showed that the impact of the polyherbal preparation on the splenocyte proliferation capacity was more pronounced than its disassembled components. Oral administration of the polyherbal preparation could significantly increase the concanavalin A-stimulated mouse spleen cells proliferation in a dose-dependent manner. Conclusion: These findings suggested that the polyherbal preparation possesses potential for augmenting the immune activity due to the polysaccharide and flavonoid content in these herbal medicines.
