l-arginine Supplementation Mitigates Dichlorvos-Induced Haematocardiotoxicity, and Oxidative Stress in Male Wistar Rats

Due to its toxicity, dichlorvos—a common organophosphate pesticide—poses significant risks to human health. This study utilized male Wistar rats to explore the potential protective effects of l-arginine supplementation against dichlorvos-induced toxicity, focusing on cardiotoxicity, haematotoxicity and oxidative stress. The rats were divided into four groups: Control, l-arginine (l), Dichlorvos (D), and l-arginine + Dichlorvos (l + D). Dichlorvos was administered to the D group, l-arginine (100 mg/kg) to the l group, and both l-arginine and dichlorvos to the l + D group. The study evaluated various parameters, including cardiovascular, oxidative stress markers, and haematological indices. Significant changes in haematological parameters such as haemoglobin (Hb), haematocrit (HCT), and red blood cell count (RBC) indicated haematotoxicity after dichlorvos administration. Additionally, elevated cardiac markers, including lactate dehydrogenase (lDH) and creatine kinase-MB (CK-MB), suggested cardiotoxic effects. Exposure to dichlorvos also resulted in decreased antioxidant enzyme levels and increased oxidative stress indicators like malondialdehyde (MDA). Remarkably, l-arginine supplementation mitigated the damage caused by dichlorvos. It normalized the altered haematological parameters, demonstrating its protective effect against haematotoxicity. The rise in cardiac markers was reduced with l-arginine supplementation, indicating protection against cardiotoxicity. Moreover, l-arginine significantly decreased oxidative stress, as evidenced by lower MDA levels and restored antioxidant enzyme activity. In conclusion, l-arginine supplementation in male Wistar rats showed promising protective effects against dichlorvos-induced cardiotoxicity, haematotoxicity and oxidative stress. This suggests that l-arginine may offer a beneficial intervention to mitigate the adverse effects of dichlorvos on blood and heart health, paving the way for potential treatments for pesticide poison

In a 2 × 3 factorial arrangement, 3 levels of l-arginine (0, 75 and 150 mg/kg BW per day) were tested at two stages of pregnancy (Early 60 days vs. late 60 days) on the maternal blood metabolites and postnatal milk composition relative to the newborn traits of Najdi ewes. Thirty adult pregnant Najdi ewes were randomly assigned into 6 treatments (n = 5). Control early (CE) ewes were orally administered 50 ml normal saline (0.9% NaCl) daily during the first 60 days of pregnancy;control late (Cl) ewes were similar to CE except saline was given at the last 60 days of pregnancy;low dose of l-arginine at early pregnancy (lE);low dose of l-arginine at late pregnancy (ll);high dose of l-arginine at early (HE) and high dose of l-arginine at late pregnancy (Hl). Blood and milk samples were collected on a weekly basis. Data of lambing and newborn traits were considered. There were non-significant differences in litter size due to l-arginine;however lamb birth and weaning weights increased by 35% in lE-ewes. arginine did not change blood protein and albumin. low arginine decreased lipid fractions, creatinine, urea, and milk fat;however, it increased AST, AlT, milk lactose and milk ash. Administration of l-arginine at 75 mg/Kg BW per day for 60 days at an early gestation in ewes improved the maternal body health and milk composition for the welfare of the neonates.

Hypertension is a major public health problem that adversely affects the health status of individuals, families and communities. l-arginine levels of a total of 90 consecutively-recruited hypertensive subjects and 50 age-matched non-hypertensive controls were studied. Plasma from subjects and control participants were analyzed for l-arginine. The mean values of l-arginine level were significantly lower among the hypertensive subjects which are (174.33 ± 78.31 μmol/l) compared to those of the 50 non-hypertensive controls (237.82 ± 261.16 μmol/l) (p = 0.04). There was no statistically significant difference in the l-arginine levels of hypertensive subjects based on gender, age and ethnici-ty (p = 0.87, 0.23 and 0.57) respectively. The l-arginine level was significantly higher among married hypertensive subjects (181.71 ± 78.17 μmol/l) compared to single or unmarried subjects (130.62 ± 65.99 μmol/l) (p = 0.03). The mean value of l-arginine level was significantly higher among hypertensive subjects with mild blood pressure (187.63 ± 77.93 μmol/l) compared to those with high blood pressure (156.93 ± 76.31 μmol/l). The difference however was not statistically significant (p = 0.05). The findings from this study confirm that the level of l-arginine is lower among hypertensive subjects compared to non-hypertensive controls. Age, gender and ethnicity did not have a significant effect on the l-arginine levels of hypertensive subjects. l-arginine level was significantly lower among single hypertensive patients and those with markedly raised blood pressure. It is recommended the l-arginine supplement be prescribed to hypertensive patient as a prophylactic measure. There is a need to enlighten hypertensive patients in the area on the need to maintain a balanced diet containing sufficient level of l-arginine.

