Interaction between hydrogen sulfide/cystathionine γ-lyase and carbon monoxide/heme oxygenase pathways in aortic smooth muscle cells

Aim:To investigate the interaction between hydrogen sulfide（H2S）/cystathion-ine γ-lyase（CSE）and carbon monoxide（CO）/heme oxygenase（HO）pathways inaortic smooth muscle cells（ASMC）.Methods:The ASMCs were divided into thefollowing groups:（1）the control group;（2）the zinc protoporphyrin（ZnPP）20μmol/L group;（3）the propargylglycine（PPG）2 mmol/L,4 mmol/L and 10 mmol/Lgroups;and（4）the sodium hydrosulfide（NariS）1×10-5mol/L,1×10-4mol/L and1×10-3mol/L *** of the groups was further divided into 6h,12h,18hand 24h *** CO level,represented by carboxyhemoglobin（HbCO）content was measured using a spectrophotometric method and H2S content wasdetected by a sensitive electrode *** and HO-1 expressions were de-tected by Western ***:The H2S content in the medium and CSEexpression by ASMC were markedly increased by ZnPP compared with the con-trol *** content in the medium and HO-1 expression by the ASMCstarted strengthening following 24h treatment with PPG at 2 mmol/L,but werefurther strengthened following 18h and 24h treatment with PPG at 4 mmol/Lcompared with the controls（P<0.01）.PPG at 10 mmol/L increased the HbCO levelin the medium following 18h treatment and increased HO-1 expression by theASMC following 12h ***,NariS at 1×10-5mol/L and 1×10-4mol/Ldecreased the HbCO level in the medium and HO-1 expression by the ASMC after6h and 12h treatment,while NariS at 1×10-3mol/L decreased them at all timepoints of the ***:The results suggested that endogenousCO/HO and H2S/CSE pathways inhibited each other in ASMC under physiologi-cal conditions.

Absorption characteristics,model,and molecular mechanism of hydrogen sulfide in morpholine acetate aqueous solution

The solubility of H_(2)S was measured in solutions of N-butyl-N-methylmorpholine acetate([Bmmorp][Ac])containing 20%-40%(mass)water at experimental temperatures ranged from 298.15 to 328.15 K and pressures up to 320 k *** total solubility of H_(2)S increased with higher temperatures,lower pressures,and reduced water *** reaction equilibrium thermodynamic model was used to correlate the solubility *** results indicate that the chemical reaction equilibrium constant decrease with increasing water content and temperature,whereas Henry constant increase with increasing water content and *** with other ionic liquids,H_(2)S exhibits a higher physical absorption enthalpy and a lower chemical absorption enthalpy in[Bmmorp][Ac]aqueous *** suggests that[Bmmorp][Ac]has a strong physical affinity for H_(2)S and low energy requirement for *** chemical methods were used to investigate the molecular mechanism of H_(2)S absorption in ionic *** interaction energy analysis revealed that the binding of H_(2)S with the ionic liquid in a1:2 ratio is more *** analyses by the methods of the interaction region indicator and the atoms in molecules were conducted to the interactions between H_(2)S and the ionic liquid.

Dynamics of liquid-phase catalytic oxidation of hydrogen sulfide removal in rural biogas

hydrogen sulfide in rural biogas was removed with liquid-phase catalytic *** using rare earth as catalyst,and sulfosalicylic acid as stabilizer,H2S purification efficiency could increase as high as 96%,and sulfur capacity of the composite solution was about 3 g/*** results show that purification efficiency was affected by catalyst addition,pH,experimental temperature,and sulfur *** parameters effects on catalytic oxidation were studied,and the optimized conditions were that Fe3+ concentration 0.08 mg/L,reaction temperature 70°C,pH 9.0,with a absorption solution volume of 50 mL,a gas flow rate 200 mL/min,and H2S mass concentration of 1.58-2.02 mg/m3.

