A Nested Case-Control Study to Explore the Association between immunoglobulin g N-glycans and Ischemic Stroke

Objective This study prospectively investigates the association between immunoglobulin g(Igg)N-glycan traits and ischemic stroke(IS) *** A nested case-control study was conducted in the China suboptimal health cohort study,which recruited 4,313 individuals in 2013–2014. Cases were identified as patients diagnosed with IS, and controls were 1:1 matched by age and sex with cases. In directly measured gPs, gP4, gP9, gP21, gP22, gP23, and gP24 were associated with the risk of IS in men after adjusting for age, waist and hip circumference,obesity, diabetes, hypertension, and dyslipidemia. Derived glycan traits representing decreased galactosylation and sialylation were associated with IS in men(FBg2S2/(FBg2 + FBg2S1 + FBg2S2): odds ratio(OR) = 0.92, 95% confidence interval(CI): 0.87–0.97;g1n: OR = 0.74, 95% CI: 0.63–0.87;g0n: OR =1.12, 95% CI: 1.03–1.22). Interaction between gold Nanoparticles and Bovine Serum Albumin or Sheep Antirabbit immunoglobulin g

The interaction between gold nanoparticles and proteins such as bovine serum albumin and immunoglobulin g under the condition of different pH values was studied based on the measurement of zeta potential and fluorescence quenching of the proteins before and after proteins were bound with gold nanoparticles. Aggregations were found in gold colloid aqueous solution after addition of proteins by TEM characterization and UV-Vis spectroscopy deterruination. The results showed that the values of zeta potential were quite different, the binding constant Kb and stoichiometry n were slightly increased with the increase of pH value. In conclusion, two factors could affect markedly the interaction between gold nanoparticles and proteins, that is, surface charge and the coordination effect between gold nanoparticles and indole group of the tryptophan residue of proteins.

Influence of birth order, birth weight, colostrum and serum immunoglobulin g on neonatal piglet survival

Background： Intake of colostrum after birth is essential to stimulate intestinal growth and function, and to provide systemic immunological protection via absorption of immunoglobulin g （Igg）. The birth order and weight of 745 piglets （from 75 litters） were recorded during a one-week period of farrowing. Only pigs weighing greater than 0.68 kg birth weight were chosen for the trial. Sow colostrum was collected during parturition, and piglets were bled between 48 and 72 hours post-birth. Piglet serum Igg and colostral Igg concentrations were determined by radial immunodiffusion. Results： Sow parity had a significant （P 〈 0.001） effect on sow colostral Igg concentration, being 5% higher in multiparous females. Sow colostral Igg concentration explained 6% and piglet birth order accounted for another 4% of the variation observed in piglet serum Igg concentration （P 〈 0.05）; however, birth weight had no detectable effect. Piglet serum Igg concentration had both a linear （P 〈 0.05） and quadratic effect （P 〈 0.05） on % survival. Piglets with 1,000 mg/dl serum Igg or less （n=24） had a 67% survival; whereas, piglets with Igg concentrations between 2250 to 2500 mg/dl （n=247） had a 91% survival. Birth order had no detectable effect on survival, but birth weight had a positive linear effect （P 〈 0.05）. Piglets weighing 0.9 kg （n = 107） at birth had a 68% survival rate, and those weighing 1.6 kg （n = 158） had an 89% survival. Conclusion： We found that the combination of sow colostrum Igg concentration and birth order can account for 10% of the variation of piglet serum Igg concentration and that piglets with less than 1,000 mg/dl Igg serum concentration and weight of 0.9 kg at birth had low survival rate when compared to their larger siblings. The effective management of colostrum uptake in neonatal piglets in the first 24 hrs post-birth may potentially improve survival from birth to weaning.

