Derivative Spectroscopy and its Application at Detecting the Weak Emission/Absorption lines

The development of spectroscopic survey telescopes like Large Sky Area Multi-Object Fiber Spectroscopic Telescope(LAMOST),Apache Point Observatory Galactic Evolution Experiment and Sloan Digital Sky Survey has opened up unprecedented opportunities for stellar classification.Specific types of stars,such as early-type emission-line stars and those with stellar winds,can be distinguished by the profiles of their spectral lines.In this paper,we introduce a method based on derivative spectroscopy(DS)designed to detect signals within complex backgrounds and provide a preliminary estimation of curve profiles.This method exhibits a unique advantage in identifying weak signals and unusual spectral line profiles when compared to other popular line detection methods.We validated our approach using synthesis spectra,demonstrating that DS can detect emission signals three times fainter than Gaussian fitting.Furthermore,we applied our method to 579,680 co-added spectra from LAMOST Medium-Resolution Spectroscopic Survey,identifying 16,629 spectra with emission peaks around the Hαline from 10,963 stars.These spectra were classified into three distinct morphological groups,resulting in nine subclasses as follows.(1)Emission peak above the pseudo-continuum line(single peak,double peaks,emission peak situated within an absorption line,P Cygni profile,Inverse P Cygni profile);(2)Emission peak below the pseudo-continuum line(sharp emission peak,double absorption peaks,emission peak shifted to one side of the absorption line);(3)Emission peak between the pseudo-continuum line.

Development and characterization of a novel common wheat–Mexico Rye T1DL·1RS translocation line with stripe rust and powdery mildew resistance

Rye(Secale cereale L., 2n=2x=14, RR) is a significant genetic resource for improving common wheat because of its resistance to multiple diseases and abiotic-stress tolerant traits. The 1RS chromosome from the German cultivated rye variety Petkus is critical in wheat breeding. However, its weakened disease resistance highlights the need to identify new resources. In the present study, a novel derived line called D27 was developed from common wheat and Mexico Rye.Cytological observations characterized the karyotype of D27 as 2n=42=21 Ⅱ. Genomic in situ hybridization indicated that a pair of whole-arm translocated Mexico Rye chromosomes were inherited typically in the mitotic and meiosis stages of D27. Experiments using fluorescence in situ hybridization(FISH) and gliadin electrophoresis showed that D27 lacked wheat 1DS chromosomes. They were replaced by 1RS chromosomes of Mexico Rye, supported by wheat simple-sequence repeat markers, rye sequence characterized amplified region markers, and wheat 40K SNP array analysis.The wheat 1DS chromosomes could not be detected by molecular markers and wheat SNP array, but the presence of rye 1RS chromosomes was confirmed. Agronomic trait assessments indicated that D27 had a higher tiller number and enhanced stripe rust and powdery mildew resistance. In addition, dough properties analysis showed that replacing 1DS led to higher viscosity and lower dough elasticity in D27, which was beneficial for cake making. In conclusion, the novel cytogenetically stable common wheat–Mexico Rye T1DL·1RS translocation line D27 offers excellent potential as outstanding germplasm in wheat breeding programs focusing on disease resistance and yield improvement. Additionally,it can be valuable for researching the rye 1RS chromosome’s genetic diversity.

Dynamic Balance Control Based on an Adaptive Gain-scheduled Backstepping Scheme for Power-line Inspection Robots

This paper presents an adaptive gain-scheduled backstepping control(AGSBC) scheme for the balance control of an underactuated mechanical power-line inspection(PLI) robotic system with two degrees of freedom and a single control input.First, a nonlinear dynamic model of the balance adjustment process of the PLI robot is constructed, and then the model is linearized at a nominal equilibrium point to overcome the computational infeasibility of the conventional backstepping technique. Second, to solve generalized stabilization control issue for underactuated systems with multiple equilibrium points,an equilibrium manifold linearized model is developed using a scheduling variable, and then a gain-scheduled backstepping control(GSBC) scheme for expanding the operational area of the controlled system is constructed. Finally, an adaptive mechanism is proposed to counteract the impact of external disturbances. The robust stability of the closed-loop system is ensured by Lyapunov theorem. Simulation results demonstrate the effectiveness and high performance of the proposed scheme compared with other control schemes.

A respiratory syncytial virus persistent-infected cell line system reveals the involvement of SOCS1 in the innate antiviral response

HEp-2 cells persistently infected with respiratory syncytial virus(RSV) are a heterogeneous mixture of viral antigen-positive and-negative variants; however, the mechanism through which viral replication becomes latent remains unclear. In this study, we investigated the potential mechanism by which RSV escapes from innate immune surveillance. Persistent-infected RSV HEp-2 cells were isolated and cell clones were passaged. The RSV-persistent cells produced viruses at a lower titer, resisted wild-type RSV re-infection, and secreted high levels of interferon-β(IFN-β), macrophage inflammatory protein-1α(Mip-1α), interleukin-8(IL-8), and Rantes. Toll-like receptor 3(TLR3), retinoic acid inducible gene-I(RIG-I), and suppressor of cytokine signaling 1(SOCS1) levels were upregulated in these cells. The silencing of TLR3 m RNA decreased the expression of SOCS1 protein and the secretion of cytokines. RSV-persistent cells are in an inflammatory state; upregulation of SOCS1 is related to the TLR3 signaling pathway, which could be associated with the mechanism of viral persistence.