Effect of exogenous hydrogen sulfide in the nucleus tractus solitarius on gastric motility in rats

BACKGROUND hydrogen sulfide(H2S)is a recently discovered gaseous neurotransmitter in the nervous and gastrointestinal *** exerts its effects through multiple signaling pathways,impacting various physiological *** nucleus tractus solitarius(NTS),a vital nucleus involved in visceral sensation,was investigated in this study to understand the role of H2S in regulating gastric function in *** To examine whether H2S affects the nuclear factor kappa-B(NF-κB)and transient receptor potential vanilloid 1 pathways and the neurokinin 1(NK1)receptor in the *** Immunohistochemical and fluorescent double-labeling techniques were employed to identify cystathionine beta-synthase(CBS)and c-Fos co-expressed positive neurons in the NTS during rat *** motility curves were recorded by inserting a pressure-sensing balloon into the pylorus through the stomach *** in gastric motility were observed before and after injecting different doses of NaHS(4 nmol and 8 nmol),physiological saline,Capsazepine(4 nmol)+NaHS(4 nmol),pyrrolidine dithiocarbamate(PDTC,4 nmol)+NaHS(4 nmol),and L703606(4 nmol)+NaHS(4 nmol).RESULTS We identified a significant increase in the co-expression of c-Fos and CBS positive neurons in the NTS after 1 h and 3 h of restraint water-immersion stress compared to the expressions observed in the control ***-NTS injection of NaHS at different doses significantly inhibited gastric motility in rats(P0.05).CONCLUSION NTS contains neurons co-expressing CBS and c-Fos,and the injection of NaHS into the NTS can suppress gastric motility in *** effect may be mediated by activating TRPV1 and NK1 receptors via the NF-κB channel.

Reactions of Group V Metal Atoms with hydrogen sulfide： Argon Matrix Infrared Spectra and Theoretical Calculations

The reaction of laser-ablated vanadium, niobium and tantalum atoms with hydrogen sulfide has been investigated using matrix isolation FTIR and theoretical calculations. The metal atoms inserted into the H-S bond of H2S to form the HMSH molecules （M=V, Nb, Ta）, which rearranged to H2MS molecules on annealing for Nb and Ta. The HMSH molecule can also further react with another H2S to form the H2M（SH）2 molecules. These new molecules were identified on the basis of the D2S and H234S isotopic substitutions. DFT （B3LYP and BPW91） theoretical calculations are used to predict energies, geometries, and vibrational frequencies for these novel metal dihydrido complexes and molecules. Reaction mechanism for formation of group V dihydrido complex was investigated by DFT internal reaction coordinate calculations. The dissociation of HVSH gave VS＋H2 on broad band irradiation and reverse reaction happened on annealing. Based on B3LYP calculation releasing hydrogen from HVSH is endothermic only by 13.5 kcal/mol with lower energy barrier of 16.9 kcal/mol.

Study on Industrial Application of hydrogen sulfide Removal by Wet Oxidation Method with High Gravity Technology

The removal of hydrogen sulfide from gas plays an important role in rational utilization of resources and environ- mental protection. In this paper, the process of hydrogen sulfide removal by wet oxidation method in a rotating packed bed was investigated in a scale for treating 10 000 Nm3/h of gas. On the basis of studying the influence of the species and con- centration of alkali source, the liquid/gas volume ratio, the high gravity factor, and the hydrogen sulfide content in feed gas on the desulfurization effect, the suitable technological conditions were obtained. The hydrogen sulfide removal efficiency could reach 98.0% under these conditions. The results of continuous operation of process facilities showed that the high gravity method has many merits including higher desulfurization rate, good stability in operation, lower liquid/gas volume ratio, greater operation elasticity, and apparent energy saving effects.

