Sevoflurane postconditioning alleviates action potential duration shortening and l-type calcium current suppression induced by ischemia/reperfusion injury in rat epicardial myocytes

Background It has been proved that sevoflurane postconditioning （SpostC） could protect the heart against myocardial ischemia/reperfusion injury, however, there has been few research focused on the electrophysiological effects of SpostC. The objective of the study was to investigate the effects of SpostC on action potential duration （APD） and l-type calcium current （Ica, l） in isolated cardiomyocytes. Methods langendorff perfused SD rat hearts were randomly assigned to one of the time control （TC）, ischemia/reperfusion （I/R, 25 minutes of ischemia followed by 30 minutes of reperfusion）, and SpostC （postconditioned with 3% sevoflurane） groups. At the end of reperfusion, epicardial myocytes were dissociated enzymatically for patch clamp studies. Results Sevoflurane directly prolonged APD and decreased peak Ica, l densities in epicardial myocytes of the TC group （P〈0.05）. I/R injury shortened APD and decreased peak Ica, l densities in epicardial myocytes of the I/R group （P〈0.05）. SpostC prolonged APD and increased peak Ica, l densities in epicardial myocytes exposed to I/R injury （P〈0.05）. SpostC decreased intracellular reactive oxygen species （ROS） levels, reduced the incidence of ventricular tachycardia and ventricular fibrillation, and decreased reperfusion arrhythmia scores compared with the I/R group （all P〈0.05）. Conclusions SpostC attenuates APD shortening and Ica, l suppression induced by I/R injury. The regulation of APD and lea, l by SpostC might be related with intracellular ROS modulation, which contributes to the alleviation of reperfusion ventricular arrhvthmia.

Objective To assess the anti-arrhythmic activity and cardioprotective effects of Wenxin Granula,a traditional Chinese formula(consisting of Salviae Miltiorrhizae Radix,Polygonati Rhizoma,Notoginseng Radix et Rhizoma,Nardostachyos Radix et Rhizoma,Angelicae Sinensis Radix,and Succinum),on heart in ischemic-induced myocardial infarction(MI) rats and compare with those of Amiodarone which have been demonstrated in *** Rats were randomly divided into Sham-operated(control),MI + Amiodarone [5 mg/(kg.d)](MI),and MI + Wenxin Granula [10 mg/(kg.d)] groups and left anterior descending coronary artery was occluded in each *** left anterior descending for 12 h,standard lead II of administration electrocardiogram was recorded in order to analyze the occurrence of *** one month,the size of the infarct area of heart was evaluated by TTC staining method and haemodynamic function was assessed to detect the heart *** scanning confocal microscope and the technique of patch clamp were used to detect the intracellular Ca2+([Ca2+]i) and l-type calcium current(ICa-l),*** Both Wenxin Granula [10 mg/(kg.d)] and Amiodarone [5 mg/(kg.d)] could markedly decrease the incidence of arrhythmia in heart of rats which were subjected to ischemic *** one month,Wenxin Granula could significantly decrease mortality to 22.22% and reduce the infarct area(P lve that Wenxin Granula attenuated [Ca2+]i decreasing in MI ***,Wenxin Granula could obviously ameliorate the impaired heart function of MI rats by decreasing the elevated left ventricular end-diastolic pressure and increasing the attenuated maximum change velocity of left ventricular pressure in the isovolumic contraction or relaxation *** the other hand,electrophysiological experiment results revealed that Wenxin Granula administration one month later also increased the reduced ICa-l density in rat ventricular

Wenxin Keli is the first state-sanctioned traditional Chinese medicine(TCM)--based antiarrhythmic *** present study aimed to examine whether long-term treatment with Wenxin Keli reduces ischemia-induced ventricular arrhythmias in rats in vivo,and if so,which mechanisms are *** rats were treated with either saline(control group)or Wenxin Keli for 3 weeks and were subjected to myocardial ischemia for 30 min with assessment of the resulting ventricular *** l-type calcium current(I)and transient outward potassium current(I)were measured by the patch clamp technique in normal rat cardiac ventricular *** the 30-min ischemia,Wenxin Keli significantly reduced the incidence of ventricular fibrillation(VF)(Plar tachycardia(VT)+VF episodes and the severity of arrhythmias were significantly reduced by Wenxin Keli administration compared to the control group(Pli inhibited I and I in a concentration-dependent *** results suggest that long-term treatment with Wenxin Keli may attenuate ischemia-induced ventricular arrhythmias in rats and that I and I may be involved in this attenuation.

AIM The present experiments were designed to investigate the effects of 5-HT on l-type calcium current (I) and transient outward potassium current (I) in normal and hypertrophied ventricular myocytes and elucidate the mechanism of 5-HT inducing arrhythmia. METHODS currents in rat ventricular myocytes were recorded by whole-cell patch-clamp techniques. RESUlTS 5-HT increased I and shifted the peak current potential from 0 mV to-10 mV in both myocytes. The augmentation of Ica induced by 5-HT was larger in hypertrophied ones. 5-HT did not markedly influence the steady-state activation kinetics. However, 5-HT shifted steady-state inactivation curve with half inactivation voltage V changing from -39.5±1.8 mV to -27.8±1.7 mV (Ple not changing the voltage responsiveness of calcium channel (slope factor к was not changed markedly). Unlike I, I was inhibited by 5-HT in similar degree in both normal and hypertrophied myocytes with the IC of 40 μM. Mianserin (5-HT receptor antagonist), reversed the inhibitory effects of 5-HT on I, suggesting that 5-HT receptor involve in the action of 5-HT on cardiac myocytes. The further studies found that compound 48/80 (phospholipase C antagonist) and chelerythrine chloride (PKC antagonist) also regressed the inhibitory effects of 5-HT on I. On the other hand, phorbol 12-myristate 13-acetate (PMA, PKC agonist) enhanced the inhibitory effect of 5-HT on Ito in normal cardiac myocytes. CONClUSION We conclude that 5-HT activates intracellular phospholipase C signal pathway, affecting the function of calcium and potassium channels through which induce alteration of action potential duration and arrhythmia. 5-HT induces a larger alteration of action potential duration by the augmentation of I, suggesting that 5-HT is more prone to induce arrhythmia in hypertrophied heart than in normal one.
