Acidosis,regardless of hypoxia involvement,is recognized as a chronic and harsh tumor microenvironment(TME)that educates malignant cells to thrive and *** overwhelming evidence supports an acidic environment as a driver or ubiquitous hallmark of cancer progression,the unrevealed core mechanisms underlying the direct effect of acidification on tumorigenesis have hindered the discovery of novel therapeutic targets and clinical ***,chemical-induced and transgenic mouse models for colon,liver and lung cancer were established,***-7 and TGF-β2 expressions were examined in clinical tissues(n=184).RNA-seq,miRNA-seq,proteomics,biosynthesis analyses and functional studies were performed to validate the mechanisms involved in the acidic TME-induced lung cancer *** data show that lung cancer is sensitive to the increased acidification of TME,and acidic TME-induced lung cancer metastasis via inhibition of miR-7-5 ***-β2 is a direct target of miR-7-5 *** reduced expression of miR-7-5 p subsequently increases the expression of TGF-β2 which enhances the metastatic potential of the lung ***,overexpression of miR-7-5 p reduces the acidic p H-enhanced lung cancer ***,the human lung tumor samples also show a reduced miR-7-5 p expression but an elevated level of activated TGF-β2;the expressions of both miR-7-5 p and TGF-β2 are correlated with patients’*** are the first to identify the role of the miR-7/TGF-β2 axis in acidic p H-enhanced lung cancer *** study not only delineates how acidification directly affects tumorigenesis,but also suggests miR-7 is a novel reliable biomarker for acidic TME and a novel therapeutic target for non-small cell lung cancer(NSCLC)*** study opens an avenue to explore the p H-sensitive subcellular components as novel therapeutic targets for cancer treatment.

Studies on the lung cancer genome are indispensable for developing a cure for lung ***-genome resequencing,genome-wide association studies,and transcriptome sequencing have greatly improved our understanding of the cancer ***,dysregulation of longrange chromatin interactions in lung cancer remains poorly *** better understand the three-dimensional(3D)genomic interaction features of the lung cancer genome,we used the A549 cell line as a model system and generated high-resolution chromatin interactions associated with RNA polymerase II(RNAPII),CCCTC-binding factor(CTCF),enhancer of zeste homolog 2(EZH2),and histone 3 lysine 27 trimethylation(H3K27me3)using long-read chromatin interaction analysis by paired-end tag sequencing(ChIA-PET).Analysis showed that EZH2/H3K27me3-mediated interactions further repressed target genes,either through loops or domains,and their distributions along the genome were distinct from and complementary to those associated with ***-related genes were highly enriched with chromatin interactions,and chromatin interactions specific to the A549 cell line were associated with oncogenes and tumor suppressor genes,such as additional repressive interactions on FOXO4 and promoter–promoter interactions between NF1 and *** of an anchor associated with chromatin interactions reversed the dysregulation of cancer-related genes,suggesting that chromatin interactions are essential for proper expression of lung cancer-related *** findings demonstrate the 3D landscape and gene regulatory relationships of the lung cancer genome.

Small non-coding RNAs are potential diagnostic biomarkers for lung cancer. Mitochondria-derived small RNA (mtRNA) is a novel regulatory small non-coding RNA that only recently has been identified and cataloged. Currently, there are no reports of studies of mtRNA in human lung cancer. Currently, normalization methods are unstable, and they often fail to identify differentially expressed small non-coding RNAs (sncRNAs). In order to identify reliable biomarkers for lung cancer screening, we used a ratio-based method using mtRNAs newly discovered in human peripheral blood mononuclear cells. In the discovery cohort (AUC = 0.981) and independent validation cohort (AUC = 0.916) the prediction model of eight mtRNA ratios distinguished lung cancer patients from controls. The prediction model will provide reliable biomarkers that will allow blood-based screening to become more feasible and will help make lung cancer diagnosis more accurate in clinical practice.

