Differential neuronal reprogramming induced by NeuroD1 from astrocytes in Grey matter versus white matter

A new technology called in vivo glia-to-neuron conversion has emerged in recent years as a promising next generation therapy for neural regeneration and repair. This is achieved through reprogramming endogenous glial cells into neurons in the central nervous system through ectopically expressing neural transcriptional factors in glial cells. Previous studies have been focusing on glial cells in the Grey matter such as the cortex and striatum, but whether glial cells in the white matter can be reprogrammed or not is unknown. To address this fundamental question, we express NeuroD1 in the astrocytes of both Grey matter(cortex and striatum) and white matter(corpus callosum) to investigate the conversion efficiency, neuronal subtypes, and electrophysiological features of the converted neurons. We discover that NeuroD1 can efficiently reprogram the astrocytes in the Grey matter into functional neurons, but the astrocytes in the white matter are much resistant to neuronal reprogramming. The converted neurons from cortical and striatal astrocytes are composed of both glutamatergic and GABAergic neurons, capable of firing action potentials and having spontaneous synaptic activities. In contrast, the few astrocyte-converted neurons in the white matter are rather immature with rare synaptic events. These results provide novel insights into the differential reprogramming capability between the astrocytes in the Grey matter versus the white matter, and highlight the impact of regional astrocytes as well as microenvironment on the outcome of glia-toneuron conversion. Since human brain has large volume of white matter, this study will provide important guidance for future development of in vivo glia-to-neuron conversion technology into potential clinical therapies. Experimental protocols in this study were approved by the Laboratory Animal Ethics Committee of Jinan University(approval No. IACUC-20180321-03) on March 21, 2018.

Construction mechanism of whitenization weight function and its application in Grey clustering evaluation

The clustering evaluation can be used to scientifically classify the objects to be evaluated according to the information aggregation of various evaluation rules. In Grey weighted clustering evaluation, the index clustering rule relies on the construction of the whitenization weight function, while the existing construction method of the linear function lacks the construction mechanism analysis and validity explanation. A normative construction principle is put forward by analyzing the construction mechanism of the function. Through proving the normative principle of the function,the basic modal function(BMF) is proposed and characterized by different function forms. Then, a new type of the whitenization weight function and its Grey clustering evaluation model algorithm are given by studying the mechanism and nature of the construction of different forms of the function. Finally, the comparative study for self-innovation capability of defense science and technology industry(DSTI) is taken as an example. The results show that the different construction ways of the function have an effect on the clustering result. The proposed construction mechanism can better explain the index clustering rules and evaluation effectiveness,which will perfect the theoretical system of Grey clustering evaluation and be applied to practice effectively.

Autoimmune diseases of the central nervous system(CNS) like multiple sclerosis(MS) are characterized by inflammation and demyelinated lesions in white and Grey matter regions. While inflammation is present at all stages of MS, it is more pronounced in the relapsing forms of the disease, whereas progressive MS(PMS) shows significant neuroaxonal damage and Grey and white matter atrophy. Hence, disease-modifying treatments beneficial in patients with relapsing MS have limited success in PMS. BAF312(siponimod) is a novel sphingosine-1-phosphate receptor modulator shown to delay progression in PMS. Besides reducing inflammation by sequestering lymphocytes in lymphoid tissues, BAF312 crosses the blood-brain barrier and binds its receptors on neurons, astrocytes and oligodendrocytes. To evaluate potential direct neuroprotective effects, BAF312 was systemically or locally administered in the CNS of experimental autoimmune encephalomyelitis mice with distinct Grey-and white-matter lesions(focal experimental autoimmune encephalomyelitis using an osmotic mini-pump). Ex-vivo flow cytometry revealed that systemic but not local BAF312 administration lowered immune cell infiltration in animals with both Grey and white matter lesions. Ex-vivo voltage-sensitive dye imaging of acute brain slices revealed an altered spatio-temporal pattern of activation in the lesioned cortex compared to controls in response to electrical stimulation of incoming white-matter fiber tracts. Here, BAF312 administration showed partial restore of cortical neuronal circuit function. The data suggest that BAF312 exerts a neuroprotective effect after crossing the blood-brain barrier independently of peripheral effects on immune cells. Experiments were carried out in accordance with German and EU animal protection law and approved by local authorities(Landesamt für Natur, Umwelt und Verbraucherschutz Nordrhein-Westfalen;87-51.04.2010.A331) on December 28, 2010.

