Objective: Inflammation and fibrosis are strongly associated with each other. glycine is present in various traditional Chinese medicines and exhibits anti-inflammatory activity. However, the effects of glycine on myocardial fibrosis(MF) in rats with myocardial infarction(MI) have not been reported. The purpose of this study is to investigate the effects of glycine therapy on MF and comprehend its underlying mechanisms. Materials and Methods: Left anterior descending artery ligation-induced MI in Sprague Dawley rats was leveraged to assess the therapeutic effects of glycine. Rats received either normal saline or glycine(0.5 mg/g bodyweight) for 7 days. Results: glycine upregulated cardiac ejection fraction and fractional shortening to improve cardiac function, as evaluated by echocardiography. Histological and immunohistochemical analyses demonstrated that glycine could decrease inflammatory cell infiltration and alleviate collagen deposition. Western blotting revealed that nuclear factor-κB(NF-κB)-mediated inflammatory signaling was also downregulated by glycine treatment. The expression of signal transducer and activator of transcription 3(STAT3), tumor necrosis factor-α, and transforming growth factor-β(TGF-β) was decreased significantly in the glycine-treated group compared to the model group. Thus, glycine plays a protective role against myocardial ischemia and subsequent MF. Conclusion: The protective effects of glycine were achieved partly through STAT3/NF-κB/TGF-β signaling pathway.

Tryptophan(Trp)carries a unique heteroaromatic indole side chain and plays a critical role in peptide or protein modification.Herein,we have reported a metal-free photoinduced N-H alkylation strategy using N-aryl glycines for specific modification of tryptophan-containing peptides.The robustness of our approach is demonstrated by its wide substrate scope,excellent isolated yields,as well as almost unobservable side effects.Using this highly efficiently metal-free condition,alkylated Trp-containing peptides can be smoothly assembled.This study provides a reliable and practical tool for the chemo-selective modification of various tryptophan containing oligopeptides.

glycine represses endoplasmic reticulum stress-related apoptosis and improves intestinal barrier by activating mammalian target of rapamycin complex 1 signaling

Endoplasmic reticulum(ER)stress has been associated with the dysfunction of intestinal barrier in humans and animals.We have previously shown that oral administration of glycine to suckling-piglets improves ER stress-related intestinal mucosal barrier impairment and jejunal epithelial apoptosis.However,the underlying mechanism remains unknown.In this study,the protective effect and the mechanism of glycine on apoptosis and dysfunction in intestinal barrier induced by brefeldin A(BFA),an ER stress inducer,was explored in porcine intestinal epithelial cells(IPEC-1).The results showed that BFA treatment led to enhanced apoptosis and upregulation of proteins involved in ER stress signaling,including inositol-requiring enzyme 1a(IRE1a),activating transcription factor 6a(ATF6a),c-Jun N-terminal kinase(JNK),and C/EBP-homologous protein(CHOP).In addition,BFA induced a dysfunction in intestinal epithelial barrier,as evidenced by the increased paracellular permeability,decreased transepithelial electrical resistance(TEER),and reduced abundance of tight junction proteins(occludin,claudin-1,zonula occludens[ZO]-1,and ZO-2).These alterations triggered by BFA were significantly abolished by glycine treatment(Pglycine on barrier function impaired by ER stress.Importantly,we found that the regulatory effect of glycine on intestinal permeability,proteins implicated in ER stress and apoptosis,as well as the morphological alterations of the ER were reversed by rapamycin.In summary,our results indicated that glycine alleviates ER stress-induced apoptosis and intestinal barrier dysfunction in IPEC-1 cells in a mammalian target of rapamycin complex 1(mTORC1)-dependent manner.The data provides in vitro evidence and a mechanism for the protective effect of glycine against the disruption of intestinal barrier integrity induced by ER stress.