Neuro-immune crosstalk connects the nervous and immune systems. Pathogenesis of nervous system diseases are related to inflammatory/immunological responses. Traditionally, immunoglobulin g （Igg） molecules are only synthesized by mature B lymphocytes. Recently, mouse hippocampus neurons were shown to produce Igg, although its functions are unknown. This study observed the expression and distribution of Igg-positive cells in the hippocampus and substantia nigra of rats, to elucidate a relationship between pathogenesis of Alzheimer’s disease, Parkinson’s disease and neural inflammatory responses. Igg immunoreactivity was visualized in plasma cells or plasmablasts from rat spleen tissues, suggesting the polyclonal antibody used in this experiment was Igg-specific. Igg protein was detected in the cytoplasm and processes of cells in many brain regions including the hippocampus and substantia nigra compacta, regions associated with Alzheimer’s disease and Parkinson’s disease. Ratios of Igg-positive cells/total cells in the hippocampal CA1, dentate gyrus and substantia nigra were 72.6%, 73.3% and 75.2%, respectively. By morphology, Igg-positive cells included neurons and neuroglia. To determine exact cell types, immunohistochemical staining with specific neuron and neuroglia markers is required. In addition, that Igg molecules are synthesized and expressed in cells of the nervous system instead of being taken up from plasma and transported across blood brain barrier or from the extracellular fluid must be confirmed. Lewy bodies are strongly immunolabeled with Igg in Parkinson’s disease brain, but it is unknown whether Igg was synthesized by nigral neurons. In this study, we found that some Igg-positive cells were similar to microglia morphologically. It has been reported that microglia express Igg receptors. immunoglobulin g(Igg)is the most abundant plasma glycoprotein and a prominent humoral immune *** composition affects the affinity of Igg to ligands and consequent immune *** modification of Igg N-glycosylation is considered to be one of the various mechanisms by which sex hormones modulate the immune *** the menstrual cycle is the central sex hormonerelated physiological process in most women of reproductive age,Igg N-glycosylation dynamics during the menstrual cycle have not yet been *** fill this gap,we profiled the plasma Igg Nglycans of 70 healthy premenopausal women at 12 time points during their menstrual cycles(every 7 days for 3 months)using hydrophilic interaction ultra-performance liquid chromatography(HILIC-UPLC).We observed cyclic periodic changes in the N-glycosylation of Igg in association with the menstrual cycle phase and sex hormone concentration in *** the integrated cohort level,the modeled average menstrual cycle effect on the abundance of Igg N-glycosylation traits was low for each trait,with the highest being 1.1%for agalactosylated ***,intrapersonal changes were relatively high in some cases;for example,the largest difference between the minimum and maximum values during the menstrual cycle was up to 21%for sialylated *** all measurements,the menstrual cycle phase could explain up to 0.72%of the variation in the abundance of a single Igg glycosylation trait of *** contrast,up to 99%of the variation in the abundance of digalactosylation could be attributed to interpersonal differences in Igg *** conclusion,the average extent of changes in the Igg N-glycopattern that occur during the menstrual cycle is small;thus,the Igg N-glycoprofiling of women in large sample-size studies can be performed regardless of menstrual cycle phase.

免疫球蛋白g(immunoglobulin g,Igg)的N-糖基化在炎症性疾病的发展中起着重要作用。本研究旨在评价Igg N-糖基在消化道癌症亚型中的诊断效能。从中国医学科学院肿瘤医院招募749名消化道癌症患者,包括食管癌(esophageal cancer,EC)、胃癌(gastric cancer,gC)、结直肠癌(colorectal cancer,CRC)和胰腺癌(pancreatic cancer,PC)患者。采用亲水交互高效液相色谱-超高效液相色谱(hydrophilic interaction liquid chromatography using ultra-performance liquid chromatography,HILIC-UPLC)分析血浆中Igg的N-糖基构成。采用Bio-Plex悬液芯片系统检测方法(Bio-Rad)进行炎症因子检测。采用典型相关分析(canonical correlation analysis,CCA)探索糖基和炎症因子之间的相关性。采用LASSO回归和logistic回归模型,基于检测到的糖基谱建立可用于区分胃肠癌症患者和健康人群诊断模型。与健康对照组相比,EC、gC、CRC和PC患者的唾液酸化和半乳糖基化水平降低,而二等分乙酰葡萄糖胺基化水平在消化道癌症患者中升高。此外,只有胰腺癌患者具有低水平的岩藻糖基化。消化道癌症组的IL-1β、IL-31和sCD40L水平均高于对照组。Igg N-糖基的组成与炎症因子相关(r=0.556)。基于糖基的模型表现出良好的诊断效能,EC、gC、CRC和PC的受试者工作特征曲线下面积(AUC)分别为0.972、0.871、0.867和0.907。这些研究结果表明,Igg N-糖基在调节消化道肿瘤的发病机制中发挥了重要作用。血清Igg N-糖基可以作为潜在的非侵入性辅助消化道癌症临床诊断的方法。