Stomatal aperture controls the balance between transpirational water loss and photosynthetic carbon dioxide(CO2)*** are surrounded by pairs of guard cells that sense and transduce environmental or stress signals to induce diverse endogenous responses for adaptation to environmental *** a recent decade,hydrogen sulfide(H2S)has been recognized as a signaling molecule that regulates stomatal *** this review,we summarize recent progress in research on the regulatory role of H2S in stomatal movement,including the dynamic regulation of phytohormones,ion homeostasis,and cell structural *** focus especially on the cross talk among H2S,nitric oxide(NO),and hydrogen peroxide(H2O2)in guard cells,as well as on H2S-mediated post-translational protein modification(cysteine thiol persulfidation).Finally,we summarize the mechanisms by which H2S interacts with other signaling molecules in plants under abiotic or biotic *** on evidence and clues from existing research,we propose some issues that need to be addressed in the future.

β-Cyclodextrin-Based Nitrosoglutathione Improves the Storage Quality of Peach by Regulating the Metabolism of Endogenous Nitric Oxide, hydrogen sulfide, and Phenylpropane

Nitrosoglutathione(GSNO)andβ-cyclodextrin(β-CD)exhibit positive roles in regulating fruit ***,there are few reports about the effects of GSNO andβ-CD on enhancing storability and boosting nitric oxide(NO),hydrogen sulfide(H2S),and phenylpropane metabolism in fruits during storage.“Xintaihong”peach were treated with 0.5,1.0,1.5mmol L−1 GSNO in 0.5%(w/v)β-CD solution(GSNO/β-CD).The effects of GSNO/β-CD on endogenous NO,H2S,and phenylpropane metabolism were *** with GSNO/β-CD increased the color difference of peach and inhibited the increase of respiratory intensity,weight loss,and relative *** with 1.0 mmol L−1 GSNO/β-CD increased the nitric oxide synthase(NOS-like)activity and L-arginine content,thereby promoting the accumulation of endogenous *** improving the activities of L-cysteine desulfhydrylase(L-CD),O-acetylserine sulfur lyase(OAS-TL),serine acetyltransferase(SAT),GSNO/β-CD increased the content of endogenous H2S in *** with GSNO/β-CD increased the activities of phenylalanine ammonia-lyase(PAL),4-coumarate-CoA ligase(4CL),and cinnamic acid-4-hydroxylase(C4H),promoted the increase of total phenols,flavonoids,and lignin in *** results indicated that GSNO/β-CD treatment better maintained the quality of peach by improving the metabolism of endogenous NO,H2S,and phenylpropane during storage.

Changes in hydrogen sulfide in Rats with Hepatic Cirrhosis in Different Stages

This study aimed to observe changes in the hydrogen sulfide（H_2S） system in the blood and liver tissue of rats with hepatic cirrhosis at different stages by studying the effect of H_2S on the course of hyperdynamic circulation in rats with hepatic cirrhosis. H_2S concentration in the blood from the portal vein and inferior vena cava of hepatic cirrhosis rat model induced with carbon tetrachloride was detected on the 15 th, 30 th, and 52 nd day. The expression of cystathionine β-synthase（CBS） and cystathionine γ-lyase（CSE） protein, and CBS and CSE mRNA in the liver was detected by immunohistochemistry and reverse transcriptase polymerase chain reaction（RT-PCR）, respectively. The results indicated that H_2S concentration in the blood from the portal vein and inferior vena cava of rats with hepatic cirrhosis was significantly lower than that in the control group. H_2S was gradually decreased with the development of the disease and significantly lower in the blood from portal vein than in the blood of inferior vena cava at the mid-stage and the late stage groups. The expression levels of CBS and CSE protein, and CBS and CSE mR NA in the livers with hepatic cirrhosis at different stages were all higher than those in the control group, and the expression gradually increased with the development of the disease. The expression of CBS was lower than CSE in the same stages. The results indicated that the CSE mRNA was expressed predominantly in the cirrhosis groups as compared with CBS mRNA. Among experimental rats, the H_2S system has an important effect on the occurrence and development of hyperdynamic circulation in rats with hepatic cirrhosis. This finding adds to the literature by demonstrating that H_2S protects vascular remodelling in the liver, and that CSE is indispensable in this process.
