Life science has a need for detection methods that are label-free and real-time. In this paper, we have selected staphylococcal protein A （SPA） and swine immunoglobulin G （igg）, and monitor the bindings between SPA and swine igg with different concentrations, as well as the dissociations of SPA-swine igg complex in different pH values of phosphate buffer by oblique-incidence reflectivity difference （OIRD） in a label-free and real-time fashion. We obtain the ON and OFF reaction dynamic curves corresponding to the bindings and dissociations of SPA and swine igg. Through our analysis of the experimental results, we have been able to obtain the damping coefficients and the dissociation time of SPA and swine igg for different pH values of the phosphate buffer. The results prove that the OIRD technique is a competing method for monitoring the dynamic processes of biomolecule interaction and achieving the quantitative information of reaction kinetics.

Nanogold particles of 10 nm were used to label goat anti-human igg (Gigg) to obtain nanogold-labeled Gigg (AuGigg). In a citrate-HCl buffer solution of pH 2.27,AuGigg showed a strong catalytic effect on the reaction between HAuCl4 and NH2OH to form big gold particles that exhibited a resonance scatter-ing (RS) peak at 796 nm. Under the chosen conditions,AuGigg combined with igg to form immuno-complex AuGigg-igg that can be removed by centrifuging at 16000 r/min. AuGigg in the centrifuging solution also showed catalytic effect on the reaction. On those grounds,an immunonanogold catalytic RS assay for igg was designed. With addition of igg,the amount of AuGigg in the centrifuging solution decreased; the RS intensity at 796 nm (I796 nm) decreased linearly. The decreased intensity ΔI796 nm was linear with respect to the igg concentration in the range of 0.08-16.0 ng·mL-1 with a detection limit of 0.02 ng·mL-1. This assay was applied to analysis of igg in sera with satisfactory sensitivity,selectivity and rapidity.

Immunoglobulin G4-related sialadenitis(igg4-RS)is an immune-mediated fibro-inflammatory disease and the pathogenesis is still not fully *** aim of this study was to explore the role and mechanism of interleukin-13(IL-13)in the cellular senescence during the progress of *** found that the expression of IL-13 and IL-13 receptorα1(IL-13Rα1)as well as the number of senescent cells were significantly higher in the submandibular glands(SMGs)of igg4-RS ***-13 directly induced senescence as shown by the elevated activity of senescence-associatedβ-galactosidase(SA-β-gal),the decreased cell proliferation,and the upregulation of senescence markers(p53 and p16)and senescence-associated secretory phenotype(SASP)factors(IL-1βand IL-6)in SMG-C6 ***,IL-13 increased the level of phosphorylated signal transducer and activator of transcription 6(p-STAT6)and mitochondrial-reactive oxygen species(mt ROS),while decreased the mitochondrial membrane potential,ATP level,and the expression and activity of superoxide dismutase 2(SOD2).Notably,the IL-13-induced cellular senescence and mitochondrial dysfunction could be inhibited by pretreatment with either STAT6 inhibitor AS1517499 or mitochondria-targeted ROS scavenger Mito ***,IL-13 increased the interaction between p-STAT6 and c AMP-response element binding protein(CREB)-binding protein(CBP)and decreased the transcriptional activity of CREB on *** together,our findings revealed a critical role of IL-13 in the induction of salivary gland epithelial cell senescence through the elevated mitochondrial oxidative stress in a STAT6–CREB–SOD2-dependent pathway in igg4-RS.

Zika virus(ZIKV) causes rash, moderate fever, conjunctivitis, and arthralgia, and has serious connection with neurological complications;therefore, it is a major threat to public health. A rapid and supersensitive method for detecting anti-ZIKV antibodies in humans and animals is thus urgently required. Here, we report an NS1-based luciferase immunosorbent assay(LISA), developed to detect ZIKV-specific igg. Fusion proteins including a reporter Nano-luciferase(NLuc) and various fragments of ZIKV NS1 protein were expressed in 293 T cells. LISA was performed using the above cell lysates containing the expressed fusion proteins. Sample panels of humans and animals infected with ZIKV were examined for sensitivity of LISA, relative to those of ZIKV RT-PCR, commercial NS1-based ELISA, and micro-neutralization(MN) *** and potential cross-reactivity were also evaluated using various convalescent serum samples derived from patients infected with dengue virus(DENV), Japanese encephalitis virus(JEV), and hepatitis C virus(HCV). Results indicated the optimal antigenic domain for anti-ZIKV igg detection was located within 172–352 amino acids(aa) of ZIKV NS1 protein. NS1-based LISA performs better than commercial ELISA in anti-ZIKV Ig G detection. LISA was shown to be at least fourfold more sensitive than commercial ELISA, and could detect anti-ZIKV Ig G in various animal hosts without the need of species-specific labeled antibody. This novel assay is potentially useful for the rapid and sensitive detection of anti-ZIKV igg in human and animal samples.

