Optimization of LPS-Induced Inflammation Model and Its Feasibility as a Fast Screening Model for Cosmetics

Objectives: The existing inflammatory models are concentrated in relatively complex medical fields, and most of them use a single type of cell, and the induction conditions are not uniform, so the current LPS-induced inflammation model is less conducive to the study of skin inflammation. The aim of this research is to enhance the existing LPS-induced inflammation model and establish a skin inflammation model that is suitable for the swift screening of anti-inflammatory agents in the cosmetics industry. Methods: LPS was used to induce inflammatory responses in KC and THP-1 cells. Enzyme-linked immunosorbent assay (ELISA) was employed to assess the levels of IL-1α, IL-8, and TNF-α in the two cell types, while the DCFH-DA probe was utilized to label the levels of reactive oxygen species (ROS) in both cell types. Results: In KC cells, 10 μg/mL of LPS induced a significant upregulation of IL-8 but did not result in elevated expression of IL-1α. However, at 100 μg/mL of LPS, both IL-8 and IL-1α were highly expressed in KC cells. LPS concentrations ranging from 0.01 to 100 μg/mL failed to stimulate TNF-α production in KC cells but induced a gradient increase in ROS levels. In THP-1 cells, LPS concentrations from 0.01 to 100 μg/mL did not induce IL-1α production but significantly elevated IL-8 and led to a gradient increase in TNF-α and ROS. After treatment with 100 μg/mL of LPS, the cosmetic ingredient Rucika KGM mitigated the elevated levels of IL-1α, IL-8, and ROS in LPS-induced KC cells and IL-8 and ROS in THP-1 cells. Conclusion: This study has successfully developed an application-oriented model suitable for investigating skin inflammation, enabling the rapid and comprehensive screening of cosmetic ingredients with anti-inflammatory activity. .

Objective:To investigate the effects of Pulmicort Respules,Ventolin combined with methylprednisolone in the adjuvant treatment of bronchiolitis in ***:A total of 100 children with bronchiolitis in our hospital from March 2018 to March 2020 were selected and divided into 2 groups with 50 cases in each group by using random number *** groups received conventional *** on this,the control group was given Pulmicort Respules and Ventolin,and the observation group was given methylprednisolone in combination with the conventional regimens on the basis of the control group treatment,the course of treatment was 5 *** levels of inflammatory factors[tumor necrosis factor-a(TNF-a),interleukin 6(IL-6)]and the time to symptom disappearance before and after treatment were compared and analyzed between the two ***:After treatment,the levels of serum TNF-a and IL-6 in the two groups decreased,and the observation group was lower than the control group,the difference was statistically significant(P<0.05);the disappearance of pulmonary moist rales,lung wheezing,cough and wheezing in the observation group were all earlier than the control group,the difference was statistically significant(P<0.05).Conclusion:Pulmicort Respules,Ventolin combined with methylprednisolone is effective in adjuvant treatment of bronchiolitis in children,which can reduce inflammation and promote the recovery of children.

[Objectives]To study the anti-inflammation and anti-swelling effect of Caulophyllum robustum Maxim( CRM) extract on secondary lesions of adjuvant arthritis( AA),associated inflammatory factors of AA rats and mice immune function. [Methods]Rats were divided into high,middle and low dose CRM extract groups( 69. 23,34. 61,17. 31 mg/kg),methotrexate group( 0. 68 mg/kg),triptolide group( 7. 88 mg/kg),model group and normal control group. The AA rat model was induced by intradermal injection of Freund's complete adjuvant( FCA). After the AA model was established successfully,the paw swelling of AA rats was observed and recorded. The levels of interleukin-1β( IL-1β),interleukin-4( IL-4),interleukin-10( IL-10),Tumor Necrosis Factor-α( TNF-α),interleukin 17( IL-17),interferon( IFN-γ),granulocyte colony-stimulating factor( G-CSF) and transforming growth factor( TGF-β) in the serum of AA rats were detected by enzyme-linked immunosorbent assay( ELISA). The spleen was removed and weighed for detecting the drug influence on the immune *** were observed by gavage administration at different dosages of 25 mg/kg,50 mg/kg,100 mg/kg for seven days,adopting carbon granules clearance test,delayed-type hypersensitivity induced by dinitrofluorobenzene( DNFB) coupled with flow cytometry evaluating lymphocytes( CD3^+,CD4^+,CD8^+) activation and serolysin test. [Results] Compared with the model group,the joint swelling,spleen index and inflammatory cytokines( IL-1β,IL-4,IL-10,TNF-α,IL-17,IFN-γ,G-CSF,TGF-β) were reduced significantly( P < 0. 05,P < 0. 01). And the activity of CRM was significantly superior to methotrexate and triptergium. Moreover,the results indicated that different dosages of 25 mg/kg,50 mg/kg,100 mg/kg of CRM could significantly reduce the carbon granules clearance index( K) and phagocytic index( a),inhibit the phagocytosis of mononuclear macrophages,reduce the mice swelling extent and inhibit DTH in mice induced by DNFB,decrease the quantity of serum hemolysin.

