Succinylated whey protein isolate as a sustained-release excipient of puerarin derivative oral tablets:Preparation,optimization and pharmacokinetics

This work was done to investigate succinylated commercial whey protein isolate(S-WPI)as an oral sustained-release delivery carrier for puerarin 5(PR-5). The succinylation condi-tions were established for S-WPIs by optimization of single factor study and Box–Beehnkendesign. The effect of succinylation degree on S-WPIs solubility was evaluated. Physicochem-ical properties of S-WPIs dried by different three methods on their flow ability, particle size,morphology and in vitro release behavior were studied. After preparing PR-5 sustained re-lease protein tablets with S-WPIs as the carrier by direct powder compression method, thedrug release were studied in vitro and the oral pharmacokinetics and bioavailability wasevaluated using in vivo dog model. It was observed that concentration of substrate has asignificant effect on succinylation. Release behavior in vitro showed spry dried S-WPIs with100% succinylation rate and 30% drug loading would be applied to the preparation of PR-5 sustained-release protein tablets based on the swelling mechanism(protein loss). Comparedwith PR-5 conventional tablet with oral administration, T max value of PR-5 sustained-releaseprotein tablets was approximately 1.58 fold greater than those of the conventional tablets asfurther evidenced by the significantly prolonged MRT and T 1/2. The findings demonstratedthat spray-dried S-WPIs has potential as a promising functional excipient for the design of PR-5 oral sustained-release tablets which can fully improve sustained-release effect and oral bioavailability.

Effect of Pressurized Soy Protein isolate upon the Growth and Antioxidants Functions of SD Rat

The objective of this study is to specify the effect of pressurized soy protein isolate (pSPI), upon the physical development, growth hormones and antioxidants functions of SD rats. The methodology depends on the selection of one hundred male SD rats, divided randomly into 5 groups. Each group consists of 20 rats. The groups will be defined as one blank control group, three groups with pSPI at low, medium and high doses and another control group with native soy protein isolates (nSPI). Low, medium and high doses are represented by 0.333 g/kg, 1.667 g/kg and 3.333 g/kg pSPI per weight, respectively. The native soy protein isolate is represented by 3.333 g/kg nSPI per weight. In every group, four animals will be taken out to collect the blood samples and analyze insulin like growth factor-I, growth hormone, thyroid stimulating hormone, thyroxine and triiodothyronin. The other SD rats will be subjected to feeding for 63 consecutive days. The body weight, the body length and food intake of each rat are measured. The total antioxidant capacity, superoxide dismutase (SOD), malondialdehyde and Glutathione Peroxidase in liver and serum of each rat will be analyzed. The results indicated that the groups with medium and high dose of pSPI result in an obvious increase in the body weight, body length and food utilization rate of SD rats. Also, pSPI has a great effect on the growth and antioxidants functions of SD rat.

Serum-Derived Bovine Immunoglobulin/Protein isolate Therapy for Patients with Refractory Irritable Bowel Syndrome

Background: A small double-blind study showed benefits of serum-derived bovine immunoglobulin/protein isolate (SBI), for diarrhea-predominant irritable bowel syndrome (IBS-D) [1]. The purpose of this chart review was to assess safety and clinical outcomes of SBI in refractory irritable bowel syndrome (IBS) patients. Methods: A retrospective review of 35 IBS patients with diarrhea or mixed diarrhea/constipation pattern (IBS-M) who were administered SBI 5 grams twice daily was performed. Clinical response (“good response” or “no response”) and adverse events were determined by follow-up after four weeks of therapy. Patients were included for evaluation if a lactulose breath test (LBT) had been performed prior to SBI. All patients were refractory to common IBS therapies. The response rate to the inclusion of SBI was calculated in three separate groups: dividing patients based on their LBT results (positive or negative), dividing patients by their IBS diagnosis (IBS-D or IBS-M) and grouping all patients together. Results: Analysis was carried out on 26 IBS-D/-M patients with LBT results. Two patients were lost to follow-up and were excluded from data analysis. The positive LBT group (N = 11) had a 73% (p = 0.117) positive response rate to SBI. The negative LBT group (N = 13) had a significant response rate of 77% (p = 0.040). If patients were divided by IBS diagnosis or grouped together, the response rate to SBI was similarly ranging from 69% - 88%. Adverse events leading to cessation of SBI occurred in 3 of 24 patients. Conclusion: SBI appeared to be a safe and effective nutritional moiety in refractory IBS-D and IBS-M patients. Larger, double-blind studies are needed.

