Functional study of hyperpolarization activated channel (I_h) in Drosophila behavior

Hyperpolarization-activated, cyclic nucleotide-gated and cation-nonselective ion channels （lh channels, or HCN channels） are known to play important roles in mammals. Their physiological functions in invertebrate remain largely unclear. Here, we re- port our studies with lh channel in Drosophila melanogaster. Drosophila lh channel mutants are found with several defects by behavioral analyses. Their lifespan is reduced, and their chemical sensitivity is shifted. In addition, their length of sleep at light-dark condition is mildly reduced. We generated transgenic flies of lh promoter-driven Gal4 and examined its expression pattern in both larvae and adult flies. Our results suggest that ih channel may play diverse roles in Drosophila and provide a basis to further expand our understanding of Drosophila ih channel function in vivo.

High temporal resolution of vision relies on the rapid kinetics of the photoresponse in the light-sensing photoreceptor *** is well known that the activation/deactivation of photoreceptor largely depends on the visual transduction cascades within ***,how photoreceptor neurons achieve the stable resting membrane potential is not *** we showed that ih channels play an essential role in regulating photoreceptor cell activity through retinal- lamina *** performed an ERG-based mutagenesis screen and isolated a mutant fly which exhibits spontaneous photoreceptor neurons firing in *** spontaneous activation of photoreceptor neuron can be suppressed by either blocking phototransduction cascades or the neurotransmitter release from photoreceptor *** immunostaining,we indentified that ih channels expressed in LI,L2 lamina neurons and retinal base glia *** studies indicated that the retina-lamina microcircuits are involved in the regulation of photoreceptor activity.

Objective Spontaneous oscillations in the hippocampus are believed to play important roles in hippocampus-dependent cognition and learning and *** studies in hippocampal slices indicate that these oscillations are mediated by the activity of intemeurons in the hippocampus,particularly via GABAA receptors. This issue remains to be further *** In this study,we developed a new system of in vitro wholecell patch-clamp recordings in whole-hippocampus *** In such preparation,robust spontaneous oscillations(from 0.8 Hz to 2 Hz)were observed in both pyramidal cells and intemeurons in the absent of any *** whole-cell recordings further revealed that the oscillations were synchronized in CA1, CA3,and across CA1-CA3 *** the presence of bicuculline-a GABA receptor blocker-in bath solution,the frequency of spontaneous oscillations was remarkably reduced to 0.1-0.2 Hz,which was found in the pyramidal cells and intemeurons of both CA1 and CA3 ***,extracellular blockade of GABA receptors by SCH50911 didn’t alter the frequency of the *** treatment of ZD7288,which blocks hyperpolarization-activated (I)channels,also slowed down the hippocampal oscillations to .4 *** Our observations from whole-hippocampus formation revealed the important roles of GABA receptors,probably coordinating with the activity of I channels,in mediating the self-generated oscillations in the hippocampus in the absence of extrahippocampus inputs.