A kind of 9 nm gold nanoparticles was prepared with the trisodium citrate and used to label goat anti-human igg to obtain an igg immunoresonance scattering spectral probe. In pH 5.8 buffer solution and in the presence of polyethylene glycol （PEG）, the immune reaction between gold-labeled goat anti-human igg and igg took place, and the resonance scattering intensity at 580 nm （I580nm） was enhanced greatly. The enhanced intensity AIRS is pro- portional to the igg concentration from 1.3 to 1.5 X 10^3 ***^-1, with a detection limit of 0.78 *** ^-1. This assay showed high sensitivity and good selectivity for quantitative determination of igg in human serum, with satisfactory results.

AIM:To indirectly determine if tissue transglutaminase(tTG)-specific T cells play a crucial role in the propagation of celiac ***:Anti-deamidated gliadin peptide(DGP) and anti-tTG IgA and igg were measured in the sera of celiac patients(both untreated and treated).The correlations were determined by Spearman's rank correlation ***:In celiac patients,we found a very significant correlation between the production of DGP IgA and igg(r = 0.75),indicating a simultaneous and ongoing production of these two isotypes reminiscent of oral vaccination ***,there was far less association between the production of tTG IgA and tTG igg in celiac patients(r = 0.52).While tTG IgA was significantly correlated with DGP IgA(r = 0.80) and DGP igg(r = 0.67),there was a weak correlation between production of anti-tTG igg and the production of anti-DGP IgA(r = 0.38) and anti-DGP igg(r = 0.43).CONCLUSION:These data demonstrate that the production of anti-tTG IgA is directly correlated to the production of anti-DGP igg and IgA,whereas anti-tTG igg is only weakly *** result therefore supports the hapten-carrier theory that in well-established celiac patients anti-tTG IgA is produced by a set of B cells that are reacting against the complex of tTG-DGP in the absence of a tTG-specific T cell.

In a previous paper, 4 hybridoma cell lines (1B3, 2D5, 2A4 and 2B6) secreting antibodiesagainst heavy chain of hmnan igg were reported. In this paper a more detailed study of theseMcAbs was introduced. First of all they were studied with double immunodiffusion test inagarose and it was demonstrated that they were reactive against 3 distinct antigenic deter-minants of human igg. This result was also confirmed by ELISA competition test. TheseMeAbs were further analysed for human igg subclass proteins and it was found that 1B3McAb gave positive reaction with all subclasses of igg while the other 3 possessed confinedsubclass reactivity, i.e. McAb 2A4 did not react with igg3 (ealled non-igg3 McAb), andboth McAbs 2D5 and 2B6 did not react with igg4 (called non-igg4 McAb). BACKGROUND In paediatric patients with complicated nephrotic syndrome(NS), rituximab(RTX) administration can induce persistent igg hypogammaglobulinemia among subjects showing low basal immunoglobulin G(igg) *** To evaluate the effect of RTX on igg levels and infections in patients with complicated NS and normal basal igg *** We consecutively enrolled all patients with complicated NS and normal basal igg levels undergoing the first RTX infusion from January 2008 to January 2016. Basal igg levels were dosed after 6 wk of absent proteinuria and with a maximal interval of 3 mo before RTX infusion. The primary outcome was the onset of igg hypogammaglobulinemia during the follow-up according to the igg normal values for age [mean ± standard deviation(SD)].RESULTS We enrolled 20 patients with mean age at NS diagnosis of 4.2 ± 3.3 years. The mean age at the first RTX infusion was 10.9 ± 3.5 years. Eleven out of twenty patients(55%) developed igg hypogammaglobulinemia. None of these patients showed severe or recurrent infections. Only one patient suffered from recurrent acute otitis media and underwent substitutive igg infusion. Three patients undergoing only the two "starting doses" experienced normalization of igg levels. Using Kaplan-Meier analysis, the cumulative proportion of patients free of igg hypogammaglobulinemia was 57.8% after the first RTX dose, 51.5% after the third dose, 44.1% after the fourth dose, and 35.5% after the fifth *** RTX can induce igg hypogammaglobulinemia in patients with pre-RTX igg normal values. None of the treated patients showed severe infections.

A DNA fragment about 1.5 kb has been isolated from spleen of adult Chinese swine （?）CR. The DNA fragment encodes immunoglobulin igg H chain gene. Sequencing analysis showed that the DNA fragment is 1 425 bp long, complete CDS. The C region of the gene has been classified as Subclass lg γ3, and is the same as reported by Sun et al., but V region of the present gene is 42 bp less by comparison. The gene has been ligated into expression vector pET-3b （NSEB）（ - ）. A protein about 52 ku has been expressed in E. coil with an expression level of about 21%.
