＜正＞Objective Previous papers reported that neural stem cells could be induced to differentiate into cholinergic neurons in the presence of retinoic acid（RA） and sonic hedgehog（SHH）.However,the mechanism was not well understood. As neural development-related genes,hes1 and mash1 are widely expressed in the developing central and peripheral nervous *** this study,we investigated the expression and impact of hes1 and mashl genes in the process of neural stem cells differentiating into cholinergic *** We isolated neural stem cells from the spinal cord of fetal *** the combination of RA,SHH,neurotrophin-3（NT-3） and brain-derived neurotrophic factor（BDNF）,the spinal cord stem cells（SCSCs） of third passage（P3） were induced to differentiate into cholinergic ***-time quantitative PCR was used to detect the expression changes of hes1,mashl and Nestin mRNA in the P3 SCSCs and that after induction for 1 and *** The expression of hes1,mashl and Nestin mRNA decreased in a significant manner after lw of induced differentiation compared with undifferentiated SCSCs,and increased slightly after 2w of induced differentiation. Conclusion The hes 1 and mash 1 gene may play an important role in the process of SCSCs differentiating into cholinergic *** study may contribute to the identification of molecular mechanisms involved in the differentiation of SCSCs into neural cells.