Functionalization of diatomite with glycine and amino silane for formaldehyde removal

Two amino-functionalized diatomite(DE)composites modified by 3-aminopropyltriethoxysilane(APTS)or glycine(GLY)(i.e.,APTS/DE and GLY/DE)were successfully synthesized via the wet chemical method for the time-and cost-efficient removal of indoor formaldehyde(HCHO).First,the optimal preparation conditions of the two composites were determined,and then their microstructures and morphologies were characterized and analyzed.Batch HCHO adsorption experiments with the two types of amino-modified DE composites were also conducted to compare their adsorption properties.Experimental results indicated that the pseudo-second-order kinetic and Langmuir isotherm models could well describe the adsorption process,and the maximum adsorption capacities of APTS/DE and GLY/DE prepared under optimal conditions at 20°C were 5.83 and 1.14 mg·g^(-1),respectively.The thermodynamic parameters of the composites indicated that the adsorption process was spontaneous and exothermic.The abundant amine groups grafted on the surface of DE were derived from the Schiff base reaction and were essential for the high-efficient adsorption performance toward HCHO.

glycine receptors contribute to cytoprotection of glycine in myocardial cells

Background The classic glycine receptor （GlyR） in the central nervous system is a ligand-gated membrane-spanning ion channel. Recent studies have provided evidence for the existence of GlyR in endothelial cells, renal proximal tubular cells and most leukocytes. In contrast, no evidence for GlyR in myocardial cells has been found so far. Our recent researches have showed that glycine could protect myocardial cells from the damage induced by lipopolysaccharide （LPS）. Further studies suggest that myocardial cells could contain GlyR or binding site of glycine. Methods In isolated rat heart damaged by LPS, the myocardial monophasic action potential （MAP）, the heart rate （HR） the myocardial tension and the activities of lactate dehydrogenase （LDH） from the coronary effluent were determined. The concentration of intracellular free calcium （[Ca^2＋]i） was measured in cardiomyocytes injured by LPS and by hypoxia/reoxygenation （H/R）, which excludes the possibility that reduced calcium influx because of LPS neutralized by glycine. Immunohistochemistry was used to detect the GlyR in myocardial tissue. GlyR and its subunit in the purified cultured cardiomyocytes were identified by Western blotting. Results Although significant improvement in the MAP/MAPD20, HR, and reduction in LDH release were observed in glycine ＋ LPS hearts, myocardial tension did not recover. Further studies demonstrated that glycine could prevent rat mycordial cells from LPS and hypoxia/reoxygenation injury （no endotoxin） by attenuating calcium influx. Immunohistochemistry exhibited a positive green-fluorescence signaling along the cardiac muscle fibers. Western blotting shows that the purified cultured cardiomyocytes express GlyR β subunit, but GlyR α1 subunit could not be detected. Conclusions The results suggest that glycine receptor is expressed in cardiomyocytes and participates in cytoprotection from LPS and hypoxia/reoxygenation injury. glycine could directly activate GlyR on the cardiomyocytes and prevent calcium influx into the cardiomyocyt

Aim:To determine whether glycine could downregulate interleukin 1 receptorassociated kinase-4(IRAK-4)expression to interfere with lipopolysaccharides(LPS)signal transduction and blunt transplantative liver ischemia-reperfusioninjury(I/RI).Methods:SD rats were randomly divided into two groups:donoranimals of the glycine group(n=40)were given glycine(1.5 mL;300 mmol/L,iv)1h before harvest,and the control group were treated with 1.5 mL physiologicalsaline(n=40).Orthotropic liver transplantation was then performed according tothe Kamada technique.Ten animals in each group were followed up for 7 d aftersurgery to assess survival.The remaining animals in each group were dividedinto 3 subgroups(n=10)at 1h,2h and 6 h after portal vein reperfusion.Levels ofLPS,serum aspartate aminotransferase(AST),alanine aminotransferase(ALT)and total bilirubin in portal circulation,as well as IRAK-4 and TNF-α expression,NF-κB transcriptional activity and morphological study of liver tissues wereanalyzed.Results:Reperfusion resulted in a significant elevation of LPS concen-trations in each group persisting to the end of our study.However,glycine,whichled to improved survival rate and liver function,significantly alleviated liver pa-renchyma cell damage by downregulating 1RAK-4,TNF-α expression and NF-κBtranscriptional activity compared with the control group.Conclusion:glycinecan attenuate hepatic I/RI by downregulating IRAK-4 to interfere with LPS signaltransduction.

