An early evaluation of translocation actions for endangered plant species on Mediterranean islands

In situ conservation is widely considered a primary conservation strategy. Plant translocation, specifically, represents an important tool for reducing the extinction risk of threatened species. However, thus far, few documented translocations have been carried out in the Mediterranean islands. The CareMediflora project, carried out on six Mediterranean islands, tackles both short-and long-term needs for the insular endangered plants through in situ and ex situ conservation actions. The project approach is based on using ex situ activities as a tool to improve in situ conservation of threatened plant species.Fifty island plants(representing 45 taxa) were selected for translocations using common criteria.During the translocations, several approaches were used, which differed in site selection method, origin of genetic material, type of propagative material, planting method, and more. Although only preliminary data are available, some general lessons can be learned from the experience of the CareMediflora project. Among the factors restricting the implementation of translocations, limited financial resources appear to be the most important. Specific preliminary management actions, sometimes to be reiterated after translocation, increase the overall cost, but often are necessary for translocation success. Translocation using juvenile/reproductive plants produces better results over the short term,although seeds may provide good results over the long run(to be assessed in the future). Regardless,plant translocation success can only be detected over long periods; therefore, proper evaluation of plant translocations requires a long-term monitoring protocol. Care-Mediflora project represents the first attempt to combine the existing approaches in a common plant conservation strategy specifically focusing on the Mediterranean islands.

Impairment of LTP in insular cortex is correlated to resilience and vulnerability to chronic stress for PTSD

OBJECTIVE Exposure to stressful events can be differently perceived by individuals depending on the level of stress resilience or vulnerability.The neural processes that underlie such clinical y and social y important differences are largely unknown.As insula cortex is important in emotional processing,we have examined whether the changes in synaptic plasticity in the insula cortex involved in stress resilience or vulnerability.METHODS Mice were divided into two groups:control and stress group.Stress group was treated by foot electric shock twice daily(0.8 mA,2 s,ten times in 1 min) in continuous two weeks.Then we used fear conditioning test to detect re-experiencing of traumatic experience,open field test to detect avoidance,pre-pulse inhibition experiment to detect hyper arousal.The changes of synaptic plasticity in the insular cortex were recorded by the multiple channels electrophysiology and whole cell patch.RESULTS According to the behavioral scores,it was divided into resilient and vulnerable group.In the fear conditioning test,the vulnerable group showed the significant freezing time decreased than that of the resilient group(Pinsular cortex.Compared with the resilient group,the LTP level was decreased(Pinsular cortex may be one of the neural mechanisms for the vulnerability to chronic stress.

Incidence evaluation of <i>Crypticerya multicicatrices</i>and <i>Maconellicoccus hirsutus</i>in Colombian Seaflower Biosphere Reserve

An evaluation of the incidence of Crypticerya multicicatrices and Maconellicoccus hirsutus in Colombian Seaflower Biosphere Reserve in San Andrés, Providence and Santa Catalina islands, from mid-August to mid-November 2012 was made. 38 locations were sampled, and 34% of sites tested show an incidence of C. multicicatrices oscillating between a range of 80%-100%, 24% with 40%-80% of incidence, 18% between 10%-40% and 24% with a range of 0%-10%. The occurrence of M. hirsutus fluctuated between 0.5% and 17%. There were seven genera of ants associated C. multicicatrices and 94 hosts of this agriculturally important insect and we found high incidence of associated sooty mold in C. multicicatrices hosts. Observations also contained some developmental stages of C. multicatrices.

Patients with temporal lobe epilepsy have inexplicable fear attack as the aura symptoms.However,the underlying neural mechanisms of seizure-modulated fear are not clarified.Recent studies identified stathmin as one of the key controlling molecules both learning and innate fear.Stathmin binds to tubulin,inhibits microtubule assembly and promotes microtubule catastrophes.Therefore we predict that stathmin plays a crucial role in associating epilepsy seizures with fear conditioning.Firstly we established pilocarpine model of epilepsy in rats,and then made the fear condition training with the epilepsy rats.The epileptic rats with fear conditioning（epilepsy+fear）had a much longer freezing time than each single stimulus.The increased freezing levels revealed a significant strengthen effect of epilepsy seizures on the learned fear of the tone-shock contextual.Then we compared the stathmin expression in the hippocampus,the amygdale,the insular cortex and the temporal lobe.We found that the dramatic change of stathmin expression happened in the insular and in the hippocampus but not in the amygdale.We compared the stathmin expression and the dendritic microtubule stability between fear and epilepsy rats.We found that epilepsy can strengthen the fear conditioning with increased expression of stathmin and decrease microtubule stability.Fear conditioning increased the expression of stathmin slightly while the epilepsy with fear conditioning increased it dramatically in the hippocampus,insular cortex and hypothalamus.The phosphorylated stathmin slightly increased in the epilepsy with fear conditioning.The increased expression of stathmin was contrary to the decrease of the stathmin MAP2 andα-tubulin in the epilepsy with fear conditioning in all three brain areas.The most significant change of the ratio of MAP2andα-tubulin/stathmin happened in the insular cortex and hippocampus.So we conclude that epilepsy can strengthen the fear conditioning with the increased stathmin and decreased microtubule stability,especially in the ins

Objective To compare the effects of intellicage and water maze test on effects of GABABR in insula on recognition memory of rats. Methods: Male Sprague-Dawley rats were randomly divided into control, sham, saline, baclofen（selective GABABR agonist） and CGP35348（GABABR antagonist） groups. Different testing procedures were employed using Intellicage and Morris water-maze test to detect changes in rat’s recognition memory. The expression of GABABR（GB1, GB2） in the insulars of rats was determined by immunofluorescence and Western Blotting at protein level.Results: the results of intellicage test showed that recognition of memory was impaired in position learning, punitive learning and punitive reversal learning by using baclofen and CGP 35348 compared with the control groups. In position reversal learning, there was no significant difference was found in the cognitive memory ability between the control groups with groups of CGP and baclofen. The results of Morris water-maze test showed that recognition of memory was impaired in the testing time. Data from immunofluorescence showed that GABABR（GB1, GB2） expression in the insular while the data of RT-PCR and Western blot showed that the relative expression of GB1 and GB2 was significantly increased in the baclofen group compared with the other control groups. The CGP35348 expression of GB1 and GB2 was significantly decreased. But there was no significant difference between the control groups. Conclusion（1） GABABR of insular plays an important role on the formation of recognition memory in rats.（2）Intellicage can add the lack of Morris water-maze test, and this behavioral system can provide more objective and accurate data.
