Discrimination of toxic ingredient between raw and processed Pinellia ternata by UPLC/Q-TOF-MS/MS with principal component analysis and T-test

Objective: To investigate the toxicity difference between raw and processed Pinelliae Rhizoma(Banxia in Chinese, BX), the rhizoma of Pinellia ternata, from the view of chemical ***: Sixteen samples of raw and processed BX were prepared and analyzed by UPLC/Q-TOF-MS/*** discrimination(chemical marker) between the two group was investigated by principal component analysis(PCA) and T-test analysis. According to the accurate charge-to-mass ratio, MS/MS fragments, and comparison of corresponding data with the reference or database, the chemical markers were identified ***: Liquiritin, liquiritigenin, and lysophosphatidylcholine(LPC) were identified as the characteristic markers. The reducing of LPC in processed BX was one of the main reasons for detoxification because LPC could induce the inflammatory response;Liquiritin and liquiritigenin showed the anti-inflammatory effect and reduced liver injury, therefore the appearance of them in processed BX was an another reason for ***: An approach to explain the mechanisms of reducing the toxicity in medicinal plants by processing was proposed. Moreover, the chemical markers of toxicity could be used to differentiate the raw material from processed herbs for the quality control and safety application in clinical practice.

Using Mo, B-Fe alloy and Fe powders as raw materials, and adding C, Cr and Ni ingredients, respectively, or C, Cr and Ni mixed powders, ternary boride hard alloy clad materials was prepared on Q235 steel substrate by means of in-situ reaction and vacuum liquid phase sintering technology. The influence of alloy ingredients on the mechanical properties of ternary boride hard alloy clad materials was investigated. The results indicate that a mixture of 0.8% C, 5% Cr and 2% Ni ingredients gives a ternary boride hard alloy clad material with optimal mechanical properties, such as high transverse rupture strength, high hardness and good wear resistance.

This paper reports on the effects of organic ingredients in facilitating direct shoot regeneration from nodal explants of Hibiscus rosa-sinensis L. This paper also compares the sterilization conditions for 3 types of explants (node, internode, and shoot tip) harvested from an open field. The optimized sterilization conditions for the explants were 40% Clorox- 20 min exposure, 10% Clorox-15 min exposure, and 5% Clorox-40 min exposure for the node, internode and shoot tip, respectively. In the direct shoot regeneration using the nodal explants, we found MS medium containing 40 g/L sucrose, 0.3% (w/v) activated charcoal, and supplementations with myo-inositol, thiamine and nicotinic acid were suitable. The in vitro shoot survival rate was 30% with a mean leaf numbers of 2.68 produced, and a mean leaf length of 1.71 cm achieved after 5 weeks of culture on the modified medium.

Dang-gui is one of the traditional Chinese medicinal herbs. Analysis of its ingredients is very important to the study of its pharmacological mechanism. Research reports on this subject have appeared in literature since 1924. From Min-Dang, a species collected from Min District of Gansu Province and identified as Angelica sinensis (Oliv.) Diels, we have isolated and identified 53 constituents in its essential oil by capillary GC/MS and 34 compounds (including 16 aminoacids by aminoacid analyzer) in its non-vola-tile fraction, most of which have not been found in Min-Dang before. In present communication, the elucidation of the structure of a new ingredient arbitrarily named as angelicide will be reported briefly.

Eucommiae Folium(EF),a traditional Chinese medicine,has been used to treat secondary hypertension,including renal hypertension and salt-sensitive hypertension,as well as hypertension caused by thoracic aortic endothelial dysfunction,a high-fat diet,and oxidized low-density *** antihypertensive components of EF are divided into four categories:flavonoids,iridoids,lignans,and phenylpropanoids,such as chlorogenic acid,geniposide acid and pinoresinol *** regulates the occurrence and development of hypertension by regulating biological processes,such as inhibiting inflammation,regulating the nitric oxide synthase pathway,reducing oxidative stress levels,regulating endothelial vasoactive factors,and lowering blood ***,its molecular antihypertensive mechanisms are still unclear and require further *** this review,by consulting the relevant literature on the antihypertensive effects of EF and using network pharmacology,we summarized the active ingredients and pharmacological mechanisms of EF in the treatment of hypertension to clarify how EF is associated with secondary hypertension,the related components,and underlying *** results of the network pharmacology analysis indicated that EF treats hypertension through a multicomponent,multi-target and multi-pathway *** particular,we discussed the role of EF targets in the treatment of hypertension,including epithelial sodium channel,heat shock protein70,rhoassociated protein kinase 1,catalase,and superoxide *** relevant signal transduction pathways,the ras homolog family member A(RhoA)/Rho-associated protein kinase(ROCK)and nicotinamide adenine dinucleotide phosphate(NADPH)oxidase/eNOS/NO/Ca^(2+)pathways,are also discussed.

Objective:To explore the effect of AIBL on Oncomelania hupensis,the intermediate snail host of Schistosoma ***:The enzyme histochemical profiles of cholineslerase, cytochrome oxidase,lactate dehydrogenase,nitric oxide synthase,and succinate dehydrogenase in the soft tissues of Oncomelania hupensis,the intermediate host snail of Schistosoma japonicum, were analyzed before and after treatment with the active ingredient of Buddleia lindleyana(AIBL), a potent and safe plant ***:Treatment with AIBL induced a notable decrease in the activities of the five enzymes(P<0.01).Conclusions:The results indicate that AIBL impairs the activities of the enzymes,thereby influencing the transfer of neurotransmitter and energy supply in Oncomelania hupensis and ultimately harming their various physiological functions, which are considered to cause death of the species.

A comparative UV-HPLC analysis of ten brands of ibuprofen tablets

Objective: To investigate the pharmaceutical equivalence of ten brands of ibuprofen tablets(400 mg) purchased from pharmacies in Benin City, ***: The drug samples were subjected to uniformity of weight, crushing strength,friability, melting point, disintegration and dissolution tests following acceptable and official protocols. The ibuprofen content was determined using UV and high performance liquid chromatography ***: Crushing strength values of the drug samples ranged between 6 and 16 kp while the disintegration times were between 7.43 and 10.40 min(for uncoated tablets) and3.25–37.32 min(for coated tablets). Friability values were less than 1% and the melting points of recrystallized ibuprofen from the samples ranged from 73.5 to 76.0 C. The amount of ibuprofen released within 1 h ranged between 18% and 102% and two brands failed the content of active ingredient in the UV method of assay while all the brands passed the test using ***: Ibuprofen(400 mg) tablets marketed in Benin City, Nigeria vary in pharmaceutical quality.
