hepatoprotective activity of the Ethanol Extract of Sarcopyramis Nepalensis

The present study examined the protective effect of the ethanol extract of Sarcopyramis nepalensis (EESN) on agents-induced hepatotoxicity in mice and the possible mechanism. Acute liver injury was induced by administration of either CCl4 or D-GalN. The animals were divided into 5 groups in terms of different treatment: normal group, CCl4 or D-GalN group, silymarin or bifendate group, low dose EESN group (10 mg/kg) and high dose EESN group (30 mg/kg). Liver function was evaluated by detecting the levels of serum alanine aminotransferase (ALT) and aspartate aminotransferase (AST). The oxidize stress markers were measured, including malondialdehyde (MDA), glutathione peroxidase (GSH) and superoxide dismutase (SOD). Liver tissues were histopathologically examined by hematoxy-lin-eosin (H&E) staining. The acute toxicity study revealed that there was no toxicity of EESN at the dose of 5 g/kg in mice. The levels of ALT and AST in serum, and the MDA level in live tissues were significantly increased and the activities of SOD and GSH substantially decreased in mice after CCl4 or D-GalN treatment. These biochemical and oxidize stress markers were profoundly improved after treatment with EESN at different doses, which was similar to the results of silymarin or bifendate treatment. The histophathological examination revealed the significant improvement in the pathological changes of the liver in EESN-treated mice as compared to those in CCl4 or D-GalN group. It was concluded that EESN possesses potential antioxidant and hepatoprotective properties and has therapeutic potential for liver diseases.

AIM: To study the hepatoprotective capacity of Sapindus mukorossi (S. mukorossi) and Rheum emodi (R. emodi) extracts in CCl4 treated male rats. METHODS: The dried powder of S. mukorossi and R. emodi was extracted successively with petroleum ether, benzene, chloroform, and ethanol and concentrated in vacuum. Primary rat hepatocyte monolayer cultures were used for in vitro studies. In vivo, the hepatoprotective capacity of the extract of the fruit pericarp of S. mukorossi and the rhizomes of R. emodi was analyzed in liver injured CCl4-treated male rats. RESULTS: In vitro: primary hepatocytes monolayer cultures were treated with CCl4 and extracts of S. mukorossi & R. emodi. A protective activity could be demonstrated in the CCl4 damaged primary monolayer culture. In vivo : extracts of the fruit pericarp of S. mukorossi (2.5 mg/mL) and rhizomes of R. emodi (3.0 mg/mL) were found to have protective properties in rats with CCl4 induced liver damage as judged from serum marker enzyme activities. CONCLUSION: The extracts of S. mukorossi and R. emodi do have a protective capacity both in vitro on primary hepatocytes cultures and in in vivo in a rat model of CCl4 mediated liver injury.

New triterpenoid saponins from the leaves of Ilex chinensis and their hepatoprotective activity

Seven new triterpenoid saponins,including five ursane-type saponins,ilexchinenosides R–V(1–5),and two oleananetype saponins,ilexchinenosides W–X(6–7),with four known triterpenoid saponins(8–11)were isolated from the leaves of Ilex *** structures were elucidated by comprehensive spectroscopic 1 D and 2 D NMR and HR-ESI-MS *** sugar moieties were determined by HPLC analysis compared with standards after hydrolysis and *** 1,2,4,9 and 10 exhibited potential hepatoprotective activity against N-acetyl-p-aminophenol(APAP)-induced Hep G2 cell injury in vitro.

Group 2 served as hepatotoxin(CCL_4 treated) *** 3 served as positive control(Silyntarin) group,and Group 4.S and ft served as(73,150 and 300 nig/kg bw p.o.)*** leaf extract treated ***,in vitro antioxidant DPPH,hydroxyl radical scavenging activity(HRSA),NO,ferric reducing antioxidant power(FRAP),lipid hydroperoxide(LPO) and super oxide dismutase(SOD) were also analyzed for the leaf ***:The levels of the serum parameters such as serum glutamic oxaloacetic transaminase(SGOT).serum glutamic pyruvic transaminase(SGPT).alkaline phosphatase(ALP),bilirubin,cholesterol(CHL).sugar and lactate dehydrogenase(LDH) were significantly increased in COL_4 treated rats when compared with the control group(Phepatoprotective effect of the *** leaf extract might be due to the presence of phenolic groups,terpenoids and alkaloids and in vitro antioxidant properties.

Background: Many kinds of orchids have significant health benefits although adequate research on their biological functions is yet to be carried out. This study investigated the paracetamol-induced liver damage-protecting effect of epiphytic Aerides odorata methanol extract (AODE). Methods: The protective effects of AODE were studied by analyzing its effect on liver function parameters, messenger RNA (mRNA) expression, and tissue histopathological architecture. The results were confirmed by ligand-receptor interaction of molecular docking and multitarget interaction of network pharmacological analyses. Results: AODE significantly (p < 0.05) minimized the dose-dependent increase in acid phosphatase, aspartate aminotransferase, alanine aminotransferase, alkaline phosphatase, gamma-glutamyl transferase, lactate dehydrogenase, and total bilirubin compared to the reference drug silymarin. Malondialdehyde level decreased, and the antioxidant genes catalase (CAT), superoxide dismutase (SOD), beta-actin, paraoxonase-1 (PON1), and phosphofructokinase-1 (PFK-1) were upregulated in AODE-treated paracetamol-intoxicated rats. A total of 376 compounds comprising phenols and flavonoids were identified using ultra-high-performance liquid chromatography-quadrupole time-of-flight-mass spectrometry (UPLC-qTOF-MS). The online toxicity assessment using SwissADME and admetSAR exhibited drug-like, nontoxic, and potential pharmacological properties. Additionally, in silico analysis showed that isoacteoside, one of the identified compounds, exhibited the best docking score (-11.42) with the liver protein human pituitary adenylate cyclase-1 (Protein Data Bank ID: 3N94). Furthermore, network pharmacology analysis identified the top 10 hub genes, namely AKT1 (protein kinase B), CTNNB1 (catenin beta-1), SRC (proto-oncogene c-Src), TNF (tumor necrosis factor), EGFR (epidermal growth factor receptor), HSP90AA1 (heat shock protein 90 alpha), MAPK3 (mitogen-activated protein kinase 3), STAT3 (signa

