The anti-tubercular activity of Melia azedarach L. and Lobelia chinensis lour. and their potential as effective anti-Mycobacterium tuberculosis candidate agents

Objective: To evaluate the anti-mycobacterial activity of Melia azedarach L.(M. azedarach) and Lobelia chinensis lour.(L. chinensis) extracts against the growth of Mycobacterium tuberculosis(M. tuberculosis).Methods: The anti-M. tuberculosis activity of M. azedarach and L. chinensis extracts were evaluated using different indicator methods such as resazurin microtiter assay(REMA) and mycobacteria growth indicator tube(MGIT) 960 system assay. The M. tuberculosis was incubated with various concentrations(50–800 mg/m L) of the extracts for 5 days in the REMA, and for 4 weeks in MGIT 960 system ***: M. azedarach and L. chinensis extracts showed their anti-M. tuberculosis activity by strongly inhibiting the growth of M. tuberculosis in a concentration-dependent manner in the REMA and the MGIT 960 system assay. Particularly, the methanol extract of M. azedarach and n-hexane extract of L. chinensis consistently exhibited their effects by effectively inhibiting the growth of M. tuberculosis in MGIT 960 system for 4 weeks with a single-treatment, indicating higher anti-M. tuberculosis activity than other extracts,and their minimum inhibitory concentrations were measured as 400 mg/m L and 800 mg/m L, ***: These results demonstrate that M. azedarach and L. chinensis extracts not only have unique anti-M. tuberculosis activity, but also induce the selective antiM. tuberculosis effects by consistently inhibiting or blocking the growth of M. tuberculosis through a new pharmacological action. Therefore, this study suggests the potential of them as effective candidate agents of next-generation for developing a new anti-tuberculosis drug, as well as the advantage for utilizing traditional medicinal plants as one of effective strategies against tuberculosis.

Anti-inflammatory effects of ethanol fraction from Disporum cantoniense （lour.） Merr. in LPS-stimulated RAW264. 7 cells

Aim To evaluate the anti-inflammatory effects of ethanol fraction prepared from Disporum cantoniense （lour.） Merr. 70% ethanol extract with a cellular model of LPS-stimulated RAW264.7 cell. Methods RAW264.7 cells were treated with different concentrations of ethanol fraction （25,50 and 100 g · L^-1 ） and stimu- the conditioned media was collected and analyzed. The quantity of ni- lated with LPS （10 μg· L^-1） for 24 hours, tric oxide （NO） was assayed by Griess reagent. The production of inflammatory mediators was determined by en- zyme-linked immunosorbent assay （ELISA）, such as prostaglandin E2 （PGE2）, tumor necrosis factor ot （TNF-ot） interleukin- 1 β （IL-1 β） and interleukin 6 （IL-6） in cell supernatant. The concentrations of inflammatory medi- 9 ators were calculated according to the standard curves generated by each of the recombinant cytokines provided with LPS can induce RAW264.7 cells to promote the pro- the ELISA kits. Results Compared with the control group, TNF-α, IL-1β and IL-6. Compared with the duction of inflammatory mediators （P 〈 0.01 ） , including NO, PGE2, model group, ethanol fraction significantly suppressed LPS induced release of inflammatory mediators such as nitric NO, PGE2, TNF-α, IL-1β and IL-6 in a good dose dependent manner （P 〈 0.05, P 〈 0.01 ）. Conclusions Eth- anol fraction could significantly inhibit the production of LPS-induced inflammatory response in RAW264.7 cells, and its anti-inflammatory effect may be related to reduce the production of inflammatory mediators NO, PGE2, TNF-α, IL-1 β and IL-6. These results demonstrate that the ethanol fraction is the bioactive component of Disporum can- toniense （lour.） Merr. , and the ethanol fraction will be further developed as a herbal remedy for preventive and/ or curative purposes in various inflammatory diseases.

[Objectives]This study aimed to optimize the extraction process of longan(Dimocarpus longan lour.)leaves by multi-indicator comprehensive evaluation method.[Methods]The contents of gallic acid ethyl ester,astragalin,quercetin,luteolin and kaempferol in longan leaves were determined simultaneously by *** the multi-indicator comprehensive evaluation method,the methanol concentration,solid to liquid ratio and extraction time for extraction of longan leaves were optimized by orthogonal test.[Results]The optimal extraction process of longan leaves are as follows:methanol concentration of 100%,solid to liquid ratio of 1∶5,and extraction time of 50 min.[Conclusions]The optimized process is simple and feasible,and it can be used to determine the contents of different ingredients in longan leaves.