Antibacterial activity of plant methanolic extracts on a field isolate of <i>Pseudomonas syringae</i>pvtomato from the Casablanca region (Morocco)

A bacterial field isolate recovered from infected tomato plants in a green-house at Sidi Rehal, a region near Casablanca city (Morocco), was identified as the gammaproteobacterium Pseudomonas syringae pv. tomato DC3000 strain, the causal agent of bacterial speck. The bacterial isolate was characterized by morphological, biochemical and molecular biological tests, its growth curves carried out in various culture media, and its phytopathogenicity verified by infection tests. A screening was performed to evaluate the antibacterial activity of methanolic extracts of 12 selected Moroccan plants against the P. syringae pv. tomato DC3000 isolate, and Agar-well diffusion and Broth microdilution methods were used to determine minimum inhibitory and minimum bactericidal concentrations. Among the methanolic extracts tested, only those of Nigella sativa, Geranuim robertianum, Aizoon canariense and Rubia peregrine showed clear inhibitory and bactericidal activities, although the highest values were achieved with N. sativa, a plant used in Morocco as a spice, condiment and medicinal treatment.

Jackfruit Seeds Protein isolate by Spray Drying Method: The Functional and Physicochemical Characteristics

With increased demand for plant based proteins by the consumers, the food manufacturers appeal for the new plant proteins with predetermined characteristics. This study aims at isolating the protein fraction from jackfruit seeds and characterizing the protein powder for functional and physicochemical properties. The protein part of the seeds was separated through pH treatments and centrifugation process and finally, the concentrate was converted into powder by spray drying method. The functional properties such as solubility, gelling capacity and emulsion properties and the physicochemical properties such as crystallinity, morphology and particle size distribution of the jackfruit seeds protein isolate (JSPI) were studied. The secondary structural elements of JSPI were also determined by Fourier-transform infrared (FTIR) spectroscopy. About 76.89% protein was estimated in the prepared JSPI with 78.44% solubility in an aquatic solvent. The least gelation concentration of JSPI was 12% in a salt solution. The pH of the solvent significantly affected the emulsifying and foaming properties. The protein isolate possessed amorphous structure, moderate bulk density and almost 75% of the particles fell in a similar size distribution range. The conformational study reported that the β-sheet is the dominant secondary structural element with the highest content of 50.28%. The observed features suggest that the JSPI holds satisfactory functional and physicochemical characteristics for being used in protein-enriched foods.

Mortality rates from a Nigerian isolate of the <i>Infectious Bursa Disease Virus</i>and passive haemagglutination antibody titer that protects chicks against challenge with the virus isolate

To determine passive haemagglutination (PHA) antibody titer that would protect chicks against Nigerian isolates of the Infectious Bursa Disease Virus (IBDV), five groups of chicks aged 30 days which had different antibody titers were challenged with a Nigerian isolate of virulent IBDV. Mortality rates of the different groups were plotted against their respective mean PHA antibody titers. A group with zero antibody titer had a mortality rate of 75% while those with PHA antibody titers of 185.6, 243.2, 256 and 307.2 had mortality rates of 40%, zero, zero and zero respectively. Linear equation generated for a line of best fit of the graph of mortality rates of the chicks on their IBD antibody titers gave antibody titer (X) at which mortality (Y) would be zero as 300. A mortality of 75% and the high antibody level needed to protect chicks suggest that the isolate may be a hypervirulent strain.

Preparation and Properties of Plywood Adhesives with Soy Protein isolate Modified by SDS

The effects of mass concentration,the reaction temperature and the reaction time of soy protein isolate(SPI) and sodium dodecyl sulfate (SDS) on the adhesive strength of the modified SPI adhesives has been studied through orthogonal *** the adhesive mechanism of the modified SPI adhesives was *** results show that the optimum formula conditions are as follows:mass concentration of SPI was 14%,that of SDS 1%,reaction temperature 35℃and the reaction time 3 *** the optimum conditions, the dry adhesive strength of the modified SPI adhesives was 1.82 MPa,the wet adhesive strength was 0.82 MPa,the viscosity was 5.8 Pa·s,the solid content was 12.96%.After SDS was added into SPI, the composites of SDS-SPI were formed,the internal hydrophobic groups among the SPI molecule structure were turned out and the water resistance of the modified SPI adhesives was enhanced with an increase of the modification *** the concentration of SDS was over a defined value,the disulfide bond was broken and the modification effect of SDS went bad.

