The purpose of the study was to construct mucosal vaccine of a recombinant *** expressing PRRSV ORF6 gene and evaluate mucosal and systemic immune response against PRRSV in mice after intranasal *** result show that the vaccine can stimulate mice to produce specific IgG in serum and remarkable special s-IgA in lung lavage fluid,at the same time,the contents of cytokines IL-2 and IFN-γ of the experimental group were significant higher than those of the control group(P<0.01),however,the contents of cytokines IL-4 was not different to the all *** summary,the constructed mucosal vaccine can significantly induce mucosal immune,humoral immunity and cellular immunity involved Th1 type cytokines,which will lay a theoretical foundation on immune mechanism and new efficient vaccines for PRRSV.

intranasal boosting with RBD-HR protein vaccine elicits robust mucosal and systemic immune responses

The emergence of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)variants has decreased the efficacy of SARs-CoV-2 vaccines in containing coronavirus disease 2019(CoVID-19)over time,and booster vaccination strategies are urgently necessitated to achieve sufficient *** immunization can improvemucosal immunity,offer-ing protection against the infection and sustaining the spread of *** this study,an intranasal booster of the RBD-HR vaccine after two doses of the mRNA vaccine significantly increased the levels of specific binding antibodies in serum,nasal lavage fluid,and bronchoal-veolar lavage fluid compared with only two doses of mRNA *** intranasal boosting with the RBD-HR vaccine,the levels of serum neutralizing antibodies against prototype and variant strains of SARS-Cov-2 pseudoviruses weremarkedly higher than those in mice receiving mRNA vaccine alone,and intranasal boosting with the RBD-HR vaccine also inhibited the bind-ing of RBD to hACE2 ***,the heterologous intranasal immunization regimen promoted extensive memory T cell responses and activated CD103+dendritic cells in the respiratory mucosa,and potently enhanced the formation of T follicular helper cells and germinal center B cells in vital immune organs,including mediastinal lymph nodes,inguinal lymph nodes,and ***,these data infer that heterologous intranasal boosting with the RBD-HR vaccine elicited broad protective immunity against SARS-CoV-2 both locallyandsystemically.

The rapid mutation and spread of SARS-CoV-2 variants urge the development of effective mucosal vaccines to provide broadspectrum protection against the initial infection and thereby curb the transmission ***,we designed a chimeric tripleRBD immunogen,3Ro-NC,harboring one Delta RBD and two Omicron RBDs within a novel protein scaffold.3Ro-NC elicits potent and broad RBD-specific neutralizing immunity against SARS-CoV-2 variants of ***,intranasal immunization with 3RoNC plus the mucosal adjuvant KFD(3Ro-NC+KFDi.n)elicits coordinated mucosal IgA and higher neutralizing antibody specificity(closer antigenic distance)against the Omicron *** Omicron-challenged human ACE2 transgenic mice,3Ro-NC+KFDi.n immunization significantly reduces the tissue pathology in the lung and lowers the viral RNA copy numbers in both the lung(85.7-fold)and the nasal turbinate(13.6-fold).Nasal virologic control is highly correlated with RBD-specific secretory IgA *** data show that 3Ro-NC plus KFD is a promising mucosal vaccine candidate for protection against SARS-CoV-2 Omicron infection,pathology and transmission potential.

＜正＞Aim To study the effects of STAg intranasally immunized on protection of mice against Toxoplasma gondii and determines the optimal dose of STAg for intranasal immunization. Methods BALB/c mice were intranasally immunized with 5μg, 10μg, 20μg, 30μg STAg per mouse twice at an internal of 2 weeks, while mice intranasally administrated with PBS in the same means were control. All mice were challenged intragastrically with 4×104 tachyzoites per mouse on day 14 after last immunization. The condition of mice infected about health and death was observed and the weight of mice was recorded every day after challenge. Mice were killed on the 30th day after challenge, the tachyzoites of their spleens and brains were counted. Serum IgG and IgA in feces were detected by ELISA. Lymphocytes of IEL were counted. Results In 20μg group and 30μg group, the survival of mice was higher than that of 5μg group, 10μg group and control （P＞0.05）. The weight of control mice gradually decreased after challenge, while that of 5μg group （P＞0.05）, 10μg group （P0.05）, 20μg group （P＜0.05） and 30μg group （P＜0.05） still remained increasing. Compared with 5μg group, lOug group and control, the tachyzoite load in spleens and brains in 20μg group and 30μg group was significantly lower （P＜0.05）. The level of IgG,IgA and IEL in all immunized mice was higher than that of control （P＞0.05）. Conclusion STAg of different doses can induce immune response against Toxoplasma gondii. 20μg and 30μg STAg can induce more protective immunity against Toxoplasma gondii.
