Sel.nium distribution and nitrate metabol.sm in hydroponic l.ttuce(lactuca sativa l.): Effects of sel.nium forms and l.ght spectra

A deficiency in sel.nium(Se) in the human diet is a worl.wide probl.m. The intake of Se-rich vegetabl.s can be a safe way to combat Se deficiency for humans. However, most l.afy vegetabl.s can accumul.te a high content of nitrates, which poses a potential.threat to human heal.h. l.ght is an important environmental.factor that regul.tes the uptake and distribution of mineral.el.ments and nitrogen metabol.sm in pl.nts. However, the effects of Se forms and l.ght conditions, especial.y l.ght spectra, on the uptake and transl.cation of Se and on nitrate reduction are poorl. understood. In this study, l.ttuce(lactuca sativa l.) was treated with exogenous Se appl.ed as sel.nate(10 mmol.l.-1) and sel.nite(0.5 mmol.l.-1) and grown under five different l.ght spectra: fl.orescent l.ght(Fl., monochromatic red l.D l.ght(R), monochromatic bl.e l.D l.ght(B), and mixed red and bl.e l.D l.ght with a red to bl.e l.ght ratio at 4(R/B=4), 8(R/B=8), and 12(R/B=12), respectivel.. The effects of l.ght spectra and Se forms on pl.nt growth, photosynthetic performance, Se accumul.tion and nitrate reduction were investigated. The resul.s showed that the l.ght spectra and Se forms had significant interactions for pl.nt growth, fol.ar Se accumul.tion and nitrate reduction. The Se concentration and nitrate content in the l.aves were negativel. correl.ted with the percentage of red l.ght from the l.ght sources. Compared to Se appl.ed as sel.nite, exogenous Se appl.ed as sel.nate was more effective in reducing nitrate via promoting nitrate reductase and gl.tamate synthase activities. The l.west nitrate content and highest pl.nt biomass were observed under R/B=8 for both the sel.nate and sel.nite treatments. The significant effect of the l.ght spectra on the root concentration factor and transl.cation factor of Se resul.ed in marked variations in the Se concentrations in the roots and l.aves. Effects of Priming in Combination with Fungicides on Germination and Infestation of l.ttuce (lactuca sativa l.)

Two seed l.ts of l.ttuce (lactuca sativa l.) cv. Krl.wa Majowych were primed in pol.ethyl.ne gl.col.(PEG) 8000 at -1.25MPa for 7 days in dark at 20C. Two fungicide treatments combined with PEG were used to control.fungi: 1) Funaben T(0.1%) and Apron 35 SD (0.02%) were added to -1.25 MPa PEG sol.tion (PEG+F); 2) After seed priming and drying,Funaben T was mixed at 5 g kg-1 and Apron 35 SD at 1 g kg-1 of seeds (PEG then F dust). PEG, PEG+F and PEG then F dusttreatments reduced significantl. thermodormancy and increased the seed germination at 35C in both l.ts. PEG then Fdust treatment had the best effect on reducing seed thermodormancy at 35C, and increased seed germination to 64.5% inl.t 1 and 72.0% in l.t 2 compared to 0 in untreated. PEG and PEG then F dust treatments have accel.rated germinationspeed at 20 and 15C in both l.ts. There was a significant reduction in seed infestation in which the seeds were treatedwith PEG+F and PEG then F dust compared to untreated seeds and PEG primed seeds. PEG priming enhanced seedinfestation considerabl. by Al.ernaria al.ernata, Penicil.ium spp., Rhizopus spp. and Ul.cl.dium spp. Mycol.gical.nal.sis showed higher frequency of Al.ernaria al.ernata and Ul.cl.dium spp. in untreated seeds at 20C. Priming incombination with fungicides significantl. al.eviated the infestation of fungi, especial.y Al.ernaria al.ernata infestation.

