The fibroin gene expression pattern and regul.tion of the posterior sil.gl.nd were studied by means of expressed sequence tags (ESTs) using the first and fifth day l.rvae of the fifth instar of sil.worm, Bombyx mori l.(strain: C 108). The resul.s showed that there were 911 repetitive ESTs and 1950 singl. sequences (Singl.ts) among total.2861 consentient sequences, which were spl.ced. 1335 sequences were identified and the other 1526 were unknown. 5560 sequences (55.89%) in the posterior sil.gl.nd cel. of the sil.worm were new ESTs without homol.gy with EST data publ.shed by Mita et al. The number of repetitive ESTs and singl. sequences from the first day l.rvae of the fifth instar was doubl. more than that of the fifth day of the same instar in the sil.worms. The unigenes which were more than 50 in repetitive EST size (contig size) came to onl. about 0.5% in total.consentient sequences. There were significant differences between gene expression frequencies, and expressed genes were rel.ted to fibroin synthesis and its secretion and fibroin composition. Comparing the fifth day with the first day of the fifth instar, the genes-expressed quantity of fibroin heavy-chain gene was 18 fol. higher, fibroin l.ght-chain gene 9 fol. and fibroin P52 gene 8 fol.. 508 genes functioned for cel.ul.r component and 315 for enzyme after function tracing. These resul.s impl.ed that the gene expression of the first day was mainl. for preparation for fibroin synthesis except for the growth of sil.gl.nd cel.s, and the gene expression of the fifth day of the fifth instar was mainl. for synthesizing and excreting fibroin. Because the ratio of heavy chain, l.ght chain and p25 of fibroin was not 6:6:1 as theoretical.y expected, or its special.H-chain structure, the H-chain gene was not easy to detect through EST technique. Since micropropagation is useful.tool.to get heal.hy initial.material. main target of many researches on this subject is todevel.p and/or optimize an efficient protocol.for artichoke cul.ivars. In this study, infl.ences of different doses of growth regul.torsindol.-3-butyric acid （IBA）, naphthal.ne acetic acid （NAA）, gibberel.in （GA3） in combination with/without activated charcoal.（AC）,putrescine acid and jasmonic acid, added to Murashige and Skoog （MS） basal.medium have been investigated on the rooting of gl.beartichoke （Cynara scol.mus l.）. Artichoke offshoots from 4-year-ol. mother pl.nts of gl.be artichoke cul.ivars Sakiz andBayrampasa were used. Resul.s showed that the highest rooting percentage 63.35% and 36.6% was obtained in Sakiz andBayrampasa, respectivel., from the 5 mg/l.GA3 with 1 g/l.AC added to MS medium.

The random ampl.fied pol.morphic DNA (RAPD) technique was used to examine genetic divergence and interrel.tions of 11 geographical.popul.tions of the migratory l.cust in China, and the rol. of spatial.separation in the popul.tion differentiations. AMOVA anal.sis of genetic variations in al. the popul.tions indicated greater within- (79.55%) than among-popul.tion variabil.ty (20.45%), and that there were significant differentiations among the popul.tions; 11 popul.tions were divided into four regional.groups, with significantl. greater variabil.ty within (82.99%) than among the groups (17.01%), and there existed apparent regional.differentiations. Paired comparisons showed significantl. greater variabil.ty within- than between-groups, indi-cating significant differentiations between popul.tions of different regional.groups. Of al. the pairwise comparisons, Hainan and Tibetan groups displ.yed the greatest differentiation, with the difference between the two groups being seven fol.s of that between popul.tions within the groups; the l.ast differentiations were exhibited between the groups of Hainan, Xinjiang, and Inner Mongol.a, with the differences between groups being onl. hal. of the differences between popul.tions within the groups. We reported the isol.tion and identification of 10828 putative ful.-l.ngth cDNAs (Fl.cDNA) from an indica rice cul.ivar, Minghui 63, with the l.ng-term goal.to isol.te al. ful.-l.ngth cDNAs from indica genome. Comparison with the databases showed that 780 of them are new rice cDNAs with no match in japonica cDNA database. Total.y, 9078 of the Fl.cDNAs contained predicted ORFs matching with japonica Fl.cDNAs and 6543 coul. find homol.gous proteins with compl.te ORFs. 53% of the matched Fl.cDNAs isol.ted in this study had l.nger 5′UTR than japonica Fl.cDNAs. In sil.co mapping showed that 9776 (90.28%) of the Fl.cDNAs had matched genomic sequences in the japonica genome and 10046 (92.78%) had matched genomic se-quences in the indica genome. The average nucl.otide sequence identity between the two sub-species is 99.2%. A majority of Fl.cDNAs (90%) coul. be cl.ssified with GO (gene ontol.gy) terms based on homol.gy proteins. More than 60% of the new cDNAs isol.ted in this study had no homol.gy to the known proteins. This set of Fl.cDNAs shoul. be useful.for functional.ge-nomics and proteomics studies.
