The Collaborative Cross mouse genetic reference population designed for dissecting complex traits

Complex traits are multifactorial traits controlled by polygenic host *** trait-related phenotypic characteristics and performance including body weight,blood chemistry,immune cell profiles,as well host susceptibility to infectious and chronic *** recent years,tremendous efforts were invested aiming to map the host genetic factors attribute to these traits and subsequently clone the gene/s underlying these *** parallel to human studies,a number of mouse models and approaches were developed aimed to enhance the mapping process and the gene *** include of using resources such as F2,backcross,advanced intercross lines,outbred populations,consomic,congenic and recombinant inbred lines(RIL).The constraints of these approaches were the limited resolution mapping of genomic regions of the quantitative trait loci(QTL)associated with the trait of interests,and the limited genetic diversity observed in the parental *** overcome these limitations,a new genetically highly diverse recombinant inbred lines of mouse population was established,namely the Collaborative Cross(CC),created from full reciprocal mating of 8 divergent strains of mice:A/J,C57BL/6J,129S1/SvI mJ,NOD/LtJ,NZO/HiL tJ,CAST/Ei,PWK/PhJ,and WSB/*** intercrossing these eight founders to generate the different CC lines,the genetic makeup of the newly developed resource is completely different from the eight parental lines,and will show heterosis,which subsequently will response differently comparing with their original ***,our results suggest that it is not essential to defining the phenotypic response of the eight parental lines,prior of assessing the CC lines,because it is believed that genetic interaction of the new genetic makeup of the new lines will reveal new phenotypic response,which completely different from the parental *** this report,we present to the community the power of the CC for dissecting variety of complex traits including host susceptib

Dark Response of Seedlings Evaluated by Chlorophyll Concentration in Maize Natural Population

Chlorophyll, one of the major chloroplast components for photosynthesis, has a positive relationship with the photosynthetic rate. The chlorophyll content is an important assessment parameter in agronomy and plant biology research. This study was conducted to evaluate the natural variation in the chlorophyll content and to determine the differential response of the chlorophyll concentration to dark treatment in a natural population containing 139 maize inbreds. A five-fold higher chlorophyll concentration was measured in the light compared with the dark. Meanwhile, the wide variation in the chlorophyll concentration showed the differential response of the natural maize population to dark. Finally, we identified some inbreds that were highly sensitive to the dark with more than 70% difference between the light and dark treatment, such as Dan598, Zheng29, Zheng35, DH29, and R08, as well as some inbreds that had lower sensitivity to the dark, with less than 35% difference in the chlorophyll content between the light and dark treatment, such as Chuan48-2, 4F1, 303WX, 9642, and LY042.

Identification of Potential F<sub>1</sub>Hybrids in Maize Responsive to Water Deficient Condition

The assessment of heterotic F1 combinations is a basic requisite for hybrid seed development. A set of 30 F1 hybrids along with their parental inbred lines were evaluated under both normal and water deficit conditions for various physiological and agronomic traits. Highly significant mean squares due to general combining ability, specific combining ability and reciprocal effect were observed for all traits under both water regimes. Components of variation exhibited greater estimates for GCA variance (б2g) than SCA variance (б2s) for majority of the traits under both normal and stress conditions depicting the predominant role of additive genetic component. inbred lines NCIL-20-20, D-157 and OH-8 were found to be the best general combiner on the basis of performance regarding grain yield per plant under water deficit condition. The F1 combinations namely, NCIL-20-20 × D-109, NCIL-20-20 × OH-8 and D-157 × NCIL-20-20 were out-performers based on yield and yield attributes under water deficit conditions. On the basis of our results, we recommend these hybrids for further exploitation to assess their potential for commercial cultivation under water deficit condition.

Tolerance of Maize(Zea mays L.) to Chlorsulfuron

Analyses of Protein and Oil of Maize Offsprings Plants Introducing Soybean DNA

[Objective] The experiment was aimed to explore character variation between different families after DNA introduction and select variant plants with good stability. [Method] The method of pollen-tube-pathway was used to introduce total DNA of soybean into normal maize inbred line 7313 for selecting generation by generation. When field characters of maize, grain colors, grain traits and panicle axis colors were stable, the crude protein, gliadin, glutelin and oil content of grains which were selected from variant strains were detected and compared. [Result] The grain crude protein, gliadin, glutelin and oil content of line 26h-4-3 were significantly different from these of control treatment. The increments of D3 and D4 generation were 10.34% and 26.70%, 6.58% and 6.28%, 15.09% and 70.34%, 55.82% and 51.52% respectively. All indexes of line 26h-3-1 were also higher than these of control treatment and the increments of D3 and D4 generation were 5.67% and 21.63%,1.91% and 2.31%, 10.85% and 62.27%,22.49% and 9.67%. [Conclusion] The crude protein, gliadin, glutelin and oil content of variant line 26h-4-3 and 26h-3-1 were stable, so variant line 26h-4-3 and 26h-3-1 were excellent variant strains which satisfied the requirement of high protein breeding.

Genetic Quality Analysis of Xiphophorus helleri RR-B Strain Using 18 Microsatellite Loci

[Objective] The study aimed to further understand the genetic quality of Xiphophorus helleri RR-B strain and estimate the feasibility of microsatellite markers in genetic monitoring of aquatic laboratory animals.[Method] Eighteen microsatellite loci which had polymorphism in wild X. helleri were used for genetic quality analysis of RR-B strain (red eyes and red body) by PCR. [Result] All these 18 microsatellite loci displayed single allelic gene band in 30 individuals of X. helleri RR-B strain and the average homozygote rate reached 100%. [Conclusion] These loci could be used as markers in genetic monitoring of X. helleri RR-B strain, which had obtained fairly high genetic homozygosity by inbreeding for more than 20 generations. This study provided basic data for establishing the molecular genetic monitoring standard of aquatic laboratory animals.
