Cloning Successive Generations of industrial hemp (Cannabis sativa) to Assess Cannabinoid Profiles

industrial hemp (Cannabis sativa) has made a remarkable impact worldwide due to the plant’s beneficial properties and versatile use. Vegetative cuttings (clones) of C. sativa are the preferred propagation technique to be the most effective in retaining the same genetic information and reducing hybridization and mutations. The objective of this project was to assess cannabinoid profile concentrations of successively cloned generations of 5 varieties: Cherry, Cherry Blossom, Cherry × Workhorse, Sour Space Candy, and The Wife. This research project focused on the idea that every cloned plant contains the exact same genetic information and, therefore, should have the same metabolic profile of cannabinoids through all the successive generations grown, which is shown to be true. Plants were cloned for multiple generations using stem cuttings and a commercial cloner. As plants matured, they were set in environmental conditions to stimulate flowering and buds were harvested and analyzed for cannabinoid contents using HPLC. Several generations of each variety were successfully cloned. As many as 17 different cannabinoids were analyzed and the results of this study show that there is not a significant difference in cannabinoids over successive generations, showing no major trends.

High salt-alkaline is one of the adverse environmental factors effecting growth and development of industrial hemp, identifying the genes that control this trait will provide important insights for industrial hemp breeding. Up to now, the molecular mechanism of saline-alkaline tolerance of industrial hemp remains unclear. In this study, RNA-seq was performed in industrial hemp under the treatment of salt(SA), alkaline(AL) and distilled water(CK) to understand the molecular mechanism of saline-alkaline tolerance in industrial hemp. A total of 450 million high-quality clean reads and mean length of 1037 bp were obtained, and 190,359 unigenes were generated for the transcriptome. Among the assembled unigenes, 1060 and 969 differentially expressed genes(DEGs) were identified in the salt and alkaline treatment, respectively. 12 of these differential expressed genes were random check for the real-time PCR, and they were all identical with the consequence of the RNA-seq. Gene annotation and pathway enrichment analysis indicated that Plant hormone signal transduction, Fructose and mannose metabolism, Flavonoid biosynthesis, Glycolysis/gluconeogenesis, Ascorbate and aldarate metabolism are significantly affected by the salt stress. The potentially important roles for underappreciated gene families, including several groups of transporters(e.g. Calcium-binding EF-hand, Aquaporin NIP5-1/NIP6-1, AQPAnG-like), transcription factors(e.g. members of WRKY, NAC) and other proteins(e.g. Cytochrome P450, Heat shock protein-like) suggested industrial hemp may be adapted to the salt-alkaline environment through these pathways. Our result may provides the important insights for the mechanism of salinity tolerance in industrial hemp and constituted a database for future studies.