Role of glycine on sulfuric acid-ammonia clusters formation:Transporter or participator

glycine(Gly) is ubiquitous in the atmosphere and plays a vital role in new particle formation(NPF).However,the potential mechanism of its on sulfuric acid(SA)-ammonia(A)clusters formation under various atmospheric conditions is still ambiguous.Herein,a(Gly)x·(SA)y·(A)z(z≤x+y≤3) multicomponent system was investigated by using density functional theory(DFT) combined with Atmospheric Cluster Dynamics Code(ACDC) at different temperatures and precursor concentrations.The results show that Gly,with one carboxyl(-COOH) and one amine(-NH2) group,can interact strongly with SA and A in two directions through hydrogen bonds or proton transfer.Within the relevant range of atmospheric concentrations,Gly can enhance the formation rate of SA-A-based clusters,especially at low temperature,low [SA],and median [A].The enhancement(R) of Gly on NPF can be up to 340 at T=218.15 K,[SA]=10^4,[A]=10^9,and [Gly]=107 molecules/cm^3.In addition,the main growth paths of clusters show that Gly molecules participate into cluster formation in the initial stage and eventually leave the cluster by evaporation in subsequent cluster growth at low [Gly],it acts as an important "transporter" to connect the smaller and larger cluster.With the increase of [Gly],it acts as a "participator" directly participating in NPF.

A highly regio-and enantioselective allylic C–H alkylation of 1,4-dienes with glycine Schiff bases has been established by chiral palladium-phosphoramidite catalysis. This reaction can proceed under mild conditions and tolerate a wide scope of 1,4-dienes,delivering structurally diverse chiral β-branched α-amino acid surrogates in moderate to high yields and with high levels of regio-, Z/E-, diastereo-and enantioselectivities.

glycine is a well-documented cytoprotective agent.However,whether it has a protective effect against myocar-dial ischemia-reperfusion injury in vivo is still unknown.By using an open-chest anesthetized rat model,we found that glycine reduced the infarct size by 21% in ischemia-reperfusion injury rats compared with that in the vehicle-treated MI/R rats.The left ventricular ejection fraction and fractional shortening were increased by 19.11% and 30.98%,respectively,in glycine-treated rats.The plasma creatine kinase levels in ischemia-reperfusion injury rats decreased following glycine treatment.Importantly,administration of glycine significantly inhibited apoptosis in post-ischemia-reperfusion myocardium,which was accompanied by suppression of phosphorylated p38 mitogen-activated protein kinase and c-Jun NH2-terminal kinase,as well as the Fas ligand.These results suggest that gly-cine attenuates myocardial ischemia-reperfusion injury in vivo by inhibiting cardiomyocytes apoptosis.

Branch number(BN)is an important agronomic attribute related to the plant architecture,adaptability,and yield of soybean.To date,few studies ofBNhave been conducted to elucidate its genetic background.We aimed to localize genetic factors affecting BN using segregating populations derived fromthe high-branching cultivar‘Kennong24’(KN24)and the low-branching cultivar‘Kenfeng19’(KF19).Composite interval mapping analysis detected a QTL(qBN-1)on chromosome 6 between the SSR markers BARCSOYSSR_06_0993 and BARCSOYSSR_06_1070 using an F2 population.To fine-map qBN-1,a RIL population was developed and genotyped with 14 SSRmarkers located in the QTL region.qBN-1 was localized to a 115.67-kb interval flanked by markers BARCSOYSSR_06_1048 and BARCSOYSSR_06_1053.The QTL was further confirmed using backcross populations of size 1305(BC2F2 with KN24 as a recurrent parent)and 1712(BC3F2 with KF19 as a recurrent parent).The fine-mapping region of qBN-1 contained only two candidate genes,Glyma.06G208800 and Glyma.06G208900,whose expression patterns were investigated by qRT-PCR.Compared to Glyma.06G208800 gene expression,Glyma.06G208900 showed the highest expression of the two genes and showed a significant difference in expression between high-and low-branching genotypes in either axillary meristem or shoot apical meristem,and showed opposite expression patterns in the two tissues at V4 and R1 stages.These results identify Glyma.06G208900 as a novel candidate gene controlling BN.Taken together,the results of this study provide a foundation for cloning and functional analysis of the qBN-1 gene and for the improvement of BN bymarker-assisted selection in soybean breeding.