Brain metastasis(BM)is the leading cause of mortality in lung cancer *** process of BM(from initial primary tumor development,migration and intravasation,dissemination and survival in the bloodstream,extravasation,to colonization and growth to metastases)is a complex process for which few tumor cells complete the entire *** research on BM of lung cancer has recently stressed the essential role of tumor microenvironment(TME)in assisting tumor cells in the completion of each BM *** review summarizes recent studies regarding the effects of TME on tumor cells in the entire process of BM derived from lung *** identification of vulnerable targets in the TME and their prospects to provide novel therapeutic opportunities are also discussed.

FRMD6,a member of the 4.1 ezrin-radixin-moesin domain-containing protein family,has been reported to inhibit tumor progression in multiple ***,we demonstrate the involvement of FRMD6 in lung cancer *** find that FRMD6 is overexpressed in lung cancer tissues relative to in normal lung *** addition,the enhanced expression of FRMD6 is associated with poor outcomes in patients with lung squamous cell carcinoma(n=75,P=0.0054)and lung adenocarcinoma(n=94,P=0.0330).Cell migration and proliferation in vitro and tumor formation in vivo are promoted by FRMD6 but are suppressed by the depletion of ***,FRMD6 interacts and colocalizes with mTOR and S6K,which are the key molecules of the mTOR signaling ***6 markedly enhances the interaction between mTOR and S6K,subsequently increasing the levels of endogenous pS6K and downstream pS6 in lung cancer ***,knocking out FRMD6 inhibits the activation of the mTOR signaling pathway in Frmd6^(−/−)gene KO MEFs and ***,our results show that FRMD6 contributes to lung cancer progression by activating the mTOR signaling pathway.

As one of the most malignant tumors worldwide,lung cancer,fueled by metastasis,has shown rising mortality ***,effective clinical strategies aimed at preventing metastasis are lacking owing to its dynamic multi-step,complicated,and progressive *** has shown promise in treating cancer metastasis by reversing the immunosuppressive network of the tumor ***,drug resistance inevitably develops due to inadequate delivery of immunostimulants and an uncontrolled immune ***,adverse effects occur,such as autoimmunity,from the non-specific immune activation and non-specific inflammation in off-target *** that improve drug solubility,permeability,stability,bioavailability,as well as sustained,controlled,and targeted delivery can effectively overcome drug resistance and enhance the therapeutic effect while reducing adverse *** particular,nanomedicine-based immunotherapy can be utilized to target tumor metastasis,presenting a promising therapeutic strategy for lung *** strategies that boost the immunotherapy effect are classified based on the metastatic cascade related to the tumor immune microenvironment;the breaking away of primary tumors,circulating tumor cell dissemination,and premetastatic niche formation cause distant secondary site *** this review,we focus on the opportunities and challenges of integrating immunotherapy with nanoparticle formulation to establish nanotechnology-based immunotherapy by modulating the tumor microenvironment for preclinical and clinical applications in the management of patients with metastatic lung *** also discuss prospects for the emerging field and the clinical translation potential of these techniques.

N6 -methyladenosine (m6 A) modification is found the most prevalent and abundant post-transcriptional mRNA modification in eukaryotic cells. It regulates almost all stages of RNA life cycle including splicing, translocation, stability, decay and translation. As a dynamic and reversible process, m6 A modification is catalyzed by the RNA methyltransferases (‘writers’), removed by the demethylases (‘erasers’), and interacts with m6 A-binding proteins (‘readers’). Recent studies have revealed that these m6 A modification regulators are frequently expressed aberrantly in various types of cancer, and involved in cell proliferation, differentiation, metabolism, particularly, in tumorigenesis and tumor progression through diverse mechanisms. In this review, the m6 A modification process and its regulatory functions in lung cancer are summarized. Furthermore, the research progress in the inhibitor development of m6 A modification, and the potential of targeting m6 A modifying proteins for clinical application are discussed.
