Nonalcoholic fatty liver disease(NAFLD),a leading chronic disease worldwide,affects approximately a quarter of the global *** steatohepatitis(NASH)is an advanced form of NAFLD and is more likely to progress to liver fibrosis than simple *** is also identified as the most rapidly growing cause of hepatocellular *** in the past decade,several phase II/III clinical trials have shown promising results in the use of novel drugs targeting lipid synthase,farnesoid X receptor signaling,peroxisome proliferatoractivated receptor signaling,hepatocellular injury,and inflammatory signaling,proven pharmaceutical agents to treat NASH are still ***,continuous exploration of the mechanism underlying the pathogenesis of NAFLD and the identification of novel therapeutic targets remain urgent tasks in the *** the current review,we summarize studies reported in recent years that not only provide new insights into the mechanisms of NAFLD development but also explore the possibility of treating NAFLD by targeting newly identified signaling *** also discuss evidence focusing on the intrahepatic targets involved in the pathogenesis of NAFLD as well as extrahepatic targets affecting liver metabolism and function.

Growth potential of human hepatocarcinoma cells in the liver of neonatal immunocompetent mice and its relation to immunological tolerance

To determine the pathological behavior of human hepatocarcinoma cells in the liver microenvironment of neonatal non-immunode-ficient mice, three human hepatocarcinoma cell lines (Bel7402, HepG2, and SK-Hep-1), traced by DiI, were transplanted into the intrahepatic or subcutaneous tissue of neonatal and adult Kunming mice. Histopathological observations showed that cells in the adult liver induced a severe immune response as early as the second day after the implantation, while the subcutaneous neoplasm underwent extensive necrosis by the end of the study. Only the cells injected into the neonatal liver underwent a delayed immunologic rejection in the organ microenvironment. These cells retained recognizable tumor features over the first seven days, and displayed an intrahepatic invasive pattern. The expression of tumor markers including alpha-fetoprotein and survivin was maintained. The quantitative ELISA for the expression patterns of IL-2 and IL-10 also confirmed that the intrahepatic immunity was non-susceptive during this period. The high serum alpha-fetoprotein level was inversely correlated with the change in immune response. Our study provided a bio-system for the research of immune responses to xenografts in the liver.

Immunological aspect of the liver and metastatic uveal melanoma

Uveal(eye)melanoma is the most common primary eye malignancy in *** optimal treatments for primary uveal melanoma,up to 50%of patients subsequently develop systemic metastasis,often in the *** hepatic metastasis develops,the survival of patients is generally short and currently available treatments fail to show meaningful improvement of *** development of immune checkpoint blockades revolutionized immunotherapy for metastatic cutaneous(skin)***,metastatic uveal melanoma is unresponsive to this approach,thus there is an unmet need to improve the treatment of metastatic uveal *** unique characteristic of uveal melanoma is that the majority of metastases first develop in the *** liver is highly specialized in development of immune tolerance to food-derived antigens and consequently serves a unique function in the immune *** the mechanisms by which the liver orchestrates immune-related responses is important to the development of an effective immunotherapy for hepatic metastases such as metastatic uveal *** this review article,the authors overview the immunological aspects of the liver and discuss approaches to improve immunotherapy for metastatic uveal melanoma.