l-arginine is an important component of amino acid injection. Its diffusion in body fluid and blood is of key importance to understand drug diffusion and drug release. As a fundamental demand for study and being a considerably valuable reference for application, in this study, the diffusion coefficients of l-arginine in polyacrylamide（PAM） aqueous solution used as non-Newtonian fluid similar to blood and body fluid were measured using a holographic interferometer. The effects of interaction among molecules and solution concentration on diffusion were analyzed and discussed, respectively. Based on the obstruction-scaling model, a novel modified model was presented for predicting diffusivity of solute in non-Newtonian fluid. Good agreement was achieved between the calculated value and the experimental data.

l-arginine is an amino acid semiessencial considered a precursor of nitric oxide, a gas mainly produced in endothelial cells. Nutritional supplements based on the amino acid l-arginine have been broadly marketed in order to increase vasodilation and the blood supply to muscle in order to optimize metabolic responses induced by exercise. The main objective of this study was to evaluate the effect of l-arginine supplementation on nitric oxide production in response to exercise. Furthermore, the biochemical parameters of muscle fatigue were assessed. Fourteen trained runners were divided in two groups, supplemented with l-arginine (ARG) and placebo (PlA). Blood samples were collected before supplementation (T0), immediately after the first exercise session (T1), immediately after the second exercise session (T2), and after 20 minutes of rest (T3). Plasma cyclic guanosine monophosphate was assessed as a marker of nitric oxide production. The biochemical parameters of muscle fatigue analyzed were plasma lactate and ammonia. There was significant increase in plasma cyclic guanosine monophosphate in both groups in response to exercise: ARG (T0: 3.6 ± 1.4;T1: 17.9 ± 5.8;T2: 15.9 ± 5.3;T3: 7.3 ± 2.5 pmol/ml) and PlA (T0: 4.1 ± 1.1;T1: 18.8 ± 9.9;T2: 16.1 ± 3.5;T3: 9.3 ± 3.7 pmol/ml). A significant reduction in plasma lactate and ammonia were observed in the recovery period after exercise (T3). However, no significant difference was observed between groups in the variables studied. Therefore, l-arginine supplementation was unable to increase the effects of exercise on nitric oxide production and did not improve the metabolic responses to exercise in runners.

A 2 × 2 factorial arrangement was designed to test effects of supplementation of a low (l, 75 mg/kg BW) vs. high (H, 150 mg/kg BW) l-arginine given at early (first 56 days) vs. late (last 56 days) pregnancy on maternal hormones and neonatal traits. Thirty Najdi pregnant ewes were randomly allocated into 6 groups. Ewes in G1 and G2 served as controls (C), given 50 ml saline at either early (CE) or late (Cl) pregnancy, respectively. G3 and G4 ewes in early pregnancy received low (lE) and high l-arginine (HE), respectively. G5 and G6 ewes in late pregnancy received low (ll) and high (Hl) l-arginine, respectively. A weekly blood sample was collected from initiation of the treatment till parturition. Serum growth hormone (GH), insulin-like growth factor-I (IGF-I), insulin, progesterone (P4) and estradiol 17 β (E2) profiles were determined. Neonatal traits were also determined. Insulin was higher (P 0.05). lamb survival rates at birth in lE ewes were highest (100%) compared to other treatments. In conclusion, supplementing pregnant ewes with low dosage of l-arginine at early stage of gestation increased lamb birth weight and survival, and improved maternal health.

Aim: To assess the effects of l-arginine (l-Arg) supplementation on pulmonary oxidative stress and antioxidant defenses in rats after exhaustive exercise. Methods: Rats were randomly divided into four groups: sedentary control (SC), sedentary control with l-Arg treatment (SC+Arg), exhaustive exercise with control diet (E) and exhaustive exercise with l-Arg treatment (E+Arg). Rats in groups SC+Arg and E+Arg received a 2% l-Arg diet. Rats in groups E and E+Arg underwent an exhaustive running test on a motorized treadmill. Pulmonary oxidative stress indices [xanthine oxidase (XO), myeloperoxidase (MPO), and malondialdehyde (MDA)] and antioxidant defense systems [superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPX), glutathione reductase (GR), and glutathione (GSH)] were investigated in this study. Results: l-Arg supplementation significantly reduced exercise-induced elevations of XO and MPO activities in lung. lArg reversed the exercise-induced increase in SOD and GR activities, but increased CAT and GPX activities. l-Arg administration also significantly increased the GSH levels in plasma. Conclusion: l-Arg supplementation can prevent elevations of XO and MPO activities in the lung and favorably influence pulmonary antioxidant defense systems after exhaustive exercise.
