Ferulic acid(FA)and vanillic acid(VA)are considered as major phenolic metabolites of cyanidin 3-glucoside,a polyphenol that widely exists in plants that possess a protective effect against oxidative stress and inflammation in our previous study.This study aimed to investigate the effect of FA and VA on inflammation,gut barrier function,and growth performance in a weaned piglet model challenged with lipopolysaccharide(LPS).Thirty-six piglets(PIC 337×C48,28 d of age)were randomly allocated into 3 treatments with 6 replicate pens(2 piglets per pen).They were fed with a basal diet or a diet containing 4,000 mg/kg of FA or VA.Dietary supplementation of VA significantly increased average daily gain(ADG)(Pgut microbiota indicated that both FA and VA increased the Firmicutes/Bacteroidetes ratio alongside reducing the relative abundance of the Prevotellaceae family including Prevotella 9 and Prevotella 2 genera,but enriched the Lachoiraceaea family including the Lachnospiraceae FCS020 group(Pgut microbiota.

Metabolic syndrome(Met S)is a chronic disease associated with the disturbance of gut microbiota homeostasis.Metabolites derived from gut microbes play essential roles in Met S prevention and therapy.Here,we focused on the inhibitory effect of the extract of millet bran protein(EMBP)on a high-fat diet(HFD)-induced Met S,aiming to identify gut microbiota and their metabolites that involve in the anti-Met S activity of EMBP.The obesity,chronic inflammation,insulin resistance in Met S mouse models were abolished after EMBP treatment.The protective mechanism of EMBP against HFD-induced Met S may depend on improved gut barrier function.Using microbiome analysis,we found that EMBP supplementation improved gut microbiome dysbiosis in Met S mice,specifically upregulating Bacteroides acidifaciens.The fecal microbiota transplantation(FMT)also demonstrated this phenomenon.In addition,metabolomic analysis showed that EMBP mediates metabolic profiling reprogramming in Met S mice.Notably,a microbiota-derived metabolite,gamma-aminobutyric acid(GABA),is enriched by EMBP.In addition,exogenous GABA treatment produced a similar protective effect to EMBP by improving NRF2-dependent gut barrier function to protect HFDinduced Met S.The results suggest that EMBP suppress host Met S by remodeling of gut microbiota as an effective candidate for next-generation medicine food dual purpose dietary supplement to intervene in MetS.

Obesity is associated with gut dysbiosis and metabolic endotoxin.Junshanyinzhen tea extract(JSTE)reduced fat accumulation and body weight in obese mice.However,the effects and mechanism of JSTE in preventing obesity were unclear.Therefore,we used different doses of JSTE(75,150 and 300 mg/(kg·day))to evaluate the effect on high-fat diet(HFD)-induced rats under 8 weeks of intervention.Here,our results showed that JSTE could significantly reduce body weight gain,blood lipid levels and fat accumulation,improve fatty damage in liver tissue(Pgut microbiota dysbiosis,intestinal barrier dysfunction,metabolic endotoxemia and chronic low-grade infl ammation in HFD-induced rats.

Non-alcoholic steatohepatitis(NASH)is a severe form of non-alcoholic fatty liver disease without effective treatment.The traditional Chinese medicine formulation Pien Tze Huang(PTH)can suppress inflammatory diseases.Here,we evaluate the effects of PTH on the evolution of NASH and its underlying mechanisms.We found that PTH prevented the development of steatohepatitis induced by various dietary models,including a high-fat high-cholesterol(HFHC)diet,choline-deficient high-fat diet(CD-HFD),and methionine-and choline-deficient(MCD)diet,along with significant suppression of liver injury,hepatic triglyceride,and lipid peroxidation.Moreover,ten days of PTH treatment after the onset of NASH significantly ameliorated MCD diet-induced steatosis and liver injury in mice.Through the metagenomic sequencing of stool samples,we found that PTH administration restored the gut microbiota with enrichment of probiotics including Lactobacillus acidophilus(L.acidophilus),Lactobacillus plantarum,Lactococcus lactis,and Bacillus subtilis.The enriched L.acidophilus prevented MCD diet-induced steatohepatitis.In addition,PTH restored the gut barrier function in mice with steatohepatitis,as evidenced by reduced intestinal permeability,decreased serum lipopolysaccharides(LPS)level,and increased epithelial tightjunction protein E-cadherin expression.Our metabolomic analysis via liquid chromatography-mass spectrometry profiling identified the alteration in the metabolism of bile acids in the portal vein of PTHtreated mice.We further confirmed that an intact gut microbiota is necessary for PTH to exhibit antisteatohepatitis effects.In conclusion,PTH protects against steatohepatitis development by modulating the gut microbiota and metabolites.PTH is a potential promising prophylactic and therapeutic option for patients with NASH.

An Experimental Study on Curative Effect of Chinese Medicine Qing Yi Tang (清胰汤) in Acute Necrotizing Pancreatitis

There are currently over 1.9 billion people who are obese or overweight, leading to considerable economical costs as well as public health challenge. A growing body of evidence suggests that the onset of obesity is influenced by genetic, environmental, and behavioral factors. While the imbalance between energy intake and energy expenditure does explain the growing incidence of obesity, the imbalance cannot on its own explain the metabolic alterations associated with obesity. Recently, there is increased attention on the link between gut microbial composition and obesity. gut microbes may be a key endocrine organ that contributes to changes in both the gut-to-liver and gut-to-adipose tissue axes characterising these metabolic disorders(e.g. obesity, type 2 diabetes, insulin resistance) via mechanisms associated with gut barrier dysfunctions. Emerging evidence that one potential contributor to the development of chronic metabolic diseases is dysbiosis, which increases intestinal permeability and induces chronic low-grade inflammatory state in the host. The intestinal barrier is a selective physical and immunological barrier that deters the translocation of harmful luminal antigens while facilitating water, electrolytes, and nutrient absorption into circulation. gut barrier is essential in maintaining intestinal homeostasis but the integrity of intestine remains vulnerable to changes by host behavior. One prominent example is the negative effect of high-fat diet on gut integrity. Higher fat intake has long been associated with chronic disease risk and increasing evidence suggests this could be partially due to increased intestinal permeability through tight-junction disruption, mucous layer diminishment, and local inflammatory response. Objective: To observe the curative effect of the traditional Chinese medicine (TCM) Qing Yi Tang (QYT, 清胰汤) in acute necrotizing pancreatitis (ANP). Methods: Twenty three dogs were randomly divided into 3 groups. In the control group (n = 7), animals underwent laparatomy only. In the ANP group (n =8), acute necrotizing pancreatitis was induced by injection of 0. 5 ml/kg 5 % sodium taurocholate with 3000 u/kg trypsin into the pancreatic duct. While in TCM group (n = 8) were fed everyday with QYT after onset of ANP. All animals were sacrificed 7 days later and organs were gathered and cultured. Mucosal and luminal floras of the intestine were analysed. Pancreas and ileal mucosa were examined histologically and ultramicroscopically, the levels of lipopolysaccharide (LPS) and amylase in blood were determined. Results: In the TCM group, histologic and ultra-structural damages in pancreas and lieal mucosa were much milder as compared with those of ANP group. In ANP group, there was a significant increase of E. colt and bacteroids, and a significant decrease of bifidobacteria, lactobacilli and enterococci in the intestinal mucosa, while in the TCM group, these changes were alleviated significantly (Pgut barrier function by alleviating the damage of intestinal mucosa and microecologic disturbance following acute pancreatitis. As a result, the chances of BT and enterogenic infection declined. These preparation might be promising in the prophylaxis and treatment of infection complicating ANP.