Degradation of Phenolic Compounds in Coal Gasification Wastewater by Biofilm Reactor with isolated Klebsiella sp.

This study was conducted to evaluate the degradation of phenolic compounds by one strain isolated from coal gasification wastewater( CGW). 16S rRNA gene sequences homology and phylogenetic analysis showed that the isolate is belonged to the genus Klebsiella sp. The effect of different phenolic compounds on the isolate was investigated by determining OD600and phenoloxidase activity,of which the results showed that the isolate can utilize phenol,4-methyl phenol,3,5-dimethyl phenol and resorcinol as carbon resources. The biofilm reactor( formed by the isolate) can resist the influent concentration of phenolic compounds as high as750 mg /L when fed with synthetic CGW and incubated at optimum conditions. The capacity of improving the biodegradability of CGW through degrading phenolic compounds was testified with fed the biofilm reactor with real CGW. Thus,it might be an effective strain for bioaugmentation of CGW treatment.

Assessment of in vitro sensitivity of Plasmodium vivax fresh isolates

Objective:To compare the applicability of the SYBK Grcen-Ⅰ assay with the standard schizont maturalion assay,for determination of sensitivity of Plasmodium vivax(***) to chloroquine and a new antifolale WR ***:The study was conducted at Mae Tao Clinic for migrant workers,Tak Province during April 2009 to July 2010.A total of 64 blood samples(1 mL blood collected into sodium heparinized plastic tube) were collected from patients with monoinfection with *** malaria prior to treatment with standard regimen of a 3-day chloroquine. In vitro sensitivity of *** isolates was evaluated by schizont maturation inhibition and SYBR Green-Ⅰ ***:A total of 30 out of 64 blood samples collected from patients with *** malaria were successfully analyzed using both the microscopic schizont maturation inhibition and SYBR Green-I *** failure rates of the schizont maturation inhibition assay(50%) and the SYBR Green-I assay(54%) were similar(P=0.51).The median IC_(10)s,IC_(50)s and IC_(90)s of both chloroquine and WR99210 were not significantly different from the clinical isolates of *** *** on the cut-off of 100 nM,the prevalences of chloroquine resistance determined by schizont maturation inhibition and SYBR Green-I assays were 19 and 11 isolates,*** strength of agreement between the two methods was very poor for both chloroquine and ***:On the basis of this condition and its superior sensitivity,the microscopic method appears better than the SYUK Green-I Green assay for assessing in vitro sensitivity of fresh *** isolates to antimalarial drugs.

Late leaf spot caused by Cercosporidium personatum is one of the most widespread groundnut leaf diseases. Along with early leaf spot and rust, it is one of the main fungal diseases hampering groundnut production worldwide. Late leaf spot accounts for significant yield losses throughout the world where groundnuts are grown. This reduction in yield caused by the disease could result in yield losses of between 50% and 70%. In Burkina Faso, the disease is present throughout the country, with incidence varying according to region and season. Could the variability in the incidence of the disease be linked to the nature of the isolates or to the conditions from each agro-ecological zone? In this study, the aim was to assess the capacity of three isolates from three agroclimatic zones of the country to defoliate and reduce groundnut yield. To this end, three isolates of Cercosporidium personatum (Berk. Et M.A Curt.) were collected in these zones and evaluated on three contrasting groundnut varieties. A split-splot design was used for the experiment. isolates were prepared from samples collected in farmers’ fields. After incubation in the laboratory, leaf spots showing good sporulation were scraped off with a scalpel after immersing the leaves in distilled water. Inoculations were carried out under controlled environment. After inoculation with the isolate, the percentage of defoliation and the reduction in yield of these varieties were then evaluated. Inoculation was carried out from the 30th day after sowing with Cercosporidium personatum spore suspensions at 105 spores/ml. The study showed that the percentage of defoliation (P = 0.0001) and the reduction in yield (P = 0.0001) were significant. The study revealed that, whatever the variety, isolate I3TF from the Upper Basins region in the South Sudanese zone caused the greatest defoliation and the greatest reduction in yield.