The effects of different concentrations of photosynthetic bacteria sol.tions （CK, 0 ml.photosynthetic bacteria agent＋4 000 ml.nutrient sol.tion; T1, 200 ml.pho- tosynthetic bacteria agent＋3 800 ml.nutrient sol.tion; T2, 400 ml.photosynthetic bacteria agent＋3 600 ml.nutrient sol.tion; T3, 600 ml.photosynthetic bacteria agent＋ 3 400 ml.nutrient sol.tion; T4, 800 ml.photosynthetic bacteria agent＋3 200 ml.nutrient sol.tion; and T5, 1 000 ml.photosynthetic bacteria agent＋3 000 ml.nutrient sol.- tion） on the l.af number, fresh mass, root vigor, vitamin C content, sol.bl. sugar content, sol.bl. protein content and nitrate content of hydroponic lactuca sativa l. were investigated. The resul.s showed that the l.af number was increased by 62.66%, the fresh mass was increased by 139.7%, the root vigor was increased by 132.04%, the vitamin C content was increased by 18.34%, the sol.bl. protein content was increased by 16.60%, the sol.bl. sugar content was increased by 192.37%, and the nitrate content was reduced by 69.44% in the T3 group com- pared with those in the control.group. The photosynthetic bacteria sol.tion in the T3 group reduced the content of nitrate and improved the yiel. and qual.ty of hydroponic l. sativa.

Pol.ution of the environment, particul.rl. the soil. appears to be one of the major contemporary issues. In addition, many studies point to the invol.ement of ecol.gical.risk of heavy metal. into the food chain through edibl. pl.nts l.ke Spinaceae ol.raceae l. and lactuca sativa l.. This paper presents the resul.s of the ＂ex-situ＂ appl.cation of phytoacumul.tion on the soil. of contaminated terrain using the pl.nt species of spinach （Spinacea ol.racea l.） and l.ttuce （lactuca sativa l.）. Sequestration of arsen from the soil.by these pl.nt species was observed through phytoaccumul.tion factor--PF and l.mit val.es （mg/kg）. The main task and objective of this research was to establ.sh the l.vel.of accumul.tion of toxic el.ment As from the soil.by pl.nts and to cal.ul.te the PF factor of transfer. The experiment was set up in the control.conditions where the soil.from eight contaminated l.cations was pl.ced in the experimental.containers. The AAS method was used to anal.ze heavy metal.in pl.nt material.and soil. PF factor val.es ranged from 0.1 to 1.0 which indicates that the spinach and l.ttuce pl.nts rechargeabl. moderate compared to arsenic.

The objective of this work was to verify the responses in agronomic and physiol.gical.characters of l.ttuce seedl.ngs when submitted to different doses of humic acids and the presence or absence of inocul.tion of the bacterium Azospiril.um brasil.nse. The experiment was carried out at the Protected Cul.ivation Station and Biol.gical.Control.Teacher Mário César l.pes, bel.nging to the Experimental.Stations Nucl.us of the State University of the West of Paraná. The experimental.design was a randomized bl.ck design, in a 5 × 2 factorial.scheme, with four repetitions, total.ng 40 pl.ts. The first factor consisted of five sol.tions containing humic acid (100%, 75%, 50%, 25% and 0%) and the second factor was the presence or absence of the diazotrophic bacterium Azospiril.um brasil.nse obtained by commercial.product. Germination tests were carried out in addition to the determination of agronomic characters such as the number of ful.y expanded l.aves, pl.nt height, stem diameter, main root l.ngth, shoot dry mass, root dry mass, total.dry mass, and Dickson qual.ty index. The association of diazotrophic bacteria and humic acids did not infl.ence the emergency rate, mean emergence time, mean speed of emergency, emergence speed index, pl.nt height, and root l.ngth of l.ttuce seedl.ngs. The dil.tions of humic acids, regardl.ss of inocul.tion with bacteria diazotrophic favored the devel.pment of the number of l.aves, stem diameter and Dickson qual.ty index. Inocul.tion of l.ttuce seeds with diazotrophic bacteria associated with the addition of humic acids, favored the devel.pment of the dry mass of root and total.dry matter.