Grey information coverage interaction relational decision making and its application

This study aims to reflect the information coverage Grey number and the interaction between attributes in Grey relational decision making. Therefore, a multi-attribute decision method based on the Grey information coverage interaction relational degree(GIRD) is proposed. Firstly, this paper defines the information coverage Grey number, and establishes the GIRD model by using the Choquet fuzzy integral and Grey relational principle. It proves that the proposed model not only is the general and unified form of the point relational degree, interval relational degree, mixed relational degree and Grey fuzzy integral relational degree, but also can effectively deal with the interaction between attributes. Further,a decision making example of evaluating the industrial operation quality for 14 cities in Hunan province of China is provided to highlight the implementation, availability, and feasibility of the proposed decision model.

Decision-making approach by employing Grey incidence analysis and group negotiation

For the problems of the consistency ranking of the group decision-making scheme,from the view of group negotiation and system coordination,the Grey incidence analysis and Nash bargaining model are used to establish a consistency group decision-making method.First,the concepts of the consensus partial decision-making program and the consensus overall ideal decision-making program are defined,and then a multi-object optimization model is constructed based on the satisfaction maximization of group negotiation and deviation minimization of system coordination to determine the consensus partial decision-making program and the consensus overall ideal decision-making program.Moreover,the Grey incidence analysis is exploited to measure the close degrees between them.Finally,a real case of the online product evaluation verifies the validity and rationality of the proposed model.

BACKGROUND Antiviral treatment of patients with chronic hepatitis B(CHB)in the Grey zone of treatment comands risk management in order to optimize the health outcome.In this sense,the identification of HBV mutants related with an increased risk of hepatocellular carcinoma(HCC)could be useful to identify subpopulations with potential indication of antiviral treatment.AIM To analyze the prevalence/persistence of hepatitis B virus(HBV)preS and basal core promoter(BCP)/precore/core variants associated to HCC development in CHB patients in the Grey zone.METHODS Work was designed as a longitudinal retrospective study,including 106 plasma samples from 31 patients with CHB in the Grey zone of treatment:Hepatitis B e antigen negative,HBV-DNA levels between 12-20000 IU/mL,normal or discordant transaminase levels during follow up and mild/moderate necroinflammatory activity in liver biopsy or Fibroscan(up to 9.5 kPa).Serum HBVDNA was tested using the Abbott Real Time HBV Assay and the BCP/precore/core and the hepatitis B surface antigen(HBsAg)coding regions were analyzed in positive samples by PCR/bulk-sequencing to identify the HCCrelated HBV mutants.RESULTS High-risk HCC related mutants were detected in 24(77%)patients:19(61%)in the BCP/precore/core,and 7(23%)in the HBsAg coding region(2 preS1 and 5 preS2 deletions).The prevalence of preS deletions was genotype-dependent:3/5(60%)patients with preS2 deletions and 1/2 with preS1 deletions were infected with the HBV-E genotype.Since HBV-E was the most prevalent in sub-Saharan patients,a correlation between preS deletions and ethnicity was also found:6/8(75%)sub-Saharan vs 1/19(5%)Caucasian patients had preS deletions(P=0.00016).Remarkably,this correlation was maintained in those patients infected with HBV-A,a minor genotype in sub-Saharan patients:2/2 patients infected with HBV-A from West Africa vs 0/6 of Caucasian origin had preS deletions.The HCC related variants were the major strains and persisted over time(up to 48 mo).Patients with preS deletions had a significant hig

Multi objective prediction and optimization of control parameters in the milling of aluminium hybrid metal matrix composites using ANN and Taguchi-Grey relational analysis

This study aims to optimize the input parameters such as mass fraction and particle size of SiC along with depth of cut,feed and cutting speed in the milling of Al5059/SiC/MoS2.The hybrid metal matrix composites are generally fabricated by reinforcing of different sizes(10,20,40 μm)of SiC with aluminium at a different levels(5%,10%& 15%)whereas the MoS2 addition is fixed as 2%.The effect of each control factor on response variables are analyzed through Taguchi S/N ratio method.Also,the most significant method for prediction of response parameters is satisfied by ANN model than the regression model.Analysis of variance(ANOVA)results envisage that mass fraction of SiC,feed rate is the most domineering factor on response variable.