Phytochemical and in vitro biological investigations of methanolic extracts of Enhydra fluctuans lour

Objective:To study the phytochemical and biological properties(antioxidant,anthelmintic and thrombolytic)of methanolic extracts of Enhydra fluctuant lour.,a plant belonging to the Asteraceae ***:The phytochemical evaluation was carried out by qualitative *** vitro antioxidant activity of extract was studied using free radical scavenging assay,ability of reduction,total phenol and total flavonoid contents determination *** anthelmintic activity was determined using paralysis and death time of Pheretima posthuma(earthworm)and thrombolytic activity by clot disruption ***:The phytochemical evaluation showed significant presence of flavonoids,triterpenes,carbohydrate,reducing sugars,saponins,phenols,diterpenes,protein and *** antioxidant activity was found significant[IC_(50)(135.20±036)μg/mL]as compared to ascorbic acid[(130.00±0.76)μg/mL].The reducing power was increased with *** phenol and total flavonoid contents were(153.08±0.38)mg/mL and(172.04±0.56)mg/mL *** paralysis and death time of earthworms for different concentrations of extract were determined and compared with *** results showed that 10 mg/mL of the crude extract had similar effect with ***,the crude extract showed a concentration depended relationship with its anthelmintic *** clot lysis activity of crude extract was compared to the standard streptokinase's clot lysis(40.13%)activity and found significant(31%).Conclusions:The study proves that the crude methanolic extract of Enhydra fluctuant lour.has significant antioxidant,anthelmintic and thrombolytic activity containing wide range of phytochemicals.

中国柽柳(Tamarix chinensis lour.)为落叶小乔木或灌木,野生于山东、辽宁、河北、河南、江苏(北部)等省,喜生于河流冲积平原、海滨、滩头、潮湿盐碱地和沙荒地,具有极强的耐盐碱能力和观赏价值。由于柽柳独特的抗逆性及盐碱地利用的不可替代性,对其研究逐步深入。2006年在种质资源调查、汇集中,在野外发现多个中国柽柳的变异单株。之后繁殖建立无性系。通过大田扦插、容器育苗多种繁育技术对中国柽柳进行扩繁。

<i>Ralstonia solanacearum</i>Induction Causes Biochemical and Oxidative Stress Isozyme Variations in Mangroves without Wilt

We evaluated the effects of Ralstonia solanacearum (Rs) induced biotic stress in three mangroves, viz., Avicennia officinalis, Derris trifoliata and Excoecaria agallocha. These plants were grown in pots as well as hydroponic systems with sufficient controls, and about 8 × 104 colony forming units of Rs suspension was injected into the healthy test plants (saplings). The plants were subjected to biochemical and isozyme analyses. Upon induction of Rs stress, highly significant (p 0.01) biochemical changes (%) were noticed in respect to controls: carbohydrate content was generally high (24-36) in all plants;hydroponic mangroves showed higher starch content, mangroves under hydroponic system showed increased reducing sugars (29-46), almost all mangroves showed increased protein content;phenolics showed a swing of decrease or increase between plants grown in pot and hydroponic systems;and all plants in general showed higher proline content. Regarding oxidative stress isozymes (OSE) and superoxide dismutase (EC1.***.1.1), mangroves showed 1 or 2 additional isozymes with comparable relative mobility;similarly 1 or 2 additional peroxidase (EC1.***.1.7) isozymes were found in mangroves grown under hydroponic system. Briefly, Rs induced biotic stress did not cause any wilt symptom in all the 3 mangroves tested, but their normal biochemical and OSE patterns, especially of those grown as hydroponics were elicited to significantly higher levels.

Preliminary Experimental Study on Anti-inflammatory Effect of Extract from Excoecaria cochinchinensis *** a National Medicine

[Objectives]To study the anti-inflammatory effects of water extract and ethanol extract from Excoecaria cochinchinensis lour.[Methods]The acute and chronic inflammation models of mouse paw edema induced by cotton ball granuloma and carrageenan were established respectively,and the anti-inflammatory effects of water and ethanol extract from *** *** acute and chronic inflammation in mice were *** experimental mice were equally divided into three groups of high,medium and low dose of *** lour.(60,40 and 20 g/kg),dexamethasone(0.006 g/kg)group and blank group(distilled water).In the cotton ball granuloma test in mice,the mice were given intragastric administration continuously for 7 d after cotton ball *** the experiment of inflammation induced by carrageenan,the mice in each group were given intragastric administration once a day for 7 d,and the inflammatory model was made after the last administration.[Results]The granuloma inhibition rates of mice in the high,medium and low dose groups of water extract and ethanol extract from *** lour.were significantly lower than those in the blank group(P<0.05).The degree of paw edema in each dose group of water and ethanol extract from *** *** significantly lower than that in the blank group(P<0.05).[Conclusions]The water and ethanol extracts from *** *** significant anti-inflammatory effects.
