[目的]建立和优化适合川芎的ISSR-PCR反应体系。[方法]以川芎叶片为材料,利用改良CTAB法提取了叶片基因组DNA,利用单因素试验分析了DNA模板、Mg2＋、dNTP、引物和TaqDNA聚合酶的浓度对ISSR-PCR反应的影响,对适合川芎的ISSR-PCR反应条件进行了优化。[结果]确定了适合川芎的ISSR-PCR反应体系为25.0μl,其中含2.5μl10×TaqDNA聚合酶缓冲液、2.1mmol/LMgCl2、300μmol/LdNTP、0.4μmol/L引物、1.0UTaqDNA聚合酶和20~40ng模板DNA。[结论]为进一步对全国17个不同分布区的川芎资源进行遗传多样性研究奠定了基础。

To investigate the taxonomic value of leaf epidermal characters in developing a classification for Ligusticum (Apiaceae), thirty-eight species (including a cultivar) of the genus were examined using both light and scanning electron microscopy. Our results show that the infrageneric classification of Ligusticum based on the characters of involucel bracteoles, mericarps, and pollen grains was not supported as few epidermal characters were relatively constant at section level. Evidence from leaf epidermis, similar to that from molecular and morphological data, supports the separation of Tilingia (i.e. L. ajanense and L. tachiroei) from Ligusticum. Ligusticopsis was found to be indistinguishable from Ligusticum in epidermal characters. Furthermore, results from epidermal analysis, together with that from cytological evidence, fruit morphology as well as molecular phylogenetics suggest that L. delavayi should be transferred out of Ligusticum.

两面针、川芎、生首乌内生真菌的分离纯化及抑菌性研究

采用组织块法从两面针(Zanthorμlum nitidum(Roxb.)DC)、川芎(Ligusticum chuanxiong Hort)生首乌(Fallopia multiflora)组织中分别分离得到7、11、6株内生真菌,以大肠杆菌(Escherichia coli)、肺炎球菌(Diplococcus pneumoniae)、金黄色葡萄球菌(Staphylococcus aureus)绿脓杆菌(Pseudomonas aeruginosa)、地衣杆菌为供试菌种,对所有两面针、川芎生首乌内生真菌进行抑菌性试验,结果显示,两面针3株内生真菌至少对1株测试病原菌有抑制作用。

Tetramethylpyrazine （TPZ） is an effective component of Ligusticum sinens Oliv. cv. Chuanxiong Hort., one of the Chinese traditional drugs. So far, its coordination effects on vital metal ions have not been reported. We have first prepared TPZ complex of copper （Ⅱ）. The complex was characterized by elemental analysis, infrared and extended X-ray absorption free structure （EXAFS） spectroscopy measurement and coordination microenvironment.