网络表示学习可以有效解决推荐面临的数据稀疏问题.本文对网络表示学习中line算法和DeepWalk算法进行改进,提出混合推荐算法并应用于电影推荐场景.该算法通过学习用户喜好特征、厌恶特征和相似用户特征,生成三个低维特征向量.将三个低维特征向量线性组合拼接成用户表示向量,以余弦相似度作为相似性指标,将相似用户关联的电影推荐给目标用户,实现电影推荐.实验结果表明,所提出的推荐算法相较于次优算法,在MovieLens数据集上的准确率和F 1指标分别提升12%和7%,在MovieTweetings数据集上的准确率和F 1指标分别提升16%和18%.本文提出的基于多维特征表示学习的推荐算法在电影推荐类场景中,具有显著的优越性.

The construction of drug-resistant cancer cell lines by CRISPR/Cas9 system for drug screening

Cancer therapy is often hampered by the rapid emergence of drug resistance. Drug-resistant cellular models are essential for understanding the drug resistance and developing new therapeutics. The low efficiency and long time required in creating these models are major obstacles hindering drug resistance research and drug screening. Herein, we report an approach that can accelerate(shortening the time from years to 3 weeks) the establishment of cancer cell line-based, inheritable drug resistance models by specific knockout of MED12 gene using CRISPR/Cas9 system. The resultant MED12^(KO) A375(melanoma)cell line was resistant to inhibitors of B-Raf proto-oncogene, serine/threonine kinase(BRAF), whereas the resultant MED12^(KO) PC9(non-small cell lung cancer) cell line was resistant to inhibitors of epidermal growth factor receptor(EGFR). Evaluation of anti-cancer drugs and their combinations shows that certain combinations of BRAF inhibitors and TGF-β receptor(TGF-βR) inhibitors are active in suppressing the growth of MED12^(KO) A375 cells, and a few combinations of EGFR inhibitors and TGF-βR inhibitors were active in suppressing the growth of MED12^(KO) PC9 cells. The drug-resistant models will be useful in screening novel drugs and drug combinations for multi-drug-resistant cancer therapy.

Metastatic human HCC model is needed for the studies on mechanism and intervention of metastatic recurrence. By using orthotopic implantation of histologically intact tissues of 30 surgical specimens, a patient like metastatic model of human HCC in nude mice (LCI-D20)and a Iow metastatic model of human HCC in nude mice LCI-D35 ) have been established. All mice with transplanted LCI-D20 tumors exhibited extremely high metastatic ability including spontaneous metastasis to liver, lungs, lymph nodes and peritoneal seeding.Remarkable difference was also found in expression of some of the invasiveness related genes and growth factors between the LCI-D20 and LCI-D35 tumors. PAI-Iincreased gradually following tumor progression in LCID20 model, and correlated with tumor size and AFP level,Phasic expression of tissue intercellular adhesion molecule-I in this model was also observed. Using corneal micropocket model, it was demonstrated that the vascular response induced by LCI-D20 tumor was stronger than that induced by LCI-D35 tumor. Similar report on metastatic human HCC model in nude mice and human HCC cell line with metastatic potential was rarely found in the literature. This LCI-D20 model has been widely used for the studies on intervention of metastasis, including antiangiogenesis, antisense approach, metalloproteinase inhibitor, differentiation inducer, etc. It is concluded that the establishment of metastatic human HCC model in nude mice and human HCC cell line with metastatic potential will provide important models for the in vivo and in vitro study of HCC invasiveness, angiogenesis as well as intervention of HCC recurrence.

DOUBLE CORTICAL line IN THE ACETABULAR ROOF OF PARAPLEGIC PATIENTS

The pelvic radiographs of 60 patients with paraplegia or quadriplegia were reviewed and double cortical line in the acetabular roofs on both sides was found in 35 patients. It was concluded that the double cortical line was a radiologic sign of disuse osteoporosis after spinal cord injury. It was noticed that the acetabular double cortical line in incomplete paralysis was not so prominent as that in complete paralysis (Pline and related factors were discussed.

A cell line （SMC-1） has been established from the resected tissue of a patient with malignant pleural mesothelioma which has been pathologically confirmed to be the mixed type. The cell line is characterized by the pleomorphy and multilaycred growth of the cells with the population-doubling time of approximately 32h, the highest mitotic index of 46—50‰ and the modal chromosome number of 69—70. Transplantation of the cells into the anireals can produce tumors bearing histological resemblance to the original material. The cultured cells, xenografted tumor cells and the cells from the host specimen show similar histochemical expressions. These data meet the requirements of a human malignant pleural mesothelioma cell line. The establishment of this cell line will serve as a useful tool in clinical research of diagnosis and treatment of this devastating disease.

Operation of natural gas and electricity networks with line pack

This paper addresses the coordinated operation of natural gas and electricity networks considering the line pack flexibility in the natural gas pipelines.The problem is formulated as a mixed integer linear programming problem.The objective is to minimize the operation cost of natural gas and electricity networks considering the price of the natural gas supply.Benders decomposition is used to solve the formulated problem.The master problem minimizes the startup and shutdown costs as well as the operation cost of the thermal units other than the gasfired generation units in the electricity network.The first subproblem validates the feasibility of the decisions made in the master problem in the electricity network.And if there is any violation,feasibility Benders cut is generated and added to the master problem.The second subproblem ensures the feasibility of the decisions of the master problem in the natural gas transportation network considering the line pack constraints.The last sub-problem ensuresthe optimality of the natural gas network operation problem considering the demand of the gas-fired generation units and line pack.The nonlinear line pack and flow constraints in the feasibility and optimality subproblems of natural gas transportation network are linearized using Newton-Raphson technique.The presented case study shows the effectiveness of the proposed approach.It is shown that leveraging the stored gas in the natural gas pipelines would further reduce the total operation cost.