Objective:To investigate the efficacy of montelukast sodium in the treatment of lung cancer patients with pulmonary ***:A total of 330 patients diagnosed with lung cancer and pulmonary infection,who were admitted to the First Affiliated Hospital of Xi’an Medical University from 2020 to 2022,were selected as research *** were randomly divided into two groups:a control group receiving conventional treatment and an observation group receiving conventional treatment combined with montelukast *** group consisted of 165 *** time required for clinical symptom improvement,the resolution of lung infection signs,and the levels of serum inflammatory factors before and after treatment were compared between the two ***:The observation group exhibited significantly faster improvement in clinical symptoms compared to the control group(Pinflammatory factors compared to the control group(P<0.001).Conclusion:Montelukast sodium demonstrates efficacy in the treatment of patients with lung cancer complicated by pulmonary *** findings suggest its potential for further verification and clinical application.

Study on the Regulation of ROS by Acupuncture and Moxibustion to Relieve Inflammation in the Treatment of Sciatica

Objective: To observe the analgesic effect of acupuncture and moxibustion on chronic constriction injury (CCI) pain model rats, and to investigate the mechanism of constriction constriction injury. Methods: 32 SPF SD rats were divided into Control Group, Sham Surgery Group, Model Group and Acupuncture and moxibustion Group, with 8 rats in each group. The Model Group, Acupuncture and moxibustion Group and sciatic nerve ligation were used to establish the CCI pain rat model. The sham operation group only separated the nerve, and the control group did not do any treatment. After modeling, the Acupuncture and moxibustion Group was given acupuncture treatment, while the control group and the Model Group did not do any intervention. 7 days for 1 course of treatment, continuous treatment for 3 courses of sampling and detection. Paw withdrawal mechanical threshold (PWMT) and Paw withdrawal thermal latency (PWTL) were observed. Biochemical detection of reactive oxygen species (ROS), malondialdehyde (MDA), Super Oxide Dismutase (SOD), glutathione (GSH);Serum, tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β) and interleukin-6 (IL-6) levels were detected by ELISA. Results: Compared with the control group, PWMT and PWTL in the Model Group were decreased (P α, IL-1β and IL-6 levels or contents were increased (P α, IL-1β and IL-6 levels or contents were decreased (P Conclusion: Acupuncture and moxibustion can reduce the ROS level, enhance the antioxidant capacity of the body, reduce the damage of sciatic nerve caused by inflammation, and relieve sciatic pain.

Liver fibrosis is an abnormal proliferation pathologic process of intrahepatic fibrous connective tissue that occurs after liver cells have been necrotized and stimulated by inflammatory factors. It is called fibrosis when the pathological change is mild, and liver cirrhosis when the change becomes so severe as to reconstruct the liver lobuli to form pseudolobuli and nodule(1). Liver fibrosis is an important pathological characteristic of chronic hepatopathy and the chief intermediate link to further develop of liver cirrhosis. No ideal remedy for treatment of chronic hepatitic cirrhosis has been found so far. Although some drugs, such as colchicine and penicillamine, had been reported to have the effect of fibrosis inhibition, their clinical application is still limited for the rather severe toxic-side effects. Certain progress have been made from the clinical and experimental studies on anti-fibrosis treatment by traditional Chinese medicine (TCM) carried out widely in China in recent ten years. And here is a general review of the drugs used.