Cloud security situation prediction method based on Grey wolf optimization and BP neural network

Aiming at the accuracy and error correction of cloud security situation prediction,a cloud security situation prediction method based on Grey wolf optimization(GWO)and back propagation(BP)neural network is proposed.Firstly,the adaptive disturbance convergence factor is used to improve the GWO algorithm,so as to improve the convergence speed and accuracy of the algorithm.The Chebyshev chaotic mapping is introduced into the position update formula of GWO algorithm,which is used to select the features of the cloud security situation prediction data and optimize the parameters of the BP neural network prediction model to minimize the prediction output error.Then,the initial weights and thresholds of BP neural network are modified by the improved GWO algorithm to increase the learning efficiency and accuracy of BP neural network.Finally,the real data sets of Tencent cloud platform are predicted.The simulation results show that the proposed method has lower mean square error(MSE)and mean absolute error(MAE)compared with BP neural network,BP neural network based on genetic algorithm(GA-BP),BP neural network based on particle swarm optimization(PSO-BP)and BP neural network based on GWO algorithm(GWO-BP).The proposed method has better stability,robustness and prediction accuracy.

Analysis Unking directly genomics, neuroimaging phenotypes and clinical measurements is crucial for understanding psychiatric disorders, but remains rare.Here, we describe a multi-scale analysis using genome-wide SNPs, gene expression, Grey matter volume (GMV), and the positive and negative syndrome scale scores (PANSS) to explore the etiology of schizophrenia. With 72 drug-naive schizophrenic first episode patients (FEPs) and 73 matched heathy controls, we identified 108 genes, from schizophrenia risk genes, that correlated significantly with GMV, which are highly co-expressed in the brain during development. Among these 108 candidates, 19 distinct genes were found associated with 16 brain regions referred to as hot clusters (HCs), primarily in the frontal cortex, sensory-motor regions and temporal and parietal regions.The patients were subtyped into three groups with distinguishable PANSS scores by the GMV of the identified HCs. Furthermore, we found that HCs with common GMV among patient groups are related to genes that mostly mapped to pathways relevant to neural signaling, which are associated with the risk for schizophrenia.Our results provide an integrated view of how genetic variants may affect brain structures that lead to distinct disease phenotypes.The method of multi-scale analysis that was described in this research, may help to advance the understanding of the etiology of schizophrenia.

Application of Grey relational analysis based on Taguchi method for optimizing machining parameters in hard turning of high chrome cast iron

High chrome white cast iron is particularly preferred in the production of machine parts requiring high wear resistance. Although the amount of chrome in these materials provides high wear and corrosion resistances, it makes their machinability difficult. This study presents an application of the Grey relational analysis based on the Taguchi method in order to optimize chrome ratio, cutting speed, feed rate, and cutting depth for the resultant cutting force (Fr) and surface roughness (Ra) when hard turning high chrome cast iron with a cubic boron nitride (CBN) insert. The effect levels of machining parameters on Fr and Ra were examined by an analysis of variance (ANOVA). A Grey relational grade (GRG) was calculated to simultaneously minimize Fr and Ra. The ANOVA results based on GRG indicated that the feed rate, followed by the cutting depth, was the main parameter and contributed to respo ses. Optimal levels of parameters were found when the chrome ratio, cutting speed, feed rate, and cutting depth were 12%, 100m/min, 0.05mm/r, and 0.1mm, respectively, based on the multiresponse optimization results obtained by considering the maximum signal to noise (SIN) ratio of GRG. Confirmation results were verified by calculating the confidence level within the interval width.
