l-arginine/nitric oxide regulates skeletal muscle development via muscle fibre-specific nitric oxide/mTOR pathway in chickens

l-arginine (l-Arg), the precursor of nitric oxide (NO), plays an important role in muscle function. Fasttwitchglycolytic fibres are more susceptible to age-related atrophy than slow-twitch oxidative *** effect of l-Arg/NO on protein metabolism of fast- and slow-twitch muscle fibres was evaluated inchickens. In Exp. 1, 48 chicks at 1 day old were divided into 4 groups of 12 birds and subjected to 4treatments: basal diet without supplementation or supplemented with 1% l-Arg, and water supplementedwith or without l-nitro-arginine methyl ester (l-NAME, 18.5 mM). In Exp. 2, 48 chicks weredivided into 4 groups of 12 birds fed with the basal diet and subjected to the following treatments: tapwater (control), tap water supplemented with l-NAME (18.5 mM), or molsidomine (MS, 0.1 mM), or18.5 mM l-NAME t 0.1 mM MS (NAMS). The regulatory effect of l-Arg/NO was further investigatedin vitro with myoblasts obtained from chicken embryo pectoralis major (PM) and biceps femoris (BF).In vivo, dietary l-Arg supplementation increased breast (t14.94%, P le mass(t23.40%, P le influence. However, l-NAME treatmentblocked the beneficial influence of l-Arg on muscle development. l-Arg decreased (P lated mTOR and ribosomal protein S6 kinase beta-1 (p70S6K) levels in breastmuscle, which was recovered by l-NAME treatment. In vitro, l-Arg or sodium nitroprusside (SNP)reduced protein synthesis rate, suppressed phosphorylated mTOR/p70S6K and decreased atrogin-1 andmuscle RING finger 1 (MuRF1) in myoblasts from PM muscle (P l-NAME abolished the inhibitoryeffect of l-Arg on protein synthesis and the mTOR/p70S6K pathway. However, myoblasts from BF muscleshowed the weak influence. Aim: To examine the effect of l-arginine, the substrate of nitric oxide (NO) synthase, on reproductive function ofmale rats. Methods: Male rats were gavaged with either l-arginine (100 or 200 mg·kg^(-1)·d^(-1)), D-arginine (200mg·kg^(-1)·d^(-1)) or vehicle (0.9% NaCl) for seven consecutive days. Their sexual behaviour and fertility were evaluat-ed using receptive females. Results: l-arginine (200 mg/kg) had no significant effect on sexual competence (interms of sexual arousal, libido, sexual vigour and sexual performance). In mating experiments, the higher dose of l-arginine effectively and reversibly inhibited fertility, whilst the lower dose and the inactive stereoisomer D-arginine hadno significant effect. The antifertility effect caused by l-arginine was due to a profound elevation in the preimplantationloss mediated possibly by impairment in epididymal sperm maturation, hyperactivated sperm motility and sperm capaci-tation. Conclusion: Elevated NO production may be detrimental to male fertility.

Water boosted CO_(2)/C_(2)H_(2)separation in l-arginine functionalized metal–organic framework

The separation of CO_(2)/C_(2)H_(2)mixture by CO_(2)-selective sorbents is an energy-efficient C_(2)H_(2)purification technique,but is strategically challenging due to their similar molecular size and physicochemical ***,water is inevitable in CO_(2)/C_(2)H_(2)mixture and it is usually a significant barrier because of its competitive adsorption with CO_(2).To address this challenge,herein,we report the first example of metal–organic framework(MOF)that exhibits water-boosted CO_(2)adsorption and CO_(2)/C_(2)H_(2)separation by anchoring l-arginine(ARG)on the Zr6 cluster of *** CO_(2)affinity and capacity in the resulting MOF-808-ARG are markedly facilitated by the presence of water,while the C_(2)H_(2)adsorption is significantly ***,CO_(2)adsorption capacities in adsorption isotherm and breakthrough measurement are increased to 143%and 184%,*** addition,the wet MOF-808-ARG exhibits the record CO_(2)/C_(2)H_(2)selectivity of 1,180 under zero *** experiments reveal that CO_(2)/C_(2)H_(2)mixture can be completely separated and the result of mass spectrometry indicates that the C_(2)H_(2)purity in the outlet is up to 99.9%.In situ infrared(IR)results and density functional theory(DFT)calculations reveal the water-promoted CO_(2)adsorption mechanism that the formation of bicarbonate products in the presence of water is thermodynamically and kinetically more favorable than that without ***,MOF-808-ARG also possesses excellent water stability and excellent regeneration of CO_(2)*** work provides a new paradigm by transforming the negative effects of water into positive ones for CO_(2)/C_(2)H_(2)separation.

The objective of this study was to investigate the effects of dietary administration of l-arginine(Arg)or N-carbamylglutamate(NCG)on hepatic energy status and mitochondrial functions in suckling Hu lambs with intrauterine growth retardation(IUGR).Forty-eight newborn Hu lambs of 7 d old were allocated into 4 treatment groups of 12 lambs each,in triplicate with 4 lambs per replicate(2 males and 2 females)as follows:CON(lambs of normal birth weight,4.25±0.14 kg),IUGR(3.01±0.12 kg),IUGR+1%Arg(2.99±0.13 kg),or IUGR+0.1%NCG(3.03±0.11 kg).The experiment lasted for 21 d,until d 28 after birth,and all lambs were fed milk replacer as a basal *** with IUGR lambs,NCG or Arg administration increased(Plevel and the activities of com-plexes I/III/IV,isocitrate dehydrogenase and citrate synthase in the *** with CON lambs,the relative mRNA levels of adenosine monophosphate-activated protein kinaseα1(AMPKα1),peroxisome proliferator-activated receptorγcoactivator-1α(PGC1α)and transcription factor A(TFAM)were increased(Pliver of IUGR lambs,but were decreased(Pliver of NCG-or Arg-treated lambs compared with those in the IUGR *** with IUGR lambs,NCG or Arg administration decreased(Pl AMPKα(tAMPKα)-to-phosphorylated AMPKα(pAMPKα)ratio and the protein expression of PGC1αand *** results suggested that dietary Arg or NCG supplements improved hepatic energy status and mitochondrial function and inhibited the AMPK-PGC1α-TFAM pathway in IUGR suckling lambs.

