Background: Circular RNAs(circRNAs), a class of covalently closed ring structure RNA molecules, which involved in various biological processes by miRNA sponges,RNA protein binding or translation. circRNAs widely appeared in eukaryotes and some DNA viruses. However, it remains largely unexplored whether RNA virus including Bombyx mori Cypovirus(BmCPV) belonged to Reoviridae with segmented double-stranded RNA(dsRNA) genome could generate viral circRNAs(vcircRNAs)with ***: The BmCPV generated vcircRNAs were assessed by circRNAome sequencing. Reverse transcription PCR and northern blotting were used to confirm the existence of vcircRNAs. Immunofluorescence assay and western blotting were employed to evaluate the peptide translated by vcircRNAs. BmN cells overexpressing vcircRNA or treated with the peptide encoded by vcircRNA were used to evaluate the effect on the infection of BmCPV. Further, to explore the potential regulatory mechanisms, the ROS burst, nuclear translocation of p65 in NF-κB signaling pathway and immune genes expression were investigated in the BmN cells treated with the peptide coded by ***: We firstly screened 295 distinct vcircRNAs in the BmCPV infected silkworm midgut, which were generated from both sense and antisense strands of BmCPV dsRNA genome by a novel hyperbacksplicing. One vcircRNA named v-circS(5))165-1018(+) was derived from region 165-1018 nucleotide(nt) of sense strand of genomic dsRNA segment S5. The viral structural protein VP7 was decreased by overexpression of v-circS(5))165-1018(+) in BmN cells. Subsequently, a peptide(termed as v-circS(5))-P21) encoded by v-circS(5))165-1018(+) was predicted and confirmed. Furthermore, v-circS(5))-P21 was identified to promote the ROS burst,nuclear translocation of p65 and antiviral genes expression in vitro.
