Microsatellite loci distributing on genome randomly act as effective genetic markers. To date, about 200 microsatellite loci were found in cervids b y transferring microsatellite PCR primers derived in bovine, ovine to cervids, a s well as a few loci derived directly from deer microsatellite library. These lo ci have been used in parentage determination, genetic diversity and population s tructure, population introgression, as genetic marker gestation length and winte ring survival et al. However, microsatellite loci presently found are untouchabl e to the demand of application. Future work should include: 1) isolating a large number of cervine microsatellite loci, 2) constructing genetic and physical map s of microsatellite loci. So that microsatelites have a strong base for advanced applications in deer.

Twenty-two nitrate nonutilizing （nit） mutants were recovered from five wild-type isolates of Fusarium graminearum and fifty nit mutants were recovered from three JS399-19-resistant mutants of F. graminearum cultured on MMC medium. Some biological properties were compared between nit mutants and their parental isolates. The results showed that there were no significant differences in growth rate, cultural characters or pathogenicity between JS399-19-resistant nit mutants and their parental isolates. But the conidial production and the sexual reproduction ability changed to some extent. There was no cross resistance toward chlorate and JS399-19 in F. graminearum and the resistance could be stable through 20-time subcultures. Therefore, the nit could be used as a genetic marker for studying the genetics of JS399-19 resistance in E graminearum, which was used to study JS399-19 resistance transferability in hyphal fusion. Resistance in JS399-19 could not be transferred by hyphal fusion or could be transferred with low chance between two compatible isolates, which would delay the development of JS399-19 resistance in the field.

The genetic structure of three stocks (K, H and S) of Penaeus chinensis was detected by RAPD method which useed 41 random oligonucleotide primers with 10 bases. Stock K was from the spawning stock of the western coastal waters of the Korea Peninsula, H was from the Huanghai Sea-Bohai Sea stock and S was from the health cultured stock in Jimo, Shandong Province, China. Three genetic markers, F-02(540), OPN-13(800) and OPN-20(2200), which could differentiate the three different stocks, have been reported. The basic data for marker assisted selection (MAS) on genetic breeding of Penaeus chinensis are provided.

Gene Transfer into Hematopoietic Cells of Mouse and Its in vivo Expression after Transplantation

We have shown previously that high-efficient gene transfer can be attained in primary hematopoietic cells using liposome-mediated gene transfer strategy. In order to examine the stability of gene expression mediated by this gene transduction protocol, we observed the expression of marker gene in vivo by using bone marrow transplantation (BMT) to engraft lethally irradiated mouse with the genetically modified hematopoietic cells. The results showed that the mouse transplanted with appropriated number of transduced cells remained alive andhealthy. The PCR analysis and G418 selection of the spleen colonies and bone marrow cells isolated from lethally irradiated animals 15 days and 30 days after injection of genetically modified bone marrow cells showed that the progeny cells of the transduced hematopoietic stem cells still contained and expressed the transduced genes, suggesting that the hematopoietic system is at least partially re-constructed by the stem cells with marker gene and that the stable expression of foreign genes in vivo can be attained by using this easy and harmless transduction protocol. These findings provide experimental basis for clinician to further investigate the biology of marrow reconstruction and the mechanism of leukemia relapse after BMT.

Frequency and Distribution of Microsatellites in the Genome of Filamentous Fungus,Neurospora crassa

A total of 38.0 Mb of publicly available DNA sequence in Neurospora crassa was researched for mono- to hexanucleotide simple sequence repeats (SSR or microsatellite) to determine the type, size and frequency. A total of 14 788 SSRs were observed in the whole genomic DNA sequence, about one every 2.57 kb, with the criteria of SSR length >15 bp and 80% matches. The most abundant microsatellite was trinucleotide repeat, the number was 4 729, followed by hexanucleotide and mononucleotide repeats, the numbers were 2 940 and 2 489 respectively, and the least abundance was dinucleotide repeat, only 691 were found. Among the 10 082 ORFs, 4 094 SSRs were harbored in 2 373 ORF (no intron) of the organism. One thousand and fifty six ORFs harbored only one SSR. Similar with other organisms, tri- and hexanucleotide repeats were predominant in ORFs, 54.1 and 48.8% of tri- and hexanucleotide repeats were distributed in ORF region. The density of these two motifs was overpresented in coding regions, because ORF region and coding region constitutes only 46 and 38.3% of genomic sequence, respectively. Upstream and downstream 300 bp of regulatory regions were high density regions of SSRs, particularly density of pentanucleotide SSR in upstream region was as high as five times of average density in genomic DNA, density of di- and tetranucleotide SSR was also more than two times of average density. The density of penta-, tetra-, di- and mononucleotide SSRs was relatively higher than average density. There were 47 SSRs in mitochondria 64 840 bp DNA sequence, their distribution is similar with genomic DNA sequence. These results suggested that SSRs were clustered in regulatory regions of genomic DNA.

Alternaria tenuissima is a fungus widely present in the environment and could cause diseases in plants and humans. In this study, through a yeast genetic approach, cDNA sequences were isolated and characterized for the AtHIS3 and AtLEU2 genes. AtHIS3 cDNA encodes a protein of 238 amino acids, while AtLEU2 cDNA encodes a protein of 363 amino acids. Based on the phylogenetic analysis of amino acid sequences of AtHis3p and AtLeu2p, A. tenuissima is closely related to the plant pathogenic fungus Phaeosphaeria nodorum. This study provides two genetic markers for studies of functions of genes regulating development, morphology, and virulence of A. tenuissima.

Changes in Tree Frog (<i>Hyla savignyi</i>) Coloration in Unstable Habitats at the Southern Border of Its Distribution

This study examined the relationship between tree frog (Hyla savignyi) coloring and its different seasonal habitats at the southern border of its distribution. The results show that tree frog color is affected by the dominant colors in its habitat, which vary seasonally, especially between winter and summer. Tree frog colors were various shades of green, white, brown, and black. No genetic marker was found to characterize the color. The ability of a small frog to infer its own time with the help of color changes occurring in the habitat on the southern border of its distribution, which are relatively broad, gives this species an advantage.

Foxtail millet （Setaria italica L. Beauv.）, as a significant food and fodder cereal crop, was widely cultivated in the Yellow River Valley and is still a kind of farming tradition of millet in Northern China. With the development of agriculture and the improvement of people＇s living standards, foxtail millet＇s planting area has be- come less and less in arid and semi-arid region now. However, because of its small diploid genome （1C genome size=420 Mb） and self-pollination, foxtail millet is very suitable for whole genome sequence and is used as an experimental model plant for C4 photosynthesis and biofuel research. In 2012, the completed genome sequence of foxtail millet had been successfully produced by the Beijing Genomics Institute （BGI） and US Department of Energy Joint Genomic Institute （JGI）, respec- tively. It will be used as an experimental crop to explore many aspects of plant ar- chitecture, physiology and biochemistry, comparative and functional genomics studies in the bioenergy grasses. To systematically understand the recent research progress in foxtail millet; we summarize the following aspects in this study： germplasm, tradi- tional breeding, physiology and biochemistry, molecular marker, construction of genet- ic linkage map, gene localization, genome sequencing and comparative genome. This may be a door to open for the further development of foxtail millet in the future.