Two new phenylpropanoid glycosides from the aerial parts of Lespedeza cuneata

Two new phenylpropanoid glycosides named cuneataside E (1) and cuneataside F (2), were isolated from the aerial parts of Lespedeza cuneata (Dum. Cours.) G. Don, whose structures were E and Z isomer, respectively. Their structures were elucidated on the basis of comprehensive spectroscopic analysis (UV, IR, HR-ESI-MS, 1D and 2D NMR). In in vitro bioassays at 10 mu mol/L, compound 1 showed moderate hepatoprotective activity against N-acetyl-p-aminophenol (APAP)-induced toxicity in HeG2 cells. Objective:To evaluate bepato protective and antioxidant capacity of Melochia corchorifolia （***）aerial part ***:Antioxidant activitywas evaluated by using three free radicals（Superoxide,Hydroxyl and DPPH） and hepatoprotective activity was assessed against CCl4 induced liver intoxication in ***:The extracts produced concentration dependent percentage protection in decrease of serum enzymes and percentage inhibition on free *** all extracts methanol extract showed better activity with percentage protection of SGOT（78.98%）,SGPT（79.65%）,ALP（82.48%） and total bilirubin（80.0%）levels against CCl4 liver intoxication and also methanolic extract showed better activity with IC50 values on superoxide. hydroxyl and DPPH radicals were 127μg.240μg and 179μ***:From the results obtained during the study it could be concluded that *** aerial part extracts have antioxidant and hepaloprotective *** study is necessary for isolation and characterization of bioactive molecules which are responsible for hepatoprotective and antioxidant activity.

An ongoing phytochemical investigation of Atractylodes lancea lead to the isolation of eight new compounds named（5 R,7 R,10 S）-14-hydroxyisopterocarpolone-11-O-β-D-glucopyranoside（1）,（1 S,7 R,10 R）-11,15-dihydroxy-4-guaien-3-one 11-O-β-D-glucopyranoside（2）,（2 E,8 E,10 E,12 R）-tetradeca-2,8,10-triene-4,6-diyne-1,12,14-triol-1-O-β-D-glucopyranoside（3）,（2 E,8 E,10 E,12 R）-tetradeca-2,8,10-triene-4,6-diyne-1,12,14-triol-1-O-β-D-apiofuranosyl-（1→6）-β-D-glucopyranoside（4）,（2 E,8 Z）-deca-2,8-diene-4,6-diyne-1,10-diol-1-O-β-D-apiofuranosyl-（1→6）-β-D-glucopyranoside（5）,（2 E,8 R）-decene-4,6-diyne-1,8-diol-O-di-β-D-glucopyranoside（6）,（2 E,8 R）-decene-4,6-diyne-1,8-diol-8-O-β-D-glucopyranoside（7）, and（2 E,8 S）-decene-4,6-diyne-1,8-diol-8-O-β-D-glucopyranoside（8）. Their structures were elucidated by means of spectroscopic and spectrometric analyses（UV, IR, 1 D and 2 D NMR, and HRESIMS）. The absolute configurations of 1-4 were established based on the experimental and calculated ECD, whereas those of 6-8 were determined by comparing the specific rotations of their aglycones. Notably, compounds 6 and 7 exhibited significant hepatoprotective activities against APAP-induced HepG2 cell injury at a concentration of 10 μ M. Compounds 4, 6, and 7 showed weak anti-inflammatory effects on LPS-induced NO production in microglia BV2 cells at a concentration of 10 μ M.

Puerariae Radix,the root of Pueraria lobata(Willd.) Ohwi,is a very important traditional Chinese medicine(TCM) with edible and pharmaceutic *** has been traditionally used for treating various diseases such as wind heat disease,head and neck pain,diabetes,alcohol poisoning,cardiovascular and other *** chemical investigations of pueraria species showed that isoflavonoids(aglycones,C-glycosides,O-glycosides) are characteristic constituents of pueraria *** the quality-marker of Pueraria lobata(Willd.) Ohwi,puerarin exhibits potent hepatoprotective effect which has been confirmed from cell culture *** our current research,to seek ingredients possessing potential hepatoprotective activities,two new isoflavone glycoside derivatives(1,2) together with nine known compounds(3-11) were obtained from the water extract of the root of Pueraria lobata(Willd.) *** the basis of chemical evidences and spectral analyses(UV,ECD,IR,ID and 2 D-NMR,HRMS),the structures of these compounds were elucidated as 8-C-α-glucofuranosyl-7,4’-dihydroxy-3’-methoxyisoflavone(1),8-C-β-glucofuranosyl-7,4’-dihydroxy-3’-methoxyisoflavone(2),puerarin(3),neopuerarin A(4),neopuerarin B(5),daidzin(6),daidzein(7),3’-methoxypuerarin(8),puerarin xyloside(9),mirificin(10),3’-hydroxypuerarin(11).Furthermore,all these isoflavones were investigated the hepatoprotective activities on HepG2 cells.10 μM tested compounds did not show obvious cytotoxic effects on HepG2 cells and shows significant hepatoprotective activities against D-galactosamine-induced toxicity in HepG2 *** results indicated that isoflavonoids(aglycones,C-glycosides,O-glycosides) exhibited hepatoprotective activities.
