Backgrounds:Hepatitis B virus(hbv)is an important human pathogen causing acute and chronic liver *** an estimated 350 million individuals are chronically infected and potentially develop liver cirrhosis or liver cell *** is a small enveloped,DNA-containing *** assembly of progeny hbv virions begins with the formation of icosahedral nucleocapsids that package the viral pregenomic RNA together with the viral *** the capsids,composed of 180 or 240 copies of the 21 kDa core protein and the partially double-stranded 3.2kbDNA genome is synthesized through reverse transcription of the pregenomic *** nucleocapsids,formed in the cytoplasm,can then be enclosed by the viral envelope *** budding of hbv is supposed to occur at intracellular *** intracellular core particles can partition into nucleus and ***,envelope protein-independent core particle budding,so-called naked core particle secretion,has been incidentally reported for hbv replicating *** functional implications of naked nucleocapsid remain ***:To assess the potential effect of naked capsids on host immunity against hbv infection,we analyzed the hbvspecific humoral and cellular immune responses in BALB/c mice hydrodynamically injected with plasmid expressing wildtype,K96R core,and Y132 core ***:Plasmids expressing wild type of hbv core protein(HBcWT),or HBcK96R,HBcY132A mutants were constructed and then transfected into hepatocyte-derived cell *** location of HBc proteins were observed by confocal *** supernatant and cell lysate of transfected cells were collected to detect the naked capsid by native agarose gel *** of HBcWT,HBcK96R or HBcY132A were hydrodynamically injected into tail vein of Balb/c *** blood were collected at different time *** were tested by *** 14 dpi,the splenocytes of mice receiving the DNA were