Main observation and conclusion An enzyme-free amperometric sensor based on a heptadecapeptide possessing an electroactive ferrocene(Fc)linker as ferrocene-Gly-Gly-Gly-Gly-Phe-Gly-His-Ile-His-Glu-Gly-Tyr-Gly-Gly-Gly-Gly-lys-(CH_(2))_(4)-dithiocyclopentane self-assembled on gold substrate was designed and fabricated for specific determination of l-arginine(l-Arg).The detection mechanism is based on conformational change of surface-immobilized peptide induced by the target l-Arg,which was confirmed via SEM,TEM,AFM,XPS,and SPR *** binding affinity and the recognition feasibility of immobilized specific and non-specific peptides were also assessed using electrochemical impedance spectroscopy(EIS),cyclic voltammetry(CV),and differential pulse voltammetry(DPV).The proposed method can serve as“signal-on”sensor for detection of l-Arg down to 31 pmol/l with broad linear range(0.0001 to 10μmol/l).Furthermore,the Fc-conjugated specific peptide sensor was successfully applied to the determination of l-Arg in pig serums with a recovery rate of 97.5%—106.9%,and its test results are in good agreement with that of chromatographic instrument,evidencing that the oligopeptide-based sensor can be served as a simple and enzyme-free biosensing platform towards l-Arg for future application.

Dynamic alterations in early intestinal development,microbiota and metabolome induced by in ovo feeding of l-arginine in a layer chick model

α,β-amyrin, a natural triterpenoid ameliorates l-arginine-induced acute pancreatitis in rats

AIM: To study the benef icial effects of triterpene α,β-amyrin and the underlying mechanisms in an experimental pancreatitis model. METHODS: Acute pancreatitis was induced in five groups of rats (n = 8) by l-arginine (2 × 2.5 g/kg, intraperitoneal, 1 h apart) and 1 h later, they received a single oral dose of α,β-amyrin (10, 30 and 100 mg/kg),methylprednisolone (30 mg/kg) and vehicle (3% Tween 80). A saline (0.9% NaCl) treated group served as a normal control. Efficacy was assessed at 24 h by determination of serum levels of amylase, lipase and proinflammatory cytokines [tumor necrosis factor (TNF)-α and interleukin (Il)-6], pancreatic myeloperoxidase (MPO) activity, lipid peroxidation [thiobarbituric acid reactive substances (TBARS)], nitrate/nitrite levels, and the wet weight/body weight ratio. Tissue histology and the immunoreactivity for TNF-α and inducible nitric oxide synthetase (iNOS) were performed. RESUlTS: α,β-amyrin and methylprednisolone treatments significantly (P l-arginine-induced increases in pancreatic wet weight/body weight ratio, and decreased the serum levels of amylase and lipase, and TNF-α and Il-6, as compared to the vehicle control. Also, pancreatic levels of MPO activity, TBARS, and nitrate/nitrite were signifi cantly lower. Histological f indings and TNF-α and iNOS immunostaining further confirmed the amelioration of pancreatic injury by α,β-amyrin. CONClUSION: α,β-amyrin has the potential to combat acute pancreatitis by acting as an anti-in? ammatory and antioxidant agent.

This study aimed to investigate the effects of dietary supplementation of underfed Hu ewes from d 35 to110 of gestation with either rumen-protected l-arginine(RP-Arg)or N-carbamylglutamate(NCG)on placental amino acid(AA)transport,angiogenic gene expression,and steroid *** d 35 of gestation,32 Hu ewes carrying twin fetuses were randomly divided into four treatment groups,each consisting of eight ewes,and were fed the following diets:A diet providing 100%of NRC’s nutrient requirements for pregnant ewes(CON);A diet providing 50%of NRC’s nutrient requirements for pregnant ewes(RES);RES diet plus 5 g/d NCG(RES+NCG);or RES diet plus 20 g/d RP-Arg(RES+ARG).On the d 110 of pregnancy,blood samples were taken from the mother,and samples were collected from type A cotyledons(COT;the fetal portions of the placenta).The levels of 17β-estradiol and progesterone in the maternal serum and both the capillary area density(CAD)and capillary surface density(CSD)in type A COT were decreased in response to Arg or NCG supplementation when compared to the RES *** concentrations of arginine,leucine,putrescine and spermidine in type A COT were higher(Plevels of inducible nitric oxide synthase(iNOS)and solute carrier family 15,member 1(SlC15A1)were increased(Ple those of progesterone receptor(PGR)and fibroblast growth factor 2(FGF2)were decreased in type A COT by supplementation with either NCG or RP-Arg compared to the RES *** results suggest that providing underfed pregnant ewes from d 35 to 110 of gestation with a diet supplemented with NCG or RP-Arg improves placental AA transport,and reduces the expression of angiogenic growth factor genes and steroid anabolism,leading to better fetal development.
