Background and Aim: Infections caused by hepatotropic pathogens, such as HBV, still raise concerns worldwidely while with no effective curable regimens. Lack of suitable animal model to directly study the immunopathogenesis of human hepatotropic viral infections is a main bottleneck for novel drug investigations. Thus, establishment of a versatile mouse model harboring both functional human immune system and human hepatocytes would be of high ***: To achieve human hepatocyte repopulation in mouse liver, enriched human fetal liver progenitors were injected into the spleen of immunodeficient mice followed with continuous treatment of anti-mouse fas antibody. Humanized mice with functional human immune system were generated by co-transplantation of human fetal thymic tissues(under renal capsules) and human fetal liver CD34+ cells(intravenous injection) into immunodeficient animals after sublethal total body ***: Secretion of human albumin in the animals transplanted with human fetal liver progenitor cells lasted over 14 weeks post transplantation, and reached up about to 400ng/m L. Immunohistochemical staining of mouse liver section by anti-human hepatocyte(Hep par 1), anti-human CK8/18, and anti-human albumin antibodies revealed that around 30-40% human hepatocytes repopulated in the humanized mouse livers. Moreover, the chimerism of human immune cells in the periphery blood and livers of the humanized mice were approximately 80% and 90% respectively at 14 weeks posttransplantation of Thy/CD34+ cells, including CD3+(both CD8 and CD8) T cells, CD19+ B cells, as well as CD14+ monocytes/macrophages and so ***: We developed a humanized mouse model with high level of human immune system and autologous human hepatocyte engraftment which may serve as a valuable model to study human hepatotropic pathogen infection.
